measuring the confluence of ipscs using an automated imaging system

이 연구는 다양한 ECM 코팅 기판에서 성장하는 iPSC가 세포 합류에 어떤 영향을 미칠 수 있는지 이해하는 데 중점을 둡니다. 성장 섭동을 피하기 위해 단일 세포 현탁액의 세포를 계산할 필요 없이 실시간으로 iPSC 합류를 평가하는 프로토콜이 확립되었습니다. 고함량 이미지 분석 시스템을 사용하여 시간이 지남에 따라 4개의 서로 다른 ECM에서 iPCS 합류를 자동화된 방식으로 평가했습니다. 부착성 iPSC의 세포 합류를 평가하기 위해 다양한 분석 설정을 사용했으며 60, 80 또는 100% 마스크가 적용되었는지 여부에 관계없이 약간의 차이(라미닌의 경우 24시간 및 48시간)만 관찰되었습니다. 우리는 또한 라미닌이 Matrigel, vitronectin 및 fibronectin에 비해 가장 좋은 합류로 이어진다는 것을 보여줍니다.
