Bacteriële invasie in longepelleiële cellen is een cruciale stap in de pathogenese van bacteriële infecties. Het proces van bacteriële invasie in de cellen kan worden opgesplitst in de volgende drie stappen: Ten eerste, de bacteriën contact en hechten zich aan het oppervlak van de epitheelcel met behulp van hun flagella. Ten tweede ondergaan de bacteriën ofwel internalisatie of dringen het cellulaire membraan binnen. Ten slotte repliceren en koloniseren de bacteriën de cellen als ze met succes ontsnappen aan cellu...

Het roken van sigaretten beïnvloedt de volksgezondheid van miljoenen mensen wereldwijd. Veel schadelijke ziekten, waaronder longkanker en chronische obstructieve longziekte (COPD), worden gemeld te worden gerelateerd aan het roken van sigaretten1,2. Het roken van sigaretten verhoogt de gevoeligheid voor acute microbiële infecties in het ademhalingssysteem3,4,5. Bovendien blijkt uit toenemend bewijs dat het roken van sigaretten de pathogenese van veel chronische aandoeningen6,7,8versterkt . Bijvoorbeeld, het roken van sigaretten kan verhogen virale of bacteriële infecties die COPD verergering veroorzaken9. Onder de bacteriële pathogenen die etiologisch bijdragen aan acute verergering van COPD, een opportunistische gram-negatieve bacillus pathogene, Pseudomonas aeruginosa, veroorzaakt infecties die correleren met slechte prognoses en hogere sterftecijfers10,11. COPD-exacerbatie verergert de ziekte door de pathologische progressie te versnellen. Er zijn geen effectieve therapieën tegen COPD-exacerbatie, behalve voor het antisymptomatische beheer12. COPD-verergering bevordert de sterfte van patiënten, vermindert de kwaliteit van leven en verhoogt de economische lasten voor de samenleving13.
De luchtwegen luchtweg is een open systeem, voortdurend onderworpen aan verschillende microbiële pathogenen die extern aanwezig zijn. Opportunistische bacteriële pathogenen worden meestal gedetecteerd in de bovenste luchtwegen, maar soms worden waargenomen in de onderste luchtwegen14,15. In diermodellen kan P. aeruginosa worden gedetecteerd in alveololatorzakjes zodra 1 uur na infectie16. Als een belangrijk afweermechanisme, immuuncellen zoals macrofagen of neutrofielen elimineren de bacteriën in de luchtwegen. Longepeppitheelcellen, als de eerste fysiologische barrière, vervullen een unieke rol in de gastheerverdediging tegen microbiële infecties. Longepeppitheelcellen kunnen microbiële invasie, kolonisatie of replicatie reguleren onafhankelijk van immuuncellen17. Sommige moleculen gevonden in epitheliale cellen, waaronder PPARg, oefenen antibacteriële functies uit, waardoor bacteriële kolonisatie en replicatie in longepitheelcellen18reguleren. Het roken van sigaretten kan de moleculen veranderen en de normale afweerfunctie in longepitheelcellen19,20aantasten. Recente studies rapporteerden directe blootstelling van sigarettenrook aan longepepelleiële cellen met behulp van robotrookapparatuur21,22. Blootstelling aan rook kan echter op andere manieren worden uitgevoerd, waaronder de toepassing van CSE. Bereiding van CSE is een reproduceerbare benadering met mogelijke toepassingen in andere celtypen, waaronder vasculaire endotheelcellen die indirect worden blootgesteld aan sigarettenrook.
