このプロトコルの目的は、熱ストレスに続く生きたショウジョウバエメラノガスター胚に集まり、核内アクチン棒を可視化することです。アクチンロッドは、神経変性疾患を含むヒト病理に付随する保存された誘導アクチンストレス応答(ASR)の特徴である。以前は、ASRが形態形成の失敗と胚の発達の生存率の低下に寄与することを示した。このプロトコルは、イメージング、遺伝学および生化学に非常に適したモデルシステムにおけるアクチンロッドアセンブリおよびASRの基礎となるメカニズムの継続的な研究を可能にする。胚は回収され、注入のためにそれらを準備するためにカバースリップに取り付けられる。ローダミン共役球状アクチン(G-アクチンレッド)を希釈し、マイクロニードルに装填する。各胚の中心に1回の注射が行われる。注射後、胚は高温でインキュベートされ、核内アクチン棒は共焦点顕微鏡で視覚化される。光漂白(FRAP)実験後の蛍光回復はアクチン棒に対して行われ得る。細胞質中のアクチンが豊富なその他の構造も画像化することができる。我々は、G-アクチンレッドが内因性G-アクチンのように重合し、単独では正常な胚の発達を妨げないことを発見した。このプロトコルの1つの制限は、胚への重大な傷害を避けるために注射中に注意を払わなければならないということです。しかし、練習では、ショウジョウバエ胚にG-actinRedを注入することは、アクチンロッドを視覚化するための迅速かつ信頼性の高い方法であり、任意の遺伝子型のハエや低酸素症や酸化ストレスを含む他の細胞ストレスの導入で簡単に使用することができます。
