Le spectre RMN 1H (Figure 2) confirme la synthèse réussie du copolymère CT-PEG et du copolymère PEI-g-PEG. Étant le principal problème de santé publique dans le monde, le cancer est largement caractérisé comme ayant un faible taux de guérison, un taux de récidive élevé et un taux de mortalité élevé1,2. Les méthodes anticancéreuse conventionnelles actuelles comprennent la chirurgie, la chimiothérapie et la radiothérapie3,parmi lesquelles la chimiothérapie est le traitement principal pour les patients atteints de cancer dans la clinique4. Les médicaments anticancéreux utilisés cliniquement comprennent principalement le paclitaxel (PTX)5 et la doxorubicine (DOX)6,7. DOX, un médicament antinéoplasique, a été largement appliqué en chimiothérapie clinique, en raison des avantages de la cytotoxicité du cancer et de l’inhibition de la prolifération des cellules cancéreuses8,9. Cependant, DOX provoque une cardiotoxicité10,11,et la courte demi-vie de DOX restreint son application dans la clinique12. Par conséquent, des porteurs de médicaments dégradables sont nécessaires pour charger dox et libérer subequently d’une manière contrôlée à une zone ciblée.
Les nanoparticules ont été largement utilisées dans les systèmes d’administration de médicaments ciblés et présentent plusieurs avantages dans le traitement du cancer (c.-à-d. rapport surface/volume important, petite taille, capacité d’encapsuler divers médicaments et chimie de surface accordable, etc.) 13,14,15. En particulier, les nanoparticules d’or (AuNPs) ont été largement utilisées dans des applications biologiques et biomédicales, telles que le traitement photothermique du cancer16,17. Les propriétés uniques des AuNPs, telles que la synthèse facile et la fonctionnalisation générale de surface, ont d’excellentes perspectives dans le domaine clinique du traitement du cancer18. En outre, les AuNPs ont été utilisés pour identifier les stratégies d’administration de médicaments, diagnostiquer les tumeurs et surmonter la résistance dans de nombreuses études19,20.
Néanmoins, les AuNPs doivent être davantage adaptés pour surmonter la résistance aux médicaments via une libération locale élevée aux lésions tumorales par perméation et rétention améliorées (EPR), telles que les propriétés de ciblage et d’accessibilité. Les polymères fonctionnalisés AuNPs ont présenté des avantages uniques, tels que l’amélioration de la solubilité dans l’eau des médicaments anticancéreux hydrophobes et le temps de circulation prolongé21,22. Divers polymères biocompatibles ont été utilisés pour les revêtements AuNP, tels que le polyéthylène glycol (PEG), la polyéthylèneimine (PEI), l’acide hyaluronique, l’héparine et la gomme xanthane. Ensuite, la stabilité, ainsi que la charge utile, des AuNPs est améliorée bien23. Plus précisément, PEI est un polymère hautement ramifié qui est composé de nombreuses unités répétitrices d’amines primaires, secondaires et tertiaires24. L’Île-du-Prince-Édouard a une excellente solubilité, une faible viscosité et un haut degré de fonctionnalité, ce qui convient au revêtement sur les AuNPs.
D’autre part, les médicaments anticancéreux doivent être livrés directement aux cellules cancéreuses avec une efficacité de chargement améliorée et une toxicité plus faible pour le traitement des tumeurs métastatiques primaires et avancées25. Les ligands ciblés ont un grand potentiel pour les systèmes d’administration ciblés de médicaments anticancéreux26. Sa sélectivité pour la liaison à la molécule cible confère une spécificité de ciblage des médicaments anticancéreux et augmente l’enrichissement des médicaments dans les tissusmalades 27. Plus de ligands comprennent des anticorps, des polypeptides et de petites molécules. Par rapport à d’autres ligands, les aptamères d’acides nucléiques peuvent être synthétisés in vitro et sont faciles à modifier. AS1411 est un oligonucléotide phosphodiétre de 26 pb non modifié qui forme une structure G-tétramère dimérique stable pour se lier spécifiquement à un récepteur protéique nucléaire cible surexprimé sur les cellules cancéreuses28,29,30. L’AS1411 inhibe la prolifération de nombreuses cellules cancéreuses mais n’affecte pas la croissance des cellules saines31,32. En conséquence, AS1411 a été utilisé pour fabriquer un système d’administration de médicaments ciblé idéal.
Dans cette étude, un copolymère PEI-g-PEG est synthétisé via une réaction d’amide, puis des nanoparticules d’or recouvertes de copolymere PEI-g-PEG (PEI-g-PEG@AuNPs) sont fabriquées. En outre, DOX et AS1411 sont liés séquentiellement à la pei-g-PEG@AuNPs préparée, comme indiqué dans Figure 1. Ce protocole détaillé vise à aider les chercheurs à éviter bon nombre des pièges courants associés à la fabrication de nouveaux pei-g-PEG@AuNPs chargés de DOX et d’AS1411.

synthesis of aptamer-pei-g-peg modified gold nanoparticles loaded with doxorubicin for targeted drug delivery

En raison de la résistance aux médicaments et de la toxicité dans les cellules saines, l’utilisation de la doxorubicine (DOX) a été limitée dans le traitement clinique du cancer. Ce protocole décrit la conception de poly(éthylénimine) greffée avec des nanoparticules d’or fonctionnalisées de polyéthylène glycol (PEI-g-PEG) avec des aptamères chargés (AS1411) et du DOX par des réactions d’amide. AS1411 est spécifiquement lié avec des récepteurs nucléoline ciblés sur les cellules cancéreuses afin que DOX cible les cellules cancéreuses au lieu des cellules saines. Tout d’abord, le PEG est carboxylé, puis greffé à l’Î.-P.-É. ramifié pour obtenir un copolymère PEI-g-PEG, ce qui est confirmé par une analyse RMN de 1H. Ensuite, les nanoparticules d’or revêtues de copolymère PEI-g-PEG (PEI-g-PEG@AuNPs) sont synthétisées, et DOX et AS1411 sont liés de manière covalente aux AuNPs progressivement via des réactions d’amide. Le diamètre de l’AS1411-g-DOX-g-PEI-g-PEG@AuNPs préparé est d’environ 39,9 nm, avec un potentiel zêta de -29,3 mV, ce qui indique que les nanoparticules sont stables dans l’eau et le milieu cellulaire. Les tests de cytotoxicité cellulaire montrent que les AuNPs chargés de DOX nouvellement conçus sont capables de tuer les cellules cancéreuses (A549). Cette synthèse démontre la disposition délicate des copolymères PEI-g-PEG, des aptamères et des DOX sur les AuNPs qui sont obtenus par des réactions séquentielles d’amide. Ces Aptamères-PEI-g-PEG fonctionnalisés AuNPs fournissent une plate-forme prometteuse pour l’administration ciblée de médicaments dans le traitement du cancer.