Dit rapport beschrijft een protocol om sigarettenrook extract te genereren om bacteriële belasting in longepelliële cellen te veranderen. CSE verhoogt de bacteriële belasting van P. aeruginosa, en het kan bijdragen aan de herhaling van bacteriële infecties meestal gezien in COPD verergering. Voor de bereiding van CSE wordt een conventionele methode gebruikt. Longepeppitheelcellen, in hun exponentiële groeistadium, worden behandeld met 4% CSE voor 3 uur. Als alternatief kunnen monolaag-gekweekte longepelleliale cellen direct worden blootgesteld aan sigarettenrook in een lucht-vloeibare interface. CSE-behandelde cellen worden vervolgens uitgedaagd met Pseudomonas bij een veelheid van infectie (MOI) van 10. De bacteriën worden gepropageerd met een bepaalde schudsnelheid om ervoor te zorgen dat de morfologie van hun flagella intact blijft om hun volledige invasieve capaciteit te behouden. Gentamycine wordt gebruikt om de bacteriën te doden die in het kweekmedium achterblijven, waardoor de potentiële besmetting tijdens de daaropvolgende bepaling van de bacteriële belasting wordt verminderd. Het protocol maakt ook gebruik van GFP-gelabelde Pseudomonas, die is gebruikt als een krachtig instrument bij het bestuderen van Pseudomonas infectie in verschillende modellen. Het roken van sigaretten is de belangrijkste etiologische oorzaak voor longemfyseem en chronische obstructieve longziekte (COPD). Het roken van sigaretten bevordert ook de gevoeligheid voor bacteriële infecties in de luchtwegen. Echter, de effecten van het roken van sigaretten op bacteriële infecties in menselijke long epitheelcellen moeten nog grondig worden bestudeerd. Hier beschreven is een gedetailleerd protocol voor de voorbereiding van sigaretten roken extracten (CSE), behandeling van menselijke long epitheelcellen met CSE, en bacteriële infectie en infectie bepaling. CSE werd bereid met een conventionele methode. Longepeppitheelcellen werden behandeld met 4% CSE voor 3 h. CSE-behandelde cellen werden toen besmet met Pseudomonas bij een veelheid van infectie (MOI) van 10. Bacteriële belastingen van de cellen werden bepaald door drie verschillende methoden. De resultaten toonden aan dat CSE de pseudomonasbelasting in longepelleelcellen verhoogde. Dit protocol biedt daarom een eenvoudige en reproduceerbare aanpak om het effect van sigarettenrook op bacteriële infecties in longepeleliële cellen te bestuderen.

Een diagram wordt gebruikt om het protocol in figuur 1te illustreren. Longepitheel BEAS-2B cellen werden behandeld met CSE en uitgedaagd met Pseudomonas. Dit protocol maakt gebruik van drie benaderingen om te bepalen of sigarettenrook pseudomonas bacteriële belasting in longepelleëlcellen beïnvloedt. Deze methode kan worden uitgebreid naar endotheelcellen of andere celkranen. Voorbereiding van de sigaret rook extractie, bacteriële infectie en de bepaling met een bacteriële belasting zijn goed beschreven in detail in deze video.Deze procedure is zeer gemakkelijk te volgen voor de nieuwe gebruikers. Teken 10 milliliter serumvrije celkweekmedium in een spuit van 60 milliliter. Bevestig het smalle uiteinde van een trim aan een milliliter pipetpunt aan het mondstuk van de spuit, als een adapter om de sigaret vast te houden.Haal het filter uit de sigaret en bevestig de sigaret aan de adapter. Niet meer dan 30 minuten voor het uitvoeren van de test brand de sigaret en trek 40 milliliter rook met lucht in de spuit. Meng de rook met het medium door de spuit krachtig te schudden.Herhaal het tekenproces totdat de sigaret volledig is uitgebrand, wat ongeveer 11 trekt in ongeveer zeven minuten. Om micro-organismen en onoplosbare deeltjes uit het medium te verwijderen, filtert u het door een filter van 0,22 micron. Breng het medium vervolgens over op een gesloten steriele buis.Inenting een tryptische sojabouillon of TSB vijzelplaat met een geselecteerde pseudomonasstam. Incubeer de plaat 's nachts op 37 graden Celsius. Bereid een buis met 20 milliliter TSB met 5%glycerol als koolstofbron.Verzamel een uitstrijkje van de vijzelplaat en inenting van de TSB. Incubeer de pseudomonas suspensie in een 37 graden Celsius shaker bij 200 RPM, gedurende ongeveer een uur, totdat de optische dichtheid op 600 nanometer 0,6 is. Na het kweken van longeppiliale cellen zoals beschreven in het manuscript, voeg een milliliter van 0,25% trypsine