大腸菌3、7、18を含む細胞間変動を定量化するために多くのアプローチが開発されている。これらの方法は、遺伝子の活性化と代謝経路を研究するためによく使用されるが、特にこれが基本的な課題である生細胞における遺伝的回路に対して、特定のノイズ源の測定や分断のような確率的ノイズダイナミクスの研究にあまり焦点を当てずに使用される。いくつかの要因は、回路自体に継承され(組み込み)、宿主細胞(外因)から派生し、遺伝的回路の連続的な性能を乱す可能性があります。本論文では、固有および外因性ノイズ源6,22を含む大腸菌細胞の全雑音を定量化することを目的としたプロトコルを開発した。全ノイズを定量化し、SNR23を評価することで、遺伝子回路の設計を改善することができます。この方法は、いくつかの蛍光タンパク質6,20を監視することにより、独立したノイズ源を別々に測定するように改変することができる。ここで説明するプロトコルについては、環境条件を十分に制御し、外部要因の影響を受けることなく細胞の活性を継続的に測定します。一つの細胞内の蛍光タンパク質からのシグナルを時間の経過とともに測定し、同時にアガロース基板の下で画像化します。結果の画像は実験室のカスタムMATLABを使用して分析される。
理想的には、細胞内の蛍光タンパク質のリアルタイム活性を連続的に測定すると、細胞の増殖と分裂を通じて正確なデータが生成されます。しかし、そのようなデータを取得することは困難です。これは、励起プロセスで放射線にさらされたときの蛍光タンパク質(光漂白7)の分解によるものです。さらに、 大腸菌 細胞も興奮に敏感であり、光毒性7を引き起こしうる。どちらの問題でも、取得できるフォトフレームの量と取得までの時間が制限されます。イメージング中に細胞を増殖させるために使用される基質および培地タイプ(例えば、リソゲニーブロス)も重要な役割を有する。蛍光性を最小限に抑え、細胞分裂時間を延長する最小限の培地を使用することを強くお勧めします。
