Kræft er en af de dødeligste sygdomme, der forårsager millioner af dødsfald hvert år på verdensplan og enorme økonomiske tab1. I klinikker, konventionelle anticancer behandlinger, såsom kirurgisk resektion, strålebehandling, og kemoterapi stadig ikke kan give en tilfredsstillende terapeutisk effekt2. Begrænsninger af disse behandlinger er høje toksiske bivirkninger, høj gentagelsesrate og høj metastaserhastighed3. For eksempel lider kemoterapi af den lave leveringseffektivitet af kemomedicin netop til tumorstedet4. Den manglende evne til lægemidler til at trænge dybt ind i tumorvævet på tværs af de biologiske barrierer, herunder ekstracellulær matrix og højt tumor interstitielt væsketryk, er også ansvarlig for den lave terapeutiske effekt5. Desuden sker tumorresistensen normalt hos de patienter, der modtog behandling med enkelt kemoterapi6. Derfor har teknikker, hvor termisk ablation af tumor opstår, såsom fototermisk behandling (PTT) og magnetisk hypertermi terapi (MHT), vist lovende resultater for at reducere tumorresistens og er dukket op i kliniske forsøg7,8,9.
PTT udløser termisk ablation af kræftceller ved virkningen af fototermiske konverteringsmidler under bestråling af laserenergien. Den genererede høje temperatur (over 50 °C) fremkalder fuldstændigcellenekrose 10. For ganske nylig blev jernoxid nanopartikler (IONPs) påvist at være et fototermisk konverteringsmiddel, der kan aktiveres af nær-infrarødt (NIR) lys11.  På trods af den lave molarabsorptionskoefficient i det nær-infrarøde område er IONP'er kandidater til fototermisk behandling ved lav temperatur (43 °C), en modificeret behandling for at reducere skader forårsaget af varmeeksponering for normalt væv og til at indlede antitumorimmunitet mod tumormetastase12. En af begrænsningerne ved TOT er laserens lave indtrængningsdybde. For dybtliggende tumorer, vekslende magnetfelt (AFM) induceret opvarmning af jernoxid nanopartikler, også kaldet magnetisk hypertermi, er en alternativ behandling for PTT13,14. Den største fordel ved MHT er den høje indtrængning af magnetfelt15. Den krævede relativt høje koncentration af IONP'er er dog fortsat en væsentlig ulempe for dens kliniske anvendelse. Leveringseffektiviteten af nanomedicin (eller nanopartikler) til faste tumorer hos dyr har været 1-10% på grund af en række forhindringer, herunder omsætning, akkumulering og penetration16,17. Derfor er en kontrolleret og målrettet IONPs leveringsstrategi med evnen til at opnå høj vævspenetration af stor interesse for kræftbehandling.
Ultralyd medieret nanopartikler levering har vist sin evne til at lette indtrængen af nanopartikler dybt ind i tumorvævet, på grund af fænomenet kaldet microbubble kavitation18,19. I denne undersøgelse fremstiller vi IONP'er afskallede mikrobobler (NSMs) gennem selvmontering, synergizing magnetisk, akustisk og optisk lydhørhed i en nanoterapeutisk platform. NSM indeholder en luftkerne og en skal af jernoxid nanopartikler med en diameter på ca. 5,4 μm. NSMs kan magnetisk guides til tumorstedet. Derefter udløses frigivelsen af IONPs af ultralyd, ledsaget af mikrobubble kavitation og mikrostreaming. Det momentum, der modtages fra mikrostreaming letter indtrængen af IONPs i tumorvævet. TOT og MHT kan opnås ved NIR laser bestråling eller AFM ansøgning, eller med kombinationen af begge.

Præcision levering af anti-cancer agenter, der sigter mod målrettet og dybt trængt levering samt en kontrolleret frigivelse på tumor site er blevet udfordret. Her fremstiller vi nanopartikler af jernoxidskaller (NSMs) gennem selvmontering, synergiserende magnetisk, akustisk og optisk lydhørhed i en nanoterapeutisk platform. Jernoxid nanopartikler tjener som både magnetiske og fototermiske midler. Når intravenøst injiceres, NSMs kan magnetisk guides til tumor site. Ultralyd udløser frigivelsen af jernoxid nanopartikler, hvilket letter indtrængen af nanopartikler dybt ind i tumoren på grund af kavitationseffekten af mikrobubbles.
