هذه الطريقة يقطع تدفق اللمفية مع الحد الأدنى من الضرر للخلايا البطانية اللمفاوية، ومع السيطرة الدقيقة على توقيت الحصار، ويمكن استخدامها لدراسة كيفية تأثير تدفق اللمفية التوازن والاستجابات المناعية في العقدة الليمفاوية. المساعدة في إثبات الإجراء ستكون (جينغنا شيويه) طالبة دراسات عليا من مختبري لإعداد جهاز حقن، وقطع ما يقرب من 30 سم من أنابيب البولي إيثيلين.قم بتوصيل طرف الإبرة A بإحدى أطراف الأنبوب. طرد بعناية إبرة أخرى، وربط الجانب المكسور إلى الطرف الآخر من أنابيب البولي إيثيلين. ثم نعلق إبرة A إلى حقنة درنة درنة المليلتر.قبل الاستخدام مباشرة، قم بإعداد خليط 10:1 الكيتامين-الزيلازين في الملح. بعد تخدير الماوس عن طريق حقن 250 ميكرولترات من خليط الكيتامين / xylazine intraperitoneally، وضمان التخدير الكامل مع قرصة إصبع قدم. حلق الفراء حول الساقين مع مقص الشعر، ثم تطبيق كريم إزالة الشعر حول الساق.بعد خمس دقائق، امسح الفراء المتبقي وكريم إزالة الشعر باستخدام أنسجة رطبة، ثم نظف الساق بالماء العقيم. رش 70٪ الإيثانول حول الساق لتعقيم منطقة العمليات. ضع الفأرة في وضعية عرضة، واستخدم الشريط الجراحي لفضح منطقة العملية على الساق اليمنى.إيندرمالي حقن خمسة ميكرولترات من 1٪ ايفانز صبغة زرقاء، أو تسعة سنتيمترات من السائل من جهاز حقن الأنابيب في لوحة القدم اليمنى من الماوس. تدليك بلطف لوحة القدم لمساعدة السوائل على دخول الأوعية اللمفاوية. تحت مجهر تشريح، تحديد موقع شق خمسة ملليمترات من الحافة السفلية من فوسا popliteal.باستخدام زوج من مقص، وجعل شق صغير ما يقرب من خمسة ملليمترات في الطول. ثم استخدام الدمى غرامة لتمتد شق وفضح الأوعية اللمفاوية جمع. تحديد كل من الأوعية اللمفاوية afferent المؤدية إلى العقد الليمفاوية الزفيرة.باستخدام حامل إبرة، أدخل بحذر إبرة الغرزة بين الأوعية اللمفاوية الوريدية والشريان الصابف. سحب بلطف الإبرة من حول الأوعية اللمفاوية afferent. سحب بعناية سلسلة خياطة، وترك حوالي سنتيمترين من سلسلة خياطة وراء.استخدم حامل الإبرة للمساعدة في ربط عقدة الجراح. تدليك بلطف لوحة القدم لضمان عدم وجود صبغة زرقاء ايفانز يمر موقع الغرز، ثم قطع السلسلة الزائدة مع مقص. خياطة الأوعية اللمفاوية الأخرى، ثم إغلاق شق الجلد مع نفس خياطة التي كانت تستخدم لالأوعية اللمفاوية.للسيطرة صورية، intradermally حقن خمسة ميكرولترات من 1٪ ايفانز صبغة زرقاء في لوحة القدم اليسرى، وتدليك لوحة القدم. افتح الجلد بشق، ثم أغلق الجرح دون خياطة الوعاء. عند مراقبة الماوس بعد الجراحة ، يجب أن تظهر الساق اليمنى وذمة ، مع انتشار صبغة زرقاء إيفانز إلى الفخذ ، في حين أن ساق التحكم ستظهر صبغة زرقاء إيفانز تقتصر على وسادة القدم.مباشرة بعد الجراحة، حقن intradermally 10 ميكرولترات من 2٪ FITC في كل من وسادة القدم اليمنى واليسار. بعد ساعتين وستة و12 ساعة من حقن فيت سي، قم بجمع الغدد الليمفاوية الزجرية من فوسا الفئران القتل الرحيم. إزالة بعناية الأنسجة الدهنية في الأنسجة الدهنية حول pLNs.تضمين pLNs في مجمع درجة حرارة القطع الأمثل مع منطقة الجيوب الأنفية النخاعية النخاعية التي تواجه إلى جانب قالب التبريد. استخدام cryotome لإعداد 20 ميكرون المجمدة المقاطع. لتحديد توزيع FITC في pLNs، صور المقاطع المبردة تحت مجهر confocal.صور Confocal القبض على اثنين وستة و 12 ساعة بعد حقن فيتف تظهر بشكل كبير انخفاض تراكم FITC في الغدد الليمفاوية الزفير بعد خياطة الأوعية اللمفاوية afferent. وتراكمت FITC المتبقية في pLNs بشكل تفضيلي في الجيوب الأنفية العقدة الليمفاوية. الصور Confocal من الأنسجة الدهنية فينودال تظهر عندما يتم حظر الأوعية اللمفاوية عن طريق خياطة، FITC يدخل الأنسجة عندما الجيوب الأنفية العقدة الليمفاوية، لكنه لا يوزع بشكل فعال في جميع أنحاء الغدد الليمفاوية.عند محاولة هذه الطريقة، من المهم أن تتذكر أن إدخال إبرة الغرز بين الأوعية الدموية والأوعية اللمفاوية أمر بالغ الأهمية. فشل هذه الخطوة قد كسر السفن. بعد تنفيذ هذه الطريقة، من الممكن تحصين الفئران بالعدوى أو غيرها من التحفيز المناعي لدراسة كيفية تنظيم التدفق اللمفاوي للحماية المناعية.وظيفة تدفق اللمفاوية ليست مفهومة جيدا. يمكن استخدام هذه الطريقة لدراسة هجرات الخلايا في العقدة الليمفاوية في غياب التدفق اللمفاوي.
