ほとんどの真核生物では、スピンドル微小管は、セントロメアと呼ばれる各染色体の単一領域に付着する必要があります。キネトコレは、セントロメアに位置するタンパク質の複合体です。セントロメアとキネトコレタンパク質の動きのタイミングとキネトコレスとセントロメレスの構造を研究することは、染色体不安定性(CIN)および癌進行を理解するために重要です。重要な問題は、染色体の位置と中心の機能(すなわち、セントロメアの同一性)がどのように決定され、それらが正確な染色体分離にどのように関与しているかである。ほとんどの種では、CENP-Aと呼ばれる特定のヒストン様タンパク質を含む特殊なヌクレオソームの存在がセントロメア同一性を定義する。そこで、CENP-Aが中心同一性の非DNA指標(エピジェネティックマーク)であることを提案する。CENP-Aがヒトのセントロメア同一性をどのように定義するかを解明することが重要です。
ホリデイ接合認識タンパク質(HJURP)は、CENP-A特異的シャペロンであり、CENTP-Aを中心核組中11、2、32,3に沈着させる。我々は、リジン124(K124)およびセントロメレローカリゼーション4のCENP-A ubiquityationに、CUL4A-RBX1-COPS8 E3リガーゼが必要であることを以前に報告している。また、我々の結果は、新たに合成されたCENP-Aのセントロメア募集には、既存のユビキタスCENP-A5が必要であることを示した。したがって、CENP-Aのユビキタス化は細胞分裂間の二量体化によって継承されることを示唆するモデルが提供された。
