本研究の目的は、生物学的研究および臨床応用のために、成人皮膚の表皮からメラノサイトを分離するためのシンプルで効果的なプロトコルを開発することであった。皮膚の基底表皮や毛包に位置するメラノサイトは、メラニン1を産生することによって皮膚および毛髪の色素沈着に重要な役割を果たす。表皮メラノサイトから得られた皮膚色素沈着は、皮膚2の下層細胞へのDNA損傷を減少/予防する紫外線照射フィルターとして作用する。皮膚におけるメラノサイトの異常増殖は、良性ネビ(ほくろ)の形成など、メラノサイトが発癌性増殖に変わる可能性があり、その後に細胞老化が続くなど、非常に一般的である。
1957年以来、ヒト原発メラノサイトの分離とその後の培養は4つ可能であったが、1982年以来、表皮5からヒトメラノサイトの培養を再現的に確立できる効率的な方法があった。表皮から一次メラノサイトを分離する従来の方法は、2段階の酵素消化を伴う。簡単に言えば、皮膚は最初に表皮を真皮から分離するためにディスパスで消化され、その後表皮をトリプシンで消化してメラノサイトおよびケラチノサイトを含む懸濁液を産生し、異なる培地で選択的に成長させることができる。現在、初期培養は通常、メラノサイトが従来の方法を用いて合流に達するまでに約3〜4週間かかり、これはそれらの単離の効率が低いためである可能性が高い。したがって、成人の皮膚組織からのメラノサイトの初期産生を増加させることは、実験室での研究と臨床応用の両方に非常に有用であろう。
多くの成長因子は、その大部分が角化細胞(例えば、α-MSH、ACTH、bFGF、NGF、ET-1、GM-CSF、HGF、LIFおよびSCF)によって分泌されることが示されている55-8、8哺乳類メラノサイトの分化および増殖を調節する。フィーダー層がない場合、これらの成長因子の欠如は、メラノサイトの増殖の減少およびそれらの増加したアポトーシス9につながる。ケラチノサイトをフィーダー細胞とメラノサイトとの共培養は、メラノサイトの増殖を加速し、アポトーシスを減少させる可能性がある。しかし、この共培養法は、より多くの皮膚組織を必要とし、ケラチノサイトを調製する必要があるが、これは実用的ではないが、メラノサイトに必要な培養条件がケラチノサイトの成長を好まないため、効率的に働くことができなかった。
これまでの研究では、ROCK阻害剤であるY-27632を成長培地に添加することで、皮膚組織10~14-14からヒト初代表皮細胞の収率を高めることができることが報告されている。したがって、ヒトの原発メラノサイトの単離がY-27632の存在から恩恵を受けるかどうかをテストすることは興味深いであろう。実際に、TpA、IBMX、Na3VO4およびdbcAMPを含む接種TIVA培地15にY-27632を添加すると、初期培養316において包皮組織から分離された一次メラノサイトの収率が有意に増加した。従来のプロトコルと比較して、この新しいプロトコルはメラノサイト16の収率を上げる上でかなり効率的である。したがって、このプロトコルは、基礎的および応用生物学的研究におけるその応用を促進するために、以前の研究16に基づいて成人皮膚組織を用いて新しい方法を詳細に記述する。
