リンパ系の空間組織は、細胞外液を効率的に除去し、抗原および抗原提示細胞(APC)を排出するLNsに輸送するための構造的および機能的なサポートを提供する。初期リンパ管(リンパ管管とも呼ばれる)は、細胞間の不連続な接合により透過性が高く、周囲の細胞外空間から液体、細胞、および他の物質を効果的に収集しやすくする1。最初のリンパ管は、細胞間の接合が緊密で、連続的な基盤膜、リンパ筋のカバレッジを有するリンパ管を採取する。リンパ管の採取は、採取したリンパを排水中のLUNに輸送し、最終的にリンパを循環2,3に戻す役割を担う。リンパを排水LNに推進する集水性リンパ管は、不発泡リンパ管4、5、6、7である。発泡性リンパ管の閉塞は、リンパ流の機能を研究する際に有用な技術であるLNsへのリンパ流を遮断する可能性がある。
これまでの研究では、リンパ流は抗原およびAPCを輸送するだけでなく、LN恒常性を維持する上で重要な役割を果たしていることを示している。組織由来のAPCは、典型的には活性化された回遊樹状細胞(DC)が、不透過性リンパ管を通過してLNに移動し、T細胞8を活性化することがよく理解されている。微生物や可溶性抗原などの自由形抗原が、LNにリンパを持って受動的に流れ、LN居住APcを活性化するという考え方は、過去10年間で9、10、11、12で受け入れられています。リンパを伴って移動する自由形抗原は、感染後数分でLNに移動し、LN常駐細胞活性化は刺激後20分以内に起こり得る。これは、ドレイン LN9に入るのに 8 時間以上かかる DC の移行のアクティブ化よりもはるかに高速です。免疫保護を開始するために抗原を輸送することに加えて、リンパはまた、その微小環境を維持するためにサイトカインおよびDCをLNに運び、免疫細胞恒常性13、14をサポートする。以前は、アフェレントリンパ管を縫合することによりリンパ流を遮断し、LN15、16、17にホメオスタティックT細胞およびB細胞ホーミングをサポートするために必要なHEV表現型を維持するために必要なリンパが必要であることを実証した。CCL21は、LN8、18におけるDCおよびT細胞の位置を指示する重要なケモカインである。リンパ流を遮断すると、LNにおけるCCL21発現が中断し、LN19におけるDCおよびT細胞の位置決めおよび/または相互作用を中断する可能性がある。したがって、リンパ流を遮断すると、LN内の免疫応答を調節するLN微小環境を破壊することによって、抗原/DCのドレインLNへの直接または間接的なアクセスを遮断することができる。リンパ流の機能をより的調するために、フットパッドからpLNに対して、マウスのリンパ流を遮断する実験プロトコル(図1)を提示する。この方法は、健康で病気の状態でリンパ機能に関する将来の研究のための重要な技術となり得る。

この方法は、リンパ内皮細胞への損傷を最小限に抑えてリンパ流を遮断し、遮断タイミングを正確に制御することで、リンパ流がリンパ節における恒常性および免疫応答にどのような影響を与えるかを調べることができる。この手順のデモンストレーションを手伝うのが、私の研究室の大学院生である景納徐さんです。注入装置を準備するために、ポリエチレンチューブのおよそ30センチメートルを切断する。針Aの先端をチューブの一端に接続します。別の針を慎重に取り外し、壊れた側をポリエチレンチューブのもう一方の端に接続します。その後、針Aを1ミリリットルの結核注射器に取り付けます。使用直前に、10:1ケタミンキシラジン混合物を生理食前に調製する。腹腔内にケタミン/キシラジン混合物を250マイクロリットル注入してマウスを麻酔した後、つま先ピンチで完全な麻酔を確実にする。毛のバリカンで脚の周りの毛皮を剃り、脚の周りに脱毛クリームを適用します。5分後、湿った組織を使用して残留ファーと脱毛クリームを拭き取り、滅菌水で脚をきれいにします。脚の周りに70%エタノールをスプレーして、手術領域を殺菌します。マウスを起こしやすい位置に置き、外科用テープを使用して右脚の手術領域を露出させます。皮内に1%エバンスブルー染料の5マイクロリットル、または注射装置から9センチメートルの液体をマウスの右足パッドに注入する。静かにリンパ管に入る流体を助けるためにフットパッドをマッサージします。解剖顕微鏡の下で、大窩窩の下端から5ミリメートルの切開部位を見つける。はさみを使用して、長さ約5ミリメートルの小さな切開を行います。その後、細かい操作鉗子を使用して切開を伸ばし、採取したリンパ管を露出させます。大窩リンパ節に至る両血管の同定。針ホルダーを使用して、眼下リンパ管と伏状動脈の間に縫合針を慎重に挿入する。軽く、尿管のリンパ管の周りに針を引き出します。縫合糸弦を慎重に引っ張り、縫合糸の約2センチメートルを後ろに残します。針ホルダーを使用して、外科医の結び目を結ぶのに役立ちます。エバンスブルー染料が縫合部位を通過しないようにフットパッドを穏やかにマッサージし、余分なひもをはさみで切ります。他の不フェレンチン性リンパ管を縫合し、リンパ管に使用したのと同じ縫合糸で皮膚切開部を閉じる。シャムコントロールのために、皮内に左足パッドに1%エバンスブルー染料の5マイクロリットルを注入し、フットパッドをマッサージする。切開で皮膚を開き、血管を縫合せずに傷口を閉じます。マウスの手術後にマウスを監視する場合、右足は浮腫を示し、エバンスブルー染料は太ももに広がり、コントロールレッグにはエバンスブルーの染料がフットパッドに制限されていることが示されます。手術直後、右足パッドと左パッドの両方に2%FITCの10マイクロリットルを皮内に注入する。FITC注射の2、6、および12時間後に、安楽死させたマウスの窩から亜大リンパ節を採取する。pRNの周りには、ペリノダル脂肪組織を慎重に取り除いてください。pRNを最適な切断温度化合物に埋め込み、髄質洞領域をクライオモールドの側面に向けます。凍結煙を使用して、20ミクロンの凍結したセクションを準備します。pRN 内の FITC 分布を決定するには、共焦点顕微鏡の下でクライオセクションを画像化します。FITC注射の2、6、および12時間後に撮影された共焦点画像は、不同性リンパ管を縫合した後の、大小リンパ節におけるFITC蓄積を大幅に減少させた。pRN中の残留FITCは、リンパ節副鼻腔に優先的に蓄積された。リンパ管が縫合によって閉塞されると、リンパ節脂肪組織の共焦点画像が示され、リンパ節が副鼻腔化するとFITCが組織に入るが、リンパ節全体に効果的に分布していない。この方法を試みるとき、血管とリンパ血管の間に縫合針を挿入することが重要であることを覚えておくことが重要です。このステップの失敗は、船舶を壊す可能性があります。この方法を行った後、感染または他の免疫刺激を有するマウスを免疫し、リンパ流が免疫保護を調節する方法を研究することが可能である。リンパ流の機能はよく理解されていない。この方法は、リンパ流がない場合のリンパ節における細胞の移動を研究するために使用することができる。
