A pesar de ser un proceso altamente complejo e ineficiente1, la metástasis es un contribuyente significativo a la morbilidad y mortalidad de los pacientes con cáncer2. De hecho, la mayoría de las muertes relacionadas con el cáncer se atribuyen a la propagación metastásica de la enfermedad3,4. Para que las células tumorales hagan metástasis con éxito, deben desprenderse del sitio primario, invadir a través del estroma contiguo, intravasar en la circulación sanguínea o linfática, viajar al lecho capilar de un sitio secundario, extravasar en el tejido secundario y proliferar o crecer para formar lesiones metastásicas5. El uso de modelos de ratón ha sido fundamental para avanzar en la comprensión de los mecanismos moleculares responsables de la siembra y el crecimiento metastásico6,7. Aquí, nos centramos en la metástasis del cáncer de mama, para la cual a menudo se utilizan tanto modelos de ratón modificados genéticamente como métodos de trasplante, cada uno con su propio conjunto de ventajas y limitaciones.
Los modelos de tumores mamarios genéticamente modificados hacen uso de promotores específicos de la glándula mamaria, incluyendo MMTV-LTR (repetición terminal larga del virus tumoral mamario de ratón) y WAP (Proteína ácida de suero de leche), para impulsar la expresión de transgenes en el epitelio mamario8. Además, estos modelos genéticos de ratón son a menudo más prohibitivos en tiempo y costo que los modelos de metástasis espontáneas o experimentales. A pesar de que la comunidad investigadora ha logrado avances significativos en la comprensión del complejo proceso de la metástasis del cáncer de mama, se estima que más de 150.000 mujeres tienen actualmente cáncer de mama metastásico16. De las pacientes con cáncer de mama en estadio IV, >36% de las pacientes presentan metástasis pulmonar17; sin embargo, el patrón específico del sitio y la incidencia de metástasis pueden variar según el subtipo molecular18,19,20,21. Las pacientes con metástasis pulmonares asociadas al cáncer de mama tienen una mediana de supervivencia de solo 21 meses, lo que pone de manifiesto la necesidad de identificar tratamientos efectivos y nuevos biomarcadores para esta enfermedad17. El uso de modelos experimentales de metástasis, incluida la inyección intravenosa de células tumorales, continuará avanzando en nuestro conocimiento de este importante desafío clínico. La metástasis, la principal causa de morbilidad y mortalidad para la mayoría de los pacientes con cáncer, puede ser difícil de modelar preclínicamente en ratones. Pocos modelos de metástasis espontáneas están disponibles. Por lo tanto, el modelo experimental de metástasis que implica la inyección de la vena de la cola de líneas celulares adecuadas es un pilar de la investigación de metástasis.