aan de cellen.Na ongeveer vijf minuten, wanneer de cellen volledig zijn losgemaakt van de onderkant van de plaat, voeg 10 milliliter van volledige Hoogten medium om de trypsine te neutraliseren. Breng de cellen over naar een buis van 15 milliliter. Vervolgens, centrifugeren de cellen gedurende vijf minuten op vier graden Celsius en 300 keer G.Verwijder en gooi de supernatant.Dan, opnieuw schorsen de cellen in milliliters van Heights medium met 10%FBS. Pipette 10 microliters van de epitheelcelsuspensie op een nieuwe plaat. Was de gentamycine-behandelde cellen twee maal met twee milliliter koude PBS.Bacteriën belasting in de long epitheelcellen kunnen vervolgens worden gedetecteerd door de druppelplaat methode, qRT-PCR, of stroom cytometrie. Voeg een milliliter cellyse buffer toe aan elke bron van longepepitheelcellen. Herstel seriële verdunningen van het cellysaat, variërend van 1 tot 10 tot 10.000.Inenting vervolgens de TSB vijzelplaat. Na 16 uur incubatie, bepalen hoeveel kolonievormende eenheden aanwezig waren in de longepelleelcellen, door het tellen van het aantal bacteriële kolonies. Voeg 0,35 milliliter van de guanidinium thiocyanaat lyse buffer toe aan elke put van longepepelleëlcellen.Verzamel de cellen met een celschraper. Pipette het lysaat in een microcentrifuge buis en meng zachtjes met de pipet. Voeg 0,35 milliliter vers bereid tot 70% ethanol toe aan het lysaat en meng goed.Breng alle monsters over naar een spinkolom die in een opvangbuis van twee milliliter wordt geplaatst. Verheerlijk de opvangbuis gedurende 30 seconden bij 10, 000 keer G en 20 tot 25 graden Celsius. Gooi de buffer in de opvangbuis weg en was de kolom met 0,7 milliliter wasbuffer.Centrifuge de kolom op 10.000 keer G gedurende 30 seconden. Was de kolom twee maal met 0,5 milliliter buffer. Herhaal de centrifugatie op 10.000 keer G gedurende twee minuten.Plaats de kolom in een nieuwe 1,5 milliliter opvangbuis en voeg 30 tot 50 microliter RNase-vrij water toe. Na het centrifugeren van de buis op 10.000 keer G gedurende een minuut, het verzamelen van de stroom door en meet de RNA-concentratie. Ter voorbereiding op een 20 microliter omgekeerde transcriptie reactie maakt een microgram van de totale RNA met 10 microliter van de reactie buffer, een microliter van omgekeerde transcriptase en RNase-vrij water.Voer de omgekeerde transcriptiereactie bij 37 graden Celsius gedurende een uur en vervolgens op 95 graden Celsius gedurende vijf minuten, volgens het protocol van de fabrikant. Meng vervolgens een microliter van elk cDNA-monster met vijf microliter van de mastermix met cyberkleurstof en één microliter van elke 200 nanomolar specifieke primer. Voeg water toe voor een totaal volume van 20 microliter.Voer PCR-analyse uit, volgens de aanbevelingen van de fabrikant. Gebruik vervolgens de vergelijkende CT-methode om de expressie te bepalen. In BEAS-2B-cellen veranderde de levensvatbaarheid niet aanzienlijk toen cellen gedurende drie uur met 4%CSE werden behandeld.Een uur als pseudomonas infectie ook niet wezenlijk van invloed op de levensvatbaarheid. Ten slotte gebruikten ze gentamycine om pseudomonas te doden in het kweekmedium had geen statistisch significant effect op de levensvatbaarheid van cellen. Zoals gemeten door de druppelplaatmethode, werd bacteriële infectie in BEAS-2B-cellen aanzienlijk verhoogd door behandeling met CSE.Hetzelfde gold voor de behandeling van HSAEC door de CSE. Bacteriële infectie van CSE-behandelde BEAS-2B-cellen werd ook beoordeeld met behulp van RT-PCR om de hoeveelheid 16S rRNA te kwantificeren, wat duidt op een toename van Pseudomonas aeruginosa. De BEAS-2B cellen behandeld met CSE en besmet met Pseudomonas fluorescens Migula werden geanalyseerd door stroom cytometrie op 509 nanometer.Het resultaat gaf ook aan dat CSE behandeling verhoogt bacteriële infectie. Resultaten van fluorescerende microscopie bleek dat GFP-gelabelde bacteriën co-gelokaliseerd met BEAS-2B cellen in een Pseudomonas fluorescens Migula infectie experiment. Cellen kunnen worden blootgesteld aan sigarettenrook met robots roken, die menselijk gedrag kunnen nabootsen en het mogelijk maken om direct de effecten van sigarettenrook op longepelische cellen te bestuderen.Deze techniek kan worden gebruikt om een sigarettenrook op andere celkranen te onderzoeken in de context van een longinfectie.
