Ce protocole décrit comment injecter G-actinRed pour visualiser l’assemblage de tiges d’actine intranucléaires dans les embryons stressés par la chaleur qui subissent une réponse inductible au stress actin (ASR)1. Nous avons développé ce protocole pour faciliter les études de l’ASR, qui dans les embryons conduit à une morphogenèse perturbée et une viabilité réduite, et dans les types de cellules humaines adultes est associée à des pathologies, y compris l’insuffisancerénale 2, myopathies musculaires3, et la maladie d’Alzheimer et de Huntington4,5,6,7,8. Cet ASR est induit par de nombreuses contraintes cellulaires, y compris le chocthermique 9,10,11, stress oxydatif4,6, réduction de la synthèse ATP12, et anormal Huntingtin ou oligomérisation β-amyloïde4,5,6,7,9,13,14,15,16. Une caractéristique de l’ASR est l’assemblage de tiges d’actine aberrantes dans le cytoplasme ou le noyau des cellules affectées, qui est entraînée par l’hyperactivation induite par le stress d’une protéine actin interagissant, Cofilin1,5,6,10. Malheureusement, des lacunes importantes subsistent en matière de connaissances concernant le RSE. Par exemple, la fonction des tiges d’actine n’est pas connue. Nous ne comprenons pas pourquoi les tiges se forment dans le cytoplasme de certains types de cellules, mais le noyau d’autres. Il n’est pas clair non plus si l’ASR est protecteur ou inadapté pour les cellules ou les embryons soumis à un stress. Enfin, nous ne connaissons toujours pas les mécanismes détaillés sous-jacents à l’hyperactivation cofiline ou à l’assemblage de tiges d’actine. Ainsi, ce protocole fournit un essai rapide et polyvalent pour sonder l’ASR en visualisant la formation et la dynamique de tige d’actine dans le système expérimental hautement tractable de l’embryon vivant de mouche de fruit.
Le protocole de microinject G-actinRouge dans les embryons vivants de Drosophila a été initialement développé pour étudier la dynamique des structures normales d’actine cytoplasmique17 pendant des événements de construction de tissu. Dans ces études, nous avons constaté que l’injection rouge de G-actin n’a pas affecté défavorablement des processus développementaux tôt dans l’embryon, y compris la cytokinésie ou la gastrulation17,18. Nous avons ensuite modifié le protocole, en adaptant la manipulation de l’embryon etl’injection rouge de G-actine pour permettre l’imagerie des tiges d’actine dans les embryons stressés par la chaleur subissant l’ASR1. D’autres méthodes que l’injection de G-actin Red peuvent être utilisées pour visualiser l’actine chez les embryons. Ces méthodes reposent sur l’expression de protéines fluorescentes (FPs) étiquetées à l’actine ou à des domaines de protéines liant l’actine, telles que utrophine-mCherry, Lifeact, F-tractin-GFP, et Moesin-GFP (examinédans 19). Toutefois, l’utilisation de ces sondes FP exige de la prudence parce qu’elles peuvent stabiliser ou perturber certaines structures d’actine, ne pas étiqueter également toutes les structures d’actine20, et dans le cas de l’actin-GFP, sont fortement surexprimées – problématiques pour l’analyse de l’assemblage de tiges qui dépend non seulement du stress, mais aussi de la concentration d’actinedépendante 1. Ainsi, G-actinRed est la sonde préférée pour les études de tige dans les embryons de mouche, et la grande taille de l’embryon permet son injection facile.
Le flux de travail de ce protocole est similaire à d’autres techniques bien établies de microinjection qui ont été utilisées pour injecter des protéines, des acides nucléiques, des médicaments et des indicateurs fluorescents dans les embryons de Drosophile 21,22,23,24,25,26,27. Cependant, à la suite de la microinjection de G-actinRouge ici, les embryons sont exposés à un léger stress thermique pour induire l’ASR et l’assemblage intranucléaire de tige d’actine. Pour les laboratoires ayant accès aux mouches et une plate-forme d’injection, cette méthode devrait être facilement implémentable et adaptable pour des lignes d’étude spécifiques en ce qui concerne l’ASR, y compris son induction par différents stress ou modulation dans des milieux génétiques distincts.

imaging intranuclear actin rods in live heat stressed drosophila embryos

Le but de ce protocole est de visualiser les tiges intranucléaires d’actine qui se réunissent dans les embryons vivants de Drosophila melanogaster suivant le stress thermique. Les tiges d’actine sont la marque d’une réponse de stress actin (ASR) conservée et inductible qui accompagne les pathologies humaines, y compris les maladies neurodégénératives. Précédemment, nous avons montré que l’ASR contribue aux échecs de morphogenèse et à la viabilité réduite des embryons en développement. Ce protocole permet d’étudier en continu les mécanismes sous-jacents à l’assemblage des tiges d’actine et à l’ASR dans un système modèle très sensible à l’imagerie, à la génétique et à la biochimie. Les embryons sont recueillis et montés sur un coverslip pour les préparer à l’injection. L’actine globulaire conjuguée à la rhodamine (G-actinRed)est diluée et chargée dans un microneedle. Une seule injection est effectuée au centre de chaque embryon. Après injection, les embryons sont incubés à haute température et les tiges d’actine intranucléaire sont ensuite visualisées par microscopie confoccale. La récupération de la fluorescence après des expériences de photobleaching (FRAP) peut être effectuée sur les tiges d’actine; et d’autres structures riches en actine dans le cytoplasme peuvent également être photographiées. Nous constatons que G-actinRed polymérise comme la G-actine endogène et n’interfère pas, à lui seul, avec le développement normal de l’embryon. Une limitation de ce protocole est que le soin doit être pris pendant l’injection pour éviter des dommages graves à l’embryon. Cependant, avec la pratique, l’injection de G-actinRouge dans les embryons de Drosophila est un moyen rapide et fiable de visualiser les tiges d’actine et peut facilement être utilisé avec des mouches de n’importe quel génotype ou avec l’introduction d’autres contraintes cellulaires, y compris l’hypoxie et le stress oxydatif.

Un flux de travail schématique de la manipulation des embryons est représenté à la figure 1,et un calendrier pour une expérience typique est présenté dans le tableau 1. Une estimation pour un bon résultat expérimental est que pour chaque 10 embryons injectés, au moins la moitié des embryons vus seront au stade de développement correct, intact, et présentent un ASR robuste avec le stress thermique à 32 °C.
