Het belang van deze methode is dat het gebruik maakt van het gevestigde protocol van micro-injectie in Drosophila embryo's21,22,23,24,25,26,27 om nieuw onderzoek met betrekking tot de ASR en de bijbehorende actin staaf assemblage mogelijk te maken. Een groot voordeel van het injecte...

Dit protocol beschrijft hoe G-actineRood te injecteren om de assemblage van intranucleaire actine staven in hitte-gestresste embryo's die een induceerbare Actin Stress Response (ASR)ondergaan visualiseren 1. We ontwikkelden dit protocol ter ondersteuning van studies van de ASR, die in embryo's leidt tot verstoorde morfogenese en verminderde levensvatbaarheid, en bij volwassen menselijke celtypen wordt geassocieerd met pathologieën zoals nierfalen2, spier myopathieën3, en Alzheimer en De Ziekte van Huntington4,5,6,7,8. Deze ASR wordt veroorzaakt door talrijke cellulaire spanningen, waaronder hitteschok9,10,11,oxidatieve stress4,6, verminderde ATP-synthese12, en abnormale Huntingtine of β-amyloïde oligomerisatie4,5,6,7,9,13,14,15,16. Een kenmerk van de ASR is de assemblage van afwijkende actinestaven in het cytoplasma of de kern van aangetaste cellen, die wordt aangedreven door stress-geïnduceerde hyperactivatie van een actine interacterend eiwit, Cofilin1,5,6,10. Helaas blijven er belangrijke kennislacunes bestaan met betrekking tot de ASR. De functie van de actinestaven is bijvoorbeeld niet bekend. We begrijpen niet waarom staven zich vormen in het cytoplasma van sommige celtypen, maar de kern van anderen. Het is ook niet duidelijk of de ASR beschermend of onaantastelijk is voor cellen of embryo's die stress ervaren. Tot slot kennen we nog steeds niet de gedetailleerde mechanismen die ten grondslag liggen aan Cofilin hyperactivatie of actine staafassemblage. Dit protocol biedt dus een snelle en veelzijdige test om de ASR te onderzoeken door actine staafvorming en dynamiek in het zeer tracteerbare experimentele systeem van het levende fruitvliegembryo te visualiseren.
Het protocol om G-actineRed te micro-injecteren in levende Drosophila-embryo's werd aanvankelijk ontwikkeld om de dynamiek van normale cytoplasmatische actinestructuren17 te bestuderen tijdens weefselopbouwgebeurtenissen. In die studies ontdekten we dat G-actineRode injectie geen negatieve invloed had op vroege ontwikkelingsprocessen in het embryo, waaronder cytokinese of gastrulatie17,18. Vervolgens hebben we het protocol gewijzigd, waarbij we de embryobehandeling en G-actineRed-injectie hebben aangepast om beeldvorming van actinestaven in hittebelaste embryo's die de ASR1ondergaan mogelijk te maken. Andere methoden naast G-actineRode injectie kunnen worden gebruikt om actine in embryo's te visualiseren. Deze methoden zijn gebaseerd op het uitdrukken van fluorescerende eiwitten (FPs) die zijn getagd op actine of op domeinen van actinebindende eiwitten, zoals Utrophin-mCherry, Lifeact, F-tractin-GFP en Moesin-GFP (herzien in19). Het gebruik van deze FP-sondes vereist echter voorzichtigheid, omdat ze sommige actinestructuren kunnen stabiliseren of verstoren, niet alle actinestructuren gelijk labelen20, en in het geval van actin-GFP, zeer overexpressie hebben – problematisch voor de analyse van staafassemblage die niet alleen stressafhankelijk is, maar ook actineconcentratieafhankelijk1. Zo is G-actinRed de voorkeursonde voor staafstudies in vliegenembryo's, en de grote omvang van het embryo maakt de eenvoudige injectie mogelijk.
De workflow van dit protocol is vergelijkbaar met andere gevestigde micro-injectietechnieken die zijn gebruikt voor het injecteren van eiwitten, nucleïnezuren, geneesmiddelen en fluorescerende indicatoren in Drosophila-embryo 's21,22,23,24,25,26,27. Echter, na de micro-injectie van G-actineRood hier, embryo's worden blootgesteld aan milde hitte stress om de ASR en intranucleaire actine staaf assemblage te induceren. Voor laboratoria met toegang tot vliegen en een injectieplatform moet deze methode gemakkelijk uitvoerbaar en aanpasbaar zijn voor specifieke studielijnen met betrekking tot de ASR, met inbegrip van de inductie ervan door verschillende spanningen of modulatie in verschillende genetische achtergronden.

imaging intranuclear actin rods in live heat stressed drosophila embryos

Het doel van dit protocol is om intranucleaire actinestaven te visualiseren die zich verzamelen in levende Drosophila melanogaster embryo's na hittestress. Actin staven zijn een kenmerk van een geconserveerde, induceerbare Actin Stress Response (ASR) die gepaard gaat met menselijke pathologieën, waaronder neurodegeneratieve ziekte. Eerder toonden we aan dat de ASR bijdraagt aan morfogenesefalen en verminderde levensvatbaarheid van het ontwikkelen van embryo's. Dit protocol maakt de voortdurende studie mogelijk van mechanismen die ten grondslag liggen aan actine staafassemblage en de ASR in een modelsysteem dat zeer vatbaar is voor beeldvorming, genetica en biochemie. Embryo's worden verzameld en op een afdeklip gemonteerd om ze voor te bereiden op injectie. Rhodamine-geconjugeerde bolvormige actine (G-actineRed) wordt verdund en in een microneedle geladen. Een enkele injectie wordt gemaakt in het midden van elk embryo. Na injectie worden embryo's geïncubeerd bij verhoogde temperatuur en intranucleaire actinestaven worden vervolgens gevisualiseerd door confocale microscopie. Fluorescentieherstel na fotobleaching (FRAP) experimenten kunnen worden uitgevoerd op de actinestaven; en andere actinerijke structuren in het cytoplasma kunnen ook in beeld worden We vinden dat G-actineRed polymeriseert als endogene G-actine en op zichzelf de normale embryoontwikkeling niet verstoort. Een beperking van dit protocol is dat er tijdens de injectie op moet worden gelet om ernstig letsel aan het embryo te voorkomen. Echter, met de praktijk, het injecteren van G-actineRood in Drosophila embryo's is een snelle en betrouwbare manier om actine staven te visualiseren en kan gemakkelijk worden gebruikt met vliegen van elk genotype of met de introductie van andere cellulaire spanningen, waaronder hypoxie en oxidatieve stress.

Een schematische workflow van embryoverwerking wordt weergegeven in figuur 1en een tijdschema voor een typisch experiment wordt gepresenteerd in tabel 1. Een schatting voor een goed experimenteel resultaat is dat voor elke 10 geïnjecteerde embryo's ten minste de helft van de bekeken embryo's zich in de juiste ontwikkelingsfase bevindt, onbeschadigd, en een robuuste ASR vertoont met hittestress bij 32 °C.
