Hier presenteerden we een protocol voor het fabriceren van ijzeroxide nanodeeltjes omhulde microbubbels (NSM's) door middel van zelfassemblage, synergetisch magnetische, akoestische en optische responsiviteit in één nanotherapeutisch platform. De IONP's waren dicht opeengepakt rond de luchtkern om een magnetische schil te vormen, die kan worden bestuurd door het externe magnetische veld voor targeting. Eenmaal geleverd, kan de afgifte van IONP's worden bereikt door ultrasone trigger. De vrijgekomen IONP's kunnen worden...

Kanker is een van de dodelijkste ziekten, die wereldwijd wereldwijd miljoenen doden en enorme economische verliezen veroorzaakt1. In klinieken kunnen conventionele antikankertherapieën, zoals chirurgische resectie, radiotherapie en chemotherapie nog steeds geen bevredigende therapeutische werkzaamheid bieden2. Beperkingen van deze therapieën zijn hoge toxische bijwerkingen, hoog recidiefpercentage en hoge metastase3. Chemotherapie lijdt bijvoorbeeld aan de lage afgifte-efficiëntie van chemogeneesmiddelen precies op de tumorplaats4. Het onvermogen van geneesmiddelen om diep in het tumorweefsel door te dringen over de biologische barrières, waaronder extracellulaire matrix en hoge tumorinterstitiële vloeistofdruk, is ook verantwoordelijk voor de lage therapeutische werkzaamheid5. Bovendien treedt de tumorresistentie meestal op bij de patiënten die werden behandeld met enkele chemotherapie6. Daarom hebben technieken waarbij thermische ablatie van tumor optreedt, zoals fotothermale therapie (PTT) en magnetische hyperthermietherapie (MHT), veelbelovende resultaten laten zien om tumorresistentie te verminderen en zijn ze opgekomen in klinische onderzoeken7,8,9.
PTT veroorzaakt thermische ablatie van kankercellen door de werking van fotothermische conversiemiddelen onder de bestraling van de laserenergie. De gegenereerde hoge temperatuur (boven 50 °C) induceert volledige celnecrose10. Zeer recent werd aangetoond dat ijzeroxide nanodeeltjes (IONP's) een fotothermisch conversiemiddel zijn dat kan worden geactiveerd door nabij-infrarood (NIR) licht11.  Ondanks de lage molaire absorptiecoëfficiënt in het nabij-infrarode gebied, zijn IONP's kandidaten voor lage temperatuur (43 °C) fotothermale therapie, een gemodificeerde therapie om de schade veroorzaakt door blootstelling aan hitte aan normale weefsels te verminderen en om antitumorimmuniteit tegen tumormetastase te initiëren12. Een van de beperkingen van PTT is de lage penetratiediepte van de laser. Voor diepgewortelde tumoren is door alternating magnetic field (AFM) geïnduceerde verwarming van ijzeroxide nanodeeltjes, ook wel magnetische hyperthermie genoemd, een alternatieve therapie voor PTT13,14. Het belangrijkste voordeel van MHT is de hoge penetratie van magnetisch veld15. De vereiste relatief hoge concentratie IONP's blijft echter een groot nadeel voor de klinische toepassing ervan. De afgifte-efficiëntie van nanogeneeskunde (of nanodeeltjes) aan vaste tumoren bij dieren is 1-10% geweest als gevolg van een reeks obstakels, waaronder circulatie, accumulatie enpenetratie 16,17. Daarom is een gecontroleerde en gerichte IONPs-toedieningsstrategie met het vermogen om een hoge weefselpenetratie te bereiken van groot belang bij de behandeling van kanker.
Echografie gemedieerde nanodeeltjesafgifte heeft aangetoond dat het de penetratie van nanodeeltjes diep in het tumorweefsel vergemakkelijkt, vanwege het fenomeen dat microbubbelcavitatie wordt genoemd18,19. In de huidige studie fabriceren we IONPs shelled microbubbles (NSM's) door middel van zelfassemblage, synergetische magnetische, akoestische en optische responsiviteit in één nanotherapeutisch platform. De NSM bevat een luchtkern en een schil van ijzeroxide nanodeeltjes, met een diameter van ongeveer 5,4 μm. De NSM's kunnen magnetisch naar de tumorplaats worden geleid. Vervolgens wordt de afgifte van IONP's geactiveerd door echografie, vergezeld van microbubbelcavitatie en microstreaming. Het momentum ontvangen van de microstreaming vergemakkelijkt de penetratie van IONP's in het tumorweefsel. De PTT en MHT kunnen worden bereikt door NIR-laserbestraling of AFM-toepassing, of met de combinatie van beide.

magnetic-, acoustic-, and optical-triple-responsive microbubbles for magnetic hyperthermia and pothotothermal combination cancer therapy

De precisieafgifte van antikankermiddelen die gericht zijn op gerichte en diep gepenetreerde toediening en een gecontroleerde afgifte op de tumorplaats is uitgedaagd. Hier fabriceren we ijzeroxide nanodeeltjes omhulde microbubbels (NSM's) door middel van zelfassemblage, synergetische magnetische, akoestische en optische responsiviteit in één nanotherapeutisch platform. IJzeroxide nanodeeltjes dienen als zowel magnetische als fotothermische middelen. Eenmaal intraveneus geïnjecteerd, kunnen NSM's magnetisch naar de tumorplaats worden geleid. Echografie veroorzaakt de afgifte van ijzeroxide nanodeeltjes, waardoor de penetratie van nanodeeltjes diep in de tumor wordt vergemakkelijkt als gevolg van het cavitatie-effect van microbubbels. Daarna kunnen magnetische hyperthermie en fotothermische therapie worden uitgevoerd op de tumor voor combinatiekankertherapie, een oplossing voor kankerresistentie als gevolg van de tumorheterogeniteit. In dit protocol werd de synthese en karakterisering van NSM's inclusief structurele, chemische, magnetische en akoestische eigenschappen uitgevoerd. Bovendien werd de werkzaamheid tegen kanker door thermische therapie onderzocht met behulp van in vitro celculturen. De voorgestelde toedieningsstrategie en combinatietherapie zijn veelbelovend in de behandeling van kanker om zowel de levering als de werkzaamheid tegen kanker te verbeteren.

De drievoudige responsieve nanodeeltjes-shelled microbubbels (NSM's) die in deze studie werden gebruikt, werden bereid door het mengsel van de oppervlakteactieve stof en IONP's te roeren. De IONP's (50 nm) assembleerden zichzelf op het grensvlak van vloeistof- en gaskern, om een dicht opeengepakte magnetische schil te vormen. De morfologie van NSM's is weergegeven in figuur 1A. De resulterende NSM's presenteerden een bolvorm en met een gemiddelde diameter van 5,41 ± 1,78 μm(figuur 1B)....
