measuring the confluence of ipscs using an automated imaging system

تركز هذه الدراسة على فهم كيف يمكن أن تؤثر زراعة iPSCs على ركائز طلاء ECM المختلفة على التقاء الخلايا. تم إنشاء بروتوكول لتقييم التقاء iPSC في الوقت الفعلي دون الحاجة إلى عد الخلايا في تعليق خلية واحدة لتجنب أي اضطراب في النمو. تم استخدام نظام تحليل الصور عالي المحتوى لتقييم التقاء iPCS على 4 ECMs مختلفة بمرور الوقت بطريقة آلية. تم استخدام إعدادات تحليل مختلفة لتقييم التقاء الخلايا من iPSCs الملتصقة ولم يلاحظ سوى اختلاف طفيف (في 24 و 48 ساعة مع laminin) سواء تم تطبيق قناع 60 أو 80 أو 100٪. نظهر أيضا أن laminin يؤدي إلى أفضل التقاء مقارنة ب Matrigel و vitronectin و fibronectin.
