Questo protocollo descrive come iniettare G-actinRed per visualizzare l'assemblaggio di barre di actina intranucleare in embrioni stressati dal calore che stanno subendo un'induttibile risposta allo stress actina (ASR)1. Abbiamo sviluppato questo protocollo per aiutare gli studi dell'ASR, che negli embrioni porta a morfogenesi interrotta e ridotta vitalità, e nei tipi di cellule umane adulte è associato a patologie tra cui insufficienza renale2,miopatiemuscolari 3e morbo di Alzheimer e Huntington4,5,6,7,8. Questa ASR è indotta da numerose sollecitazioni cellulari, tra cui shocktermico 9,10,11,stress ossidativo4,6,ridotta sintesi ATP12e huntingtina anomala o oligomerizzazione β-amiloide4,5,6,7,9,13,14,15,16. Un segno distintivo dell'ASR è l'assemblaggio di aste di actina aberrante nel citoplasma o nel nucleo delle cellule colpite, che è guidato dall'iperattivazione indotta dallo stress di una proteina che interagisce con l'actina, Cofilin1,5,6,10. Sfortunatamente, permangono lacune chiave in materia di conoscenze per quanto riguarda l'ASR. Ad esempio, la funzione delle aste actin non è nota. Non capiamo perché le aste si formano nel citoplasma di alcuni tipi di cellule, ma il nucleo di altri. Né è chiaro se l'ASR sia protettivo o disadattiva per cellule o embrioni sottoposti a stress. Il protocollo per microiniettare G-actinRed in embrioni viventi di Drosophila è stato inizialmente sviluppato per studiare la dinamica delle normali strutture dell'actina citoplasmatica17 durante gli eventi di costruzione dei tessuti. In questi studi, abbiamo scoperto che l'iniezione di G-actinaRed non ha influito negativamente sui primi processi di sviluppo nell'embrione, tra cui citocinesi o gastrulazione17,18. Abbiamo quindi modificato il protocollo, adattando la manipolazione dell'embrione e l'iniezione G-actinRed per consentire l'imaging di aste di actina in embrioni stressati dal calore sottoposti all'ASR1. Altri metodi oltre all'iniezione di G-actinRed possono essere utilizzati per visualizzare l'actina negli embrioni. Questi metodi si basano sull'esprimere proteine fluorescenti (FP) taggate ad actina o a domini di proteine che legano l'actina, come Utrophin-mCherry, Lifeact, F-tractin-GFP e Moesin-GFP (recensito in19). Tuttavia, l'uso di queste sonde FP richiede cautela perché possono stabilizzare o interrompere alcune strutture actina, non etichettano allo stesso modo tutte le strutture actin20e, nel caso dell'actina-GFP, sono altamente sovraespresse – problematiche per l'analisi dell'assemblaggio dell'asta che non dipende solo dallo stress, ma agisce anche nella concentrazionedipendente 1. Pertanto, G-actinRed è la sonda preferita per gli studi sulle aste negli embrioni di mosca e le grandi dimensioni dell'embrione ne consentono la facile iniezione.
Il flusso di lavoro di questo protocollo è simile ad altre consolidate tecniche di microiniezione che sono state utilizzate per iniettare proteine, acidi nucleici, farmaci e indicatori fluorescenti negli embrioni di Drosophila 21,22,23,24,25,26,27. Tuttavia, in seguito alla microiniezione di G-actinRed qui, gli embrioni sono esposti a un leggero stress termico per indurre l'ASR e l'assemblaggio intranucleare dell'asta di actina. imaging intranuclear actin rods in live heat stressed drosophila embryos

Lo scopo di questo protocollo è quello di visualizzare le barre di actina intranucleare che si assemblano in embrioni di melanogaster Drosophila vivi a seguito dello stress termico. Le canne Actin sono un segno distintivo di una risposta allo stress actina conservata e induttibile (ASR) che accompagna patologie umane, tra cui la malattia neurodegenerativa. In precedenza, abbiamo dimostrato che l'ASR contribuisce ai fallimenti della morfogenesi e alla ridotta vitalità degli embrioni in via di sviluppo. Questo protocollo consente di continuare lo studio dei meccanismi alla base dell'assemblaggio dell'asta di actina e dell'ASR in un sistema modello altamente suscettibile di imaging, genetica e biochimica. Gli embrioni vengono raccolti e montati su una coverlip per prepararli all'iniezione. L'actina globulare coniugata con rodamina (G-actinaRossa)viene diluita e caricata in un microneedle. Una singola iniezione viene effettuata al centro di ogni embrione. Dopo l'iniezione, gli embrioni vengono incubati a temperatura elevata e le barre di actina intranucleare vengono quindi visualizzate mediante microscopia confocale. Il recupero della fluorescenza dopo esperimenti di fotobleaching (FRAP) può essere eseguito sulle barre di actina; e altre strutture ricche di actina nel citoplasma possono anche essere immagini. Scopriamo che G-actinRed polimerizza come l'endogeno G-actina e non interferisce, da solo, con il normale sviluppo embrionale. Una limitazione di questo protocollo è che occorre fare attenzione durante l'iniezione per evitare gravi lesioni all'embrione. Tuttavia, con la pratica, iniettare G-actinaRossa negli embrioni di Drosophila è un modo rapido e affidabile per visualizzare le aste di actina e può essere facilmente utilizzato con mosche di qualsiasi genotipo o con l'introduzione di altri stress cellulari, tra cui ipossia e stress ossidativo.

Un flusso di lavoro schematico di gestione degli embrioni è illustrato nella figura 1e un calendario per un esperimento tipico è presentato nella tabella 1. Una stima per un buon risultato sperimentale è che per ogni 10 embrioni iniettati, almeno la metà degli embrioni visualizzati sarà nella fase di sviluppo corretta, integra, e mostrerà una ROBUSTA ASR con stress termico a 32 °C.
