Denne protokollen beskriver hvordan du injiserer G-actinRed for å visualisere montering av intranukleære aktinstenger i varmestressede embryoer som gjennomgår en induserbar Actin Stress Response (ASR)1. Vi utviklet denne protokollen for å hjelpe studier av ASR, som i embryoer fører til forstyrret morfogenese og redusert levedyktighet, og hos voksne menneskelige celletyper er forbundet med patologier, inkludert nyresvikt2,muskel myopatier3, og Alzheimers og Huntingtons sykdom4,5,6,7,8. Denne ASR er indusert av mange cellulære påkjenninger, inkludertvarmesjokk 9,10,11, oksidativt stress4,6, redusert ATP syntese12, og unormal Huntingtin eller β-amyloid oligomerization4,5,6,7,9,13,14,15,16. Et kjennetegn på ASR er montering av avvikende aktinstenger i enten cytoplasma eller kjernen av berørte celler, som drives av stressindusert hyperaktivering av et aktininteraktiverende protein, Cofilin1,5,6,10. Det er heller ikke klart om ASR er beskyttende eller maladaptiv for celler eller embryoer som gjennomgår stress. Dermed gir denne protokollen en rask og allsidig analyse for å sondere ASR ved å visualisere actin stangdannelse og dynamikk i det svært tractable eksperimentelle systemet til det levende fruktfluembryoet.
Protokollen til mikroinject G-actinRed til levende Drosophila embryoer ble opprinnelig utviklet for å studere dynamikken i normale cytoplasmatiske aktin strukturer17 under vev bygge hendelser. I disse studiene fant vi at G-actinRed injeksjon ikke påvirket tidlige utviklingsprosesser i embryoet negativt, inkludert cytokinese eller gastrulation17,18. Vi endret deretter protokollen, tilpasser embryohåndtering og G-actinRød injeksjon for å tillate avbildning av actin stenger i varmestressede embryoer som gjennomgår ASR1. Arbeidsflyten av denne protokollen ligner på andre veletablerte mikroinjeksjon teknikker som har blitt brukt til å injisere proteiner, nukleinsyrer, narkotika, og fluorescerende indikatorer i Drosophila embryoer21,22,23,24,25,26,27. Men etter mikroinjeksjon av G-actinRed her, embryoer er utsatt for mild varmestress for å indusere ASR og intranukleær actin stang montering. For laboratorier med tilgang til fluer og en injeksjonsrigg, bør denne metoden være lett gjennomførbar og tilpasningsdyktig for spesifikke studielinjer i forhold til ASR, inkludert induksjon av ulike påkjenninger eller modulering i distinkt genetisk bakgrunn.

imaging intranuclear actin rods in live heat stressed drosophila embryos

Formålet med denne protokollen er å visualisere intranukleære aktinstenger som monteres i levende Drosophila melanogaster embryoer etter varmestress. Actin stenger er et kjennetegn på en bevart, induserbar Actin Stress Response (ASR) som følger med menneskelige patologier, inkludert nevrodegenerativ sykdom. Tidligere viste vi at ASR bidrar til morfogenesefeil og redusert levedyktighet for å utvikle embryoer. Denne protokollen tillater fortsatt studie av mekanismer underliggende actin stang montering og ASR i et modellsystem som er svært mottagelig for avbildning, genetikk og biokjemi. Embryoer samles og monteres på en dekkslip for å forberede dem til injeksjon. Rhodamin-konjugert kuleformede actin (G-actinRed) fortynnes og lastes inn i et mikroneedle. En enkelt injeksjon er gjort i midten av hvert embryo. Etter injeksjon inkuberes embryoer ved forhøyet temperatur og intranukleære aktinstenger visualiseres deretter ved konfokal mikroskopi. Fluorescens utvinning etter fotobleaching (FRAP) eksperimenter kan utføres på actin stenger; og andre aktinrike strukturer i cytoplasma kan også avbildes. Vi finner at G-actinRed polymeriserer som endogene G-actin og ikke alene forstyrrer normal embryoutvikling. En begrensning av denne protokollen er at forsiktighet må tas under injeksjon for å unngå alvorlig skade på embryoet. Men med praksis er injeksjon av G-actinRed i Drosophila embryoer en rask og pålitelig måte å visualisere actin stenger og kan enkelt brukes med fluer av hvilken som helst genotype eller med innføring av andre cellulære påkjenninger, inkludert hypoksi og oksidativt stress.

En skjematisk arbeidsflyt for embryohåndtering er avbildet i figur 1, og en tidsplan for et typisk eksperiment presenteres i tabell 1. Et anslag for et godt eksperimentelt utfall er at for hver 10 embryoer injisert, vil minst halvparten av embryoene som vises være på riktig utviklingsstadium, uskadet, og utvise en robust ASR med varmestress ved 32 °C.
