תאי גזע פלוריפוטנטיים מושרים (iPSCs) מתקבלים מתאים סומטיים וניתן למיין אותם לסוגי תאים שונים. הם משמשים לעתים קרובות כמערכת למודל פתוגנזה של מחלות או לביצוע בדיקת תרופות, וגם מציעים את הפוטנציאל לשמש בהקשר של רפואה מותאמת אישית. מאז iPSCs יש פוטנציאל גדול, חשוב לאפיין אותם באופן מלא לשימוש כמערכת מודל אמין. בעבר הראינו את החשיבות של גידול תאי גזע פלוריפוסיים בסביבה היפוקסית, שכן תאים אלה מסתמכים על גליקוליזה וסביבה אירובית עלולה לגרום לחוסר איזון חמצון-חיזור1. תאי גזע פלוריפוטנטיים עצביים פגיעים גם לתנאי תרבית אחרים, במיוחד לסביבה החוץ-תאית. אופטימיזציה של תנאי התרבות היא נושא מרכזי כדי לשמור עליהם בריאים ומתרבים. תרבית iPSC בריאה תוביל לתאים ממוינים בריאים שהם בדרך כלל נקודת הקצה של המודל המשמש להבנת תכונות מולקולריות, תאיות ותפקודיות של הפרעות אנושיות ספציפיות או תהליכים תאיים.

measuring the confluence of ipscs using an automated imaging system

מחקר זה מתמקד בהבנת האופן שבו גידול תאי גזע פלוריפוטנטיים על מצעי ציפוי ECM שונים יכול להשפיע על מפגש התאים. פרוטוקול להערכת מפגש iPSC בזמן אמת נקבע ללא צורך לספור תאים בתרחיף של תא בודד כדי למנוע כל הפרעה בגדילה. מערכת ניתוח תמונות בעלת תוכן גבוה שימשה להערכת מפגש iPCS על 4 ECMs שונים לאורך זמן באופן אוטומטי. הגדרות ניתוח שונות שימשו להערכת מפגש תאים של iPSCs דבקים ורק הבדל קל (ב 24 ו 48 שעות עם laminin) נצפתה אם מסכה 60, 80 או 100% הוחל. אנו גם מראים כי laminin להוביל את המפגש הטוב ביותר לעומת Matrigel, vitronectin ו fibronectin.

במחקר זה חקרנו את המפגש של תאי גזע פלוריפוטנטיים (iPSCs) כאשר הם גדלים בתנאי ציפוי שונים. באמצעות ציטומטר, הצלחנו להשיג תוצאות אינפורמטיביות בקלות במשולשים תוך 5 ימים. מאחר שתאי גזע פלוריפוטנטיים כמעט ואינם נצמדים לכלי פלסטיק ויש צורך בציפוי כדי לתמוך בהתפשטותם, החלטנו לעקוב אחר המפגש של תאי ...
