Als das größte Problem der öffentlichen Gesundheit weltweit, Krebs wird weithin als mit einer niedrigen Heilungsrate, hohe Rezidivrate und hohe Sterblichkeitsrate1,2gekennzeichnet. Aktuelle konventionelle Anti-Krebs-Methoden umfassen Chirurgie, Chemotherapie und Strahlentherapie3, unter denen Chemotherapie ist die primäre Behandlung für Krebspatienten in der Klinik4. Klinisch verwendete Anti-Krebs-Medikamente gehören hauptsächlich Paclitaxel (PTX)5 und Doxorubicin (DOX)6,7. DOX, ein antineoplastisches Medikament, wurde in der klinischen Chemotherapie allgemein angewendet, aufgrund der Vorteile der Krebszytotoxizität und Hemmung der Proliferation von Krebszellen8,9. Jedoch, DOX verursacht Kardiotoxizität10,11, und die kurze Halbwertszeit von DOX beschränkt seine Anwendung in der Klinik12. Nanopartikel wurden häufig in gezielten Medikamentenabgabesystemen eingesetzt und haben mehrere Vorteile in der Krebsbehandlung (d. h. großes Oberflächen-Volumen-Verhältnis, geringe Größe, Fähigkeit, verschiedene Medikamente zu kapseln, und abstimmbare Oberflächenchemie usw.) 13,14,15. Insbesondere Gold-Nanopartikel (AuNPs) sind in biologischen und biomedizinischen Anwendungen wie der photothermischen Krebstherapie16,17weit verbreitet. Die einzigartigen Eigenschaften von AuNPs, wie einfache Synthese und allgemeine Oberflächenfunktionalisierung, haben hervorragende Perspektiven im klinischen Bereich der Krebstherapie18. Auch, AuNPs wurden verwendet, um Medikamentenabgabestrategien zu identifizieren, Tumoren zu diagnostizieren, und Resistenzen in vielen Studien zu überwinden19,20.
Ungeachtet dessen müssen AuNPs weiter darauf zugeschnitten werden, die Arzneimittelresistenz durch hohe lokale Freisetzung bei Tumorläsionen durch verbesserte Permeation und Retention (EPR) zu überwinden, wie z. B. die Targeting- und Barrierefreiheitseigenschaften. Polymer funktionalisierte AuNPs haben einzigartige Vorteile gezeigt, wie verbesserte Wasserlöslichkeit von hydrophoben Anti-Krebs-Medikamente und längere Durchblutungszeit21,22. Für AuNP-Beschichtungen wurden verschiedene biokompatible Polymere verwendet, wie Polyethylenglykol (PEG), Polyethylenimin (PEI), Hyaluronsäure, Heparin und Xanthan. Insbesondere ist PEI ein hoch verzweigtes Polymer, das aus vielen sich wiederholenden Einheiten von primären, sekundären und tertiären Aminen24besteht. PEI verfügt über eine ausgezeichnete Löslichkeit, geringe Viskosität und ein hohes Maß an Funktionalität, das für die Beschichtung auf AuNPs geeignet ist.
Auf der anderen Seite müssen Krebsmedikamente direkt mit verbesserter Belastbarkeit und geringerer Toxizität für die Behandlung von primären und fortgeschrittenen metastasierenden Tumoren an Krebszellen abgegeben werden25. Gezielte Liganden haben ein großes Potenzial für Anti-Krebs-Medikamente gezielte Abgabesysteme26. Seine Selektivität für die Bindung von Zielmolekülen verleiht Anti-Krebs-Medikament auf Spezifität und erhöht die anreicherung von Medikamenten in erkrankten Geweben27. Mehr Liganden enthalten Antikörper, Polypeptide und kleine Moleküle. Im Vergleich zu anderen Liganden können Nukleinsäureaptamere in vitro synthetisiert werden und sind leicht zu modifizieren. AS1411 ist ein unverändertes 26 bp Phosphodiester-Oligonukleotid, das eine stabile dimerische G-Tetramer-Struktur bildet, um speziell an einen überexprimierten nuklearen Zielproteinrezeptor an Krebszellen28,29,30zu binden. AS1411 hemmt die Proliferation vieler Krebszellen, hat aber keinen Einfluss auf das Wachstum gesunder Zellen31,32. Infolgedessen wurde AS1411 verwendet, um ein ideales gezieltes Arzneimittelabgabesystem herzustellen.
In dieser Studie wird ein PEI-g-PEG-Copolymer über eine Amidreaktion synthetisiert, dann werden PEI-g-PEG Copolymer beschichtete Gold-Nanopartikel (PEI-g-PEG@AuNPs) hergestellt. Darüber hinaus sind DOX und AS1411 sequenziell mit dem vorbereiteten PEI-g-PEG@AuNPs verknüpft, wie in Abbildung 1dargestellt. Dieses detaillierte Protokoll soll Forschern helfen, viele der häufigen Fallstricke zu vermeiden, die mit der Herstellung neuer PEI-g-PEG@AuNPs verbunden sind, die mit DOX und AS1411 beladen sind.

synthesis of aptamer-pei-g-peg modified gold nanoparticles loaded with doxorubicin for targeted drug delivery

Aufgrund der Arzneimittelresistenz und Toxizität in gesunden Zellen ist die Anwendung von Doxorubicin (DOX) in der klinischen Krebstherapie begrenzt. Dieses Protokoll beschreibt die Konstruktion von Poly(Ethylenimin) mit Polyethylenglykol (PEI-g-PEG) copolymer funktionalisierten Gold-Nanopartikeln (AuNPs) mit belastetem Aptamer (AS1411) und DOX durch Amidreaktionen. AS1411 ist speziell mit gezielten Nucleolin-Rezeptoren auf Krebszellen verbunden, so dass DOX Krebszellen anstelle von gesunden Zellen anspricht. Zuerst wird PEG carboxyliert und dann an verzweigte PEI transplantiert, um ein PEI-g-PEG-Copolymer zu erhalten, das durch 1H NMR-Analyse bestätigt wird. Als nächstes werden PEI-g-PEG Copolymer-beschichtete Gold-Nanopartikel (PEI-g-PEG@AuNPs) synthetisiert, und DOX und AS1411 werden nach und nach über Amidreaktionen kovalent mit AuNPs verbunden. Der Durchmesser des präparierten AS1411-g-DOX-g-PEI-g-PEG@AuNPs beträgt 39,9 nm, mit einem Zetapotential von -29,3 mV, was darauf hinweist, dass die Nanopartikel in Wasser und Zellmedium stabil sind. Zellzytotoxizitätstests zeigen, dass die neu entwickelten DOX-geladenen AuNPs in der Lage sind, Krebszellen abzutöten (A549). Diese Synthese demonstriert die empfindliche Anordnung von PEI-g-PEG-Copolymeren, Aptamern und DOX auf AuNPs, die durch sequenzielle Amidreaktionen erreicht werden. Solche aptamer-PEI-g-PEG funktionalisierten AuNPs bieten eine vielversprechende Plattform für die gezielte Medikamentenabgabe in der Krebstherapie.
