De hier beschreven methoden bieden een gegeneraliseerde workflow om vectorcompetentieanalyses uit te voeren. Als algemeen kader worden veel van deze methodologieën in de literatuur bewaard. Er is echter aanzienlijke ruimte voor wijzigingen (besproken in Azar en Weaver1). Van virussen (bijv. virale afstamming, opslag van challenge-virus, virale passagegeschiedenis), entomologie (bijv. laboratoriumkolonisatie van muggenpopulaties, aangeboren immuniteit, het muggenmicrobioom / viroom) en experimente...

Vectorcompetentie wordt gedefinieerd als het vermogen op het niveau van soort, populatie en zelfs een individu van een bepaalde geleedpotige zoals een mug, teek of flebotomine zandvlieg, om een agens biologisch te verwerven en over te dragen met replicatie of ontwikkeling in de geleedpotige1,2. Met betrekking tot muggen en door geleedpotigen overgedragen virussen (d.w.z. arbovirussen), wordt het middel door een vrouwelijke mug uit een viremische gastheer geïmpregneerd. Na inname moet het virus productief een van een kleine populatie midgut-epitheelcellen infecteren3, waarbij verschillende fysiologische obstakels worden overwonnen, zoals proteolytische afbraak door spijsverteringsenzymen, de aanwezigheid van de microbiota (midgut-infectiebarrière of MIB) en de uitgescheiden peritrofische matrix. Infectie van het midgut-epitheel moet worden gevolgd door replicatie van het virus en uiteindelijk ontsnapping uit het midgut in de open bloedsomloop van de mug, of hemolymfe, wat het begin vertegenwoordigt van een gedissemineerde infectie die de midgut-ontsnappingsbarrière (MEB) overwint. Op dit punt kan het virus infecties van secundaire weefsels (bijv. Zenuwen, spieren en vetlichamen) vaststellen en zich blijven vermenigvuldigen, hoewel een dergelijke secundaire replicatie mogelijk niet strikt noodzakelijk is voor het virus om de acinaire cellen van de speekselklieren te infecteren (het overwinnen van de speekselklierinfectiebarrière). Uitgaan van de speekselklier acinar cellen in hun apicale holtes en vervolgens beweging in het speekselkanaal maakt inenting van het virus in volgende gastheren bij bijten mogelijk en voltooit de transmissiecyclus1,2,4,5,6,7.
Gezien dit goed gekarakteriseerde en over het algemeen geconserveerde mechanisme van verspreiding binnen een muggenvector, zijn laboratoriumvectorcompetentiebeoordelingen vaak methodologisch vergelijkbaar, hoewel er verschillen in protocollen bestaan1,2. Over het algemeen worden muggen na orale blootstelling aan het virus ontleed, zodat individuele weefsels zoals het midgut, benen, eierstokken of speekselklieren kunnen worden getest op virale infectie, gedissemineerde infectie, gedissemineerde infectie / potentiële transovariale overdracht en gedissemineerde infectie / potentiële overdrachtscompetentie, respectievelijk8. De loutere aanwezigheid van een virus in de speekselklieren is echter geen definitief bewijs van overdrachtsvermogen, gezien het bewijs van een speekselklier ontsnapping / uitgang barrière (SGEB) in sommige vector / virus combinaties1,2,4,5,7,9. De standaardmethode om overdrachtscompetentie aan te tonen blijft muggenoverdracht op een vatbaar dier10,11,12. Aangezien dit voor veel arbovirussen echter het gebruik van immuungecompromitteerde muizenmodellen13,14,15,16vereist, is deze methode vaak onbetaalbaar. Een veelgebruikt alternatief is het verzamelen van het muggenspeeksel, dat kan worden geanalyseerd door reverse transcription-polymerase chain reaction (RT-PCR) of een infectieuze test om de aanwezigheid van respectievelijk het virale genoom of infectieuze deeltjes aan te tonen. Het is vermeldenswaard dat dergelijke in vitro speekselverzamelingsmethoden12 kunnen overschatten of17 de hoeveelheid virus die tijdens in vivo voeding wordt afgezet, kunnen onderschatten, wat aangeeft dat dergelijke gegevens met de nodige voorzichtigheid moeten worden geïnterpreteerd. Niettemin is de in vitro-methode zeer waardevol wanneer geanalyseerd vanuit het perspectief van de loutere aanwezigheid van virus in het speeksel, wat wijst op transmissiepotentieel.
Er bestaan twee belangrijke benaderingen voor het bepalen van de rol van muggenvectoren bij uitbraken van arbovirale ziekten. De eerste methode omvat veldbewaking, waarbij muggen worden verzameld in het kader van actieve overdracht18,19,20,21,22,23,24. Aangezien de infectiepercentages echter doorgaans vrij laag zijn (bijv. de geschatte 0,061% infectiegraad van muggen in gebieden met actieve Zika-virus (ZIKV) circulatie in de Verenigde Staten21), kan de beschuldiging van potentiële vectorsoorten sterk worden beïnvloed door vangmethode25,26 en willekeurige kans (bijv. Bemonstering van één geïnfecteerd individu op 1.600 niet-geïnfecteerden)21 . Hiermee rekening houdend, kan een bepaald onderzoek mogelijk niet voldoende muggen verwerven in zowel ruwe aantallen als soortendiversiteit om muggen die betrokken zijn bij overdracht nauwkeurig te bemonsteren. Vectorcompetentieanalyses worden daarentegen uitgevoerd in een laboratoriumomgeving, waardoor strikte controle van parameters zoals orale dosis mogelijk is. Hoewel ze niet volledig in staat zijn om de ware complexiteit van muggeninfectie en overdrachtscapaciteit in een veldomgeving weer te geven, blijven deze laboratoriumbeoordelingen krachtige hulpmiddelen op het gebied van arbovirologie.
Op basis van verschillende vectorcompetentieanalyses met ZIKV in verschillende muggensoorten, populaties en methoden27,28,29,30,31,32,evenals een recent overzicht van vectorcompetentiebeoordelingen1,beschrijven we hier verschillende protocollen die verband houden met een typische vectorcompetentieworkflow. In deze experimenten werden drie Ae. aegypti-populaties uit Amerika (de stad Salvador, Brazilië; de Dominicaanse Republiek; en de lagere Rio Grande-vallei, TX, VS) blootgesteld aan een enkele stam van ZIKV (Mex 1-7, GenBank Accession: KX247632.1) bij 4, 5 of 6 log10 focusvormende eenheden (FFU) / ml-doses door middel van kunstmatige bloedmeel. Vervolgens werden ze geanalyseerd op bewijs van infectie, gedissemineerde infectie en transmissiecompetentie na verschillende tijden van extrinsieke incubatie (2, 4, 7, 10 en 14 dagen) door middel van dissectie en een op celcultuur gebaseerde infectieuze test. Hoewel de huidige workflow/protocollen zijn geoptimaliseerd voor ZIKV, zijn veel elementen direct vertaalbaar naar andere door muggen overgedragen arbovirussen in geleedpotige insluiting en bioveiligheidsniveaus 2 en 3 (ACL/BSL2 of ACL/BSL3).
