これは、細胞間カルシウムシグナル伝達の研究のための第一次脳の脳の分離および培養のための簡単で効果的な方法です。得られた細胞培養は、ほぼ均質な近母細胞集団であり、カルシウムイメージングのためにペリサイトをロードするのは簡単です。ここで説明する方法は、この分野の他の研究者に、ペリサイト生物学を研究するための強力なツールと、体外の細胞内シグナル伝達を提供する必要があります。37°Cの水浴で毛細血管の1バイアルを解凍して始めます。毛細血管が解凍されたら、30ミリリットルのDMEMコンプを備えた遠心管に移し、G.500回5分間遠心分離機を出し、チューブから培地を取り出し、新鮮なDMEMコンプを10ミリリットルでパレットを再中断します。懸濁液をコーティングされたT-75フラスコに移し、毛細血管が10%の二酸化炭素を供給する37°Cインキュベーターで4〜6時間底に付着するようにします。インキュベーション後、フラスコを光顕微鏡で検査します。毛細血管の分画はフラスコの底部に付着する必要があります。毛細血管を播種した4日目に、顕微鏡下で検査し、約60〜70%コンフルエントであることを確認します。培地を吸引し、PBSで細胞を静かに洗います。解凍したトリプシン-EDTAを2ミリリットル加え、フラスコを1〜3分間保育器に残し、フラスコを頻繁に取り出し、顕微鏡で細胞剥離を観察する。内皮細胞が切り上がり始めたら、フラスコを軽くたたいて取り外します。細胞が剥離したら、フラスコに10ミリリットルのDMEMコンプを加えてトリプシン化を止める。フラスコを培地で数回洗い流し、内皮細胞懸濁液を吸引し、他の目的に使用できるようになりました。フラスコに10ミリリットルのDMEMコンプを加え、光顕微鏡の下でチェックして、ペリサイトがまだ存在し、底に取り付けられていることを確認します。次に、フラスコをインキュベーターに戻して、ペリサイトを豊かにした培養物を成長させます。コーティングされた96ウェルプレートにペリサイトを播種するには、テキスト原稿に記載されているように取り外します。培地を吸引し、細胞ペレットを乱さないよう気をつけて、新鮮なDMEMコンプの1ミリリットルでペレットを再懸濁した。計数チャンバーを使用して細胞を数え、ウェルあたり10,000個の細胞をシードするために各ウェルに添加すべき懸濁液の体積を計算します。細胞懸濁液を加え、十分なDMEMコンプを細胞に加え、ウェルあたり200マイクロリットルの総体積を達成します。フラ-2 AMカルシウムインジケーター染料で海中球をロードする準備ができたら、培養器から細胞と96ウェルプレートを取り出し、ウェルから培地を吸引します。細胞をアッセイバッファーで2回洗浄し、各ウェルに100マイクロリットルのローディング溶液を加えます。写真の漂白を避けるためにtinfoilでプレートを包み、室温で30 RPMの揺れをして45分間インキュベートします。インキュベーション後、ローディングバッファーを吸引し、アッセイバッファーで細胞を2回洗浄する。100マイクロリットルの新鮮なアッセイバッファーを加え、細胞を残して30分間インキュベートします。インキュベーション後、プレートリーダーの温度を摂氏37度に設定し、細胞を含む96ウェルプレートをサンプルプレート位置に移し、次に試薬プレート位置にアゴニストを用いて試薬プレートを置きます。まず、細胞の積み込みを測定して、すべてのウェルでのFura-2 AMの等負荷を確保し、340〜380ナノメートルの励起蛍光波長、510ナノメートルの発光波長で測定を行います。50マイクロリットルのアゴニストを、毎秒150マイクロリットルの速度で、試薬プレートから細胞を用いた各ウェルに加える。カバースリップをセルチャンバーに取り付け、漏れを避けるために締めます。DMEMコンプと計算されたセルサスペンションの体積を各チャンバに加え、最終的な体積は500マイクロリットルです。セルチャンバーを摂氏37度、炭酸ガス10%で6日間、またはコンフルエントになるまでインキュベートします。細胞がコンフルエントになったら、培養器から細胞室を取り出し、培地を吸引する。細胞をアッセイバッファーで2回洗浄し、各チャンバに500マイクロリットルのローディングバッファーを加え、室温で45分間インキュベートします。インキュベーション後、ローディングバッファーを吸引し、アッセイバッファーで細胞を2回洗浄し、次に各チャンバーに500マイクロリットルの新鮮なアッセイバッファーを加え、室温でさらに30分間インキュベートし、AMエステルの切断を可能にする。バッファーを500マイクロリットルの新鮮なアッセイバッファーに交換し、共焦点顕微鏡でのライブイメージングを進めます。細胞の乱れを避けるために、顕微鏡のステージ上にできるだけ穏やかに取り付けます。励起波長を488ナノメートルに、発光を515ナノメートルに、5秒間隔で2分間連続画像取得し、x、y画像サイズを512 x 512ピクセルに設定します。細胞室に100ミリモルATPの300マイクロリットルを加え、画像の取得を開始します。このプロトコルは、牛の脳毛細血管からペリサイトを浄化するために使用されました。播種毛細血管からの細胞の成長は、9日間にわたって画像化された。毛細血管は1日目にフラスコに完全に付着し、2日目までに内皮発芽が見えた。4日後、細胞の成長は特徴的であり、内皮細胞はトリプシン法を介して除去された。毛細血管の残骸はトリプシン化後に存在していたが、次の日にフラスコから消失した。内皮層を除去した後、包皮細胞は合流するまで増殖させた。9日目には、海中球が約80%の合流に達し、島を形成しました。フラ-2にATPを添加すると、細胞ホルモンカルシウム濃度が増加した。反応は、ATPを周皮細胞に添加した直後に起こり、測定された期間にわたってゆっくりと低下した。カルシウム応答もリアルタイム共焦点イメージングで測定した。ベースライン蛍光を測定し、その後ATPを64秒で添加し、強い細胞内カルシウム応答を明らかにした。その直後、細胞内で細胞内に区画化された細胞体カルシウム、および細胞領域の減少が見られた。300秒までに細胞面積は大幅に減少し、蛍光はほぼベースラインレベルまで低下した。この方法は、初代脳毛細血管の脳内細胞から最初の細胞内カルシウム測定を得るために使用され、ATPを介して心球が刺激され、そしてインビトロで収縮できることを実証した。
