심방 및 심실 조직에서 심근세포 및 비근구의 적절하고 복제적인 절연을 확인하기 위해, 세포는 문화권에서 다양한 날에 관찰 및 촬영하였다(도2). 또한, 정량적 역전사 폴리머라제 연쇄 반응(qRT-PCR)은 세포 유형 특이적 전사의 수준을 측정하기 위해 수행되었다. 심장 근육 트로포닌 T(Tnnt2)는심장 심근 세포의 마커이며 심방 및 심실 심장 심근 세포 배양(도 3A)에서강력하게 발현되었다. 대조적으로, 심방 나트리요성펩티드(Nppa는,전형적으로 생리적인 조건 하에서 성인 심방 심장 근세포에서 독점적으로 표현되는) 및 근신광사슬2(Myl2,심실 심세포 특이 유전자)는 각각 심방 및 심실 심장 심근세포 배양에서 견고하고 구체적으로 발현하였다(그림3,C).
섬유아세포 마커, 전사 인자(21),혈소판 유래 성장 인자 수용체 A(Pdgfra) 및 단핵 유래 세포 마커 클러스터(68(Cd68)는 심방 및 심실 챔버(도3D-F)로부터분리된 비심낭 배양물에서 독점적으로 발현되었다. 비근구가 모든 심장 세포의 ~65%를 손상시키며 이들 중 대다수가 섬유아세포 또는 단핵구 유래혈통(18,19,23,24)에서유래한 것으로 추정된다. 따라서, 이 두 혈통에 대한 마커는 심장 병리학의 다양한 모델 및 병인의 연구에서 이러한 세포 집단에 대한 관심을 감안할 때 대표로 선택되었다.
t-튜블러 마커 디하이드로피리딘(DHPR)에 대한 AMAMMs 및 AMVM의 면역염색(DHPR은 전압 의존성(L)형 칼슘 채널)과 라야노딘 수용체(RYR2)를 통해 절연 및 장기 배양(도4A,B)을통해 손상되지 않은 t-tubules를 입증하였다. 심방과 심실 심근 세포에 특이하고 독특한 DHPR및 국소화의 풍요로움은 t-tubules의 존재를 표시했습니다. 더욱이, RYR2 면역염색을 가진 DHPR의 공동지역화는 그대로 이화된 구조물의 표시기이었습니다. 심방 및 심실 심장 근세포에서 육종 단백질 알파 액티닌에 대한 면역 염색은 예상 육종 자구 패턴의 결과. 육종자 살리기 패턴은 TOPRO-3(도4C, D;보라색 및 빨간색)와 함께 막대 모양의 형태 및 핵 염색과 함께 분리된 심장 근구의 순도와 생존가능성을 평가하는 데 사용되었습니다. 예상대로 심실 심장 심근세포는 평균 ~150mm의 길이를 나타내는 반면, 심방 심장 심근세포는 평균 ~75mm를 나타냈다. 더욱이, 면역염색 분석에 따라, 심방 심장 근세포(심실 심장 근세포는 아님)는 내피성 망상및 분비 과립에 국소화의 특징이었던 염색 패턴에서 심방 내트리요성 펩티드(ANP)의 견고한 발현을나타냈다(도 4C,D;녹색).
심방 심장 심근세포에 독특한 특징은 수축 세포 이외에 내분비 세포로서의 분류입니다. 심방 근세포는 기저 조건 하에서 ANP를 분비하는 동안, 분비분비 (즉, 알파-adrenergic 작용제, 페닐레프린 [PE])에 대한 응답으로 증가합니다. 더욱이, 심방 심근세포는 ANP를 분비하고 호르몬의 일부를 전구체 상태(Pro-ANP, 15 kD)로부터 펩타이드(ANP 3kD)16,17로공동 처리한다. 이러한 분비 능력은 급성 PE 치료에 대한 응답으로 격리된 심방 심장 근세포의 매체에서 ANP의 면역블롯 검출을 통해 정량화될 수있다(도 4E). 심방 심장 근세포의 이 분비 및 처리 능력은 배양 조건에 민감한 것으로 나타났습니다. 따라서, 심방 심근세포 도금 매체는 덱사메타손, 인슐린, 이송린 및 셀레늄으로 보충되는 것이 필수적이다.
