Bruken av iPSC for sykdomsmodellering og fremtidig legemiddelscreening sammen med deres mulige anvendelse i presisjonsmedisin gjør det til en teknologi av stor relevans, og av denne grunn tror vi at det er nødvendig å tydelig forstå in vitro-dyrkingstilstanden som bedre ligner den fysiologiske situasjonen til embryonale stamceller. Induserte pluripotente stamceller (iPSCs) er hentet fra somatiske celler og kan differensieres i forskjellige celletyper. De brukes ofte som et system for å modellere sykdomspatogenese eller utføre narkotikascreening, og tilbyr også potensialet til å bli brukt i sammenheng med personlig medisin. Siden iPSC-er har stort potensial, er det viktig å karakterisere dem fullt ut for bruk som et pålitelig modellsystem. Vi har tidligere vist viktigheten av å dyrke iPSC-er i et hypoksisk miljø, da disse cellene er avhengige av glykolyse og et aerobt miljø kan forårsake redoks ubalanse1. iPSC-er er også sårbare for andre kulturforhold, spesielt det ekstracellulære miljøet. Optimalisering av kulturforhold er et sentralt tema for å holde dem sunne og spredende. En sunn iPSC-kultur vil føre til sunne differensierte celler som generelt er endepunktet for modellen som brukes til å forstå molekylære, cellulære og funksjonelle trekk ved spesifikke menneskelige lidelser eller cellulære prosesser.
I denne studien har en enkel protokoll blitt brukt til å teste sammenløpet av iPSC-er ved hjelp av forskjellige beleggforhold i separate brønner. iPSCs krever et materlag av murine embryonale fibroblaster (MEF) for å festes riktig, men sameksistensen av iPSCs og MEF gjør det vanskelig å utføre analyser som RNA eller proteinekstraksjon siden to populasjoner av celler er tilstede. For å unngå materlaget har forskjellige proteiner som tilhører den ekstracellulære matrisen (ECM) blitt brukt til å gjenskape den naturlige cellenisjen og å ha materfri iPSC-kultur. Spesielt er Matrigel et løselig kjellermembranpreparat ekstrahert fra Engelbreth-Holm-Swarm (EHS) musesarkom, som er beriket i ekstracellulære matriksproteiner (dvs. laminin, kollagen IV, heparansulfatproteoglykaner, entactin / nidogen og vekstfaktorer)2,3. De andre brukte beleggforholdene er i stedet rensede proteiner med kjent relevans i byggingen av ECM-ene: laminin-521 er kjent for å bli utskilt av humane pluripotente stamceller (hPSCs) i embryoets indre cellemasse, og det er en av de vanligste lamininene i kroppen etter fødselen 4,5,6,7,8,9, 10,11; vitronektin er en xenofri cellekulturmatrise kjent for å støtte vekst og differensiering av hPSC 12,13,14,15,16; fibronektin er et ECM-protein som er viktig for virveldyrutvikling og vedlegg og vedlikehold av embryonale stamceller i en pluripotent tilstand 17,18,19,20,21,22,23,24,25. measuring the confluence of ipscs using an automated imaging system

Denne studien fokuserer på å forstå hvordan voksende iPSC-er på forskjellige ECM-beleggsubstrater kan påvirke cellekonfluens. En protokoll for å vurdere iPSC-sammenløp i sanntid er etablert uten behov for å telle celler i enkeltcellesuspensjon for å unngå vekstforstyrrelser. Et bildeanalysesystem med høyt innhold ble brukt til å vurdere iPCS-sammenløp på 4 forskjellige ECM-er over tid på en automatisert måte. Ulike analyseinnstillinger ble brukt til å vurdere cellekonfluens av adherente iPSC, og bare en liten forskjell (ved 24 og 48 timer med laminin) er observert om en 60, 80 eller 100% maske ble påført. Vi viser også at laminin fører til best samløp sammenlignet med Matrigel, vitronektin og fibronektin.

I denne studien undersøkte vi iPSCs-sammenløp når de dyrkes på forskjellige beleggforhold. Ved hjelp av et cytometer kunne vi oppnå lett informative resultater i tre eksemplarer på 5 dager. Siden iPSC-er knapt festes til plastbeholdere og et belegg er nødvendig for å støtte spredningen, bestemte vi oss for å overvåke sammenløpet av humane iPSC-er, da det indikerer helsen til cellekulturen, og det kan reflektere over deres differensieringspotensial.
