Este protocolo descreve como injetar G-actinRed para visualizar a montagem de hastes de actina intranuclear em embriões estressados pelo calor que estão passando por uma Resposta de Estresse Actina indutível (ASR)1. Desenvolvemos este protocolo para auxiliar estudos do ASR, que em embriões leva à morfogênese interrompida e à viabilidade reduzida, e em tipos de células humanas adultas está associado a patologias incluindo insuficiência renal2,miopatias musculares3,e Alzheimer e Doença de Huntington4,5,6,7,8. Este ASR é induzido por numerosos estresses celulares, incluindo choque térmico9,10,11, estresse oxidativo4,6, síntese ATP reduzida12, e huntingtin anormal ou β-amilóide oligomerização4,5,6,7,9,13,14,15,16. Uma marca registrada do ASR é a montagem de hastes de actina aberrantes no citoplasma ou núcleo das células afetadas, que é impulsionada pela hiperativação induzida pelo estresse de uma proteína interativa de actina, Cofilin1,5,6,10. Infelizmente, permanecem as principais lacunas de conhecimento em relação ao ASR. Por exemplo, a função das hastes de actin não é conhecida. Não entendemos por que as hastes se formam no citoplasma de alguns tipos de células, mas o núcleo de outros. Também não está claro se o ASR é protetor ou mal adaptável para células ou embriões submetidos a estresse. Finalmente, ainda não sabemos os mecanismos detalhados subjacentes à hiperativação de Cofilin ou ao conjunto de hastes actin. Assim, este protocolo fornece um ensaio rápido e versátil para sondar o ASR visualizando a formação e dinâmica da haste actin no sistema experimental altamente tratável do embrião de mosca de fruta viva.
O protocolo para microinjetar G-actinRed em embriões Drosophila vivos foi inicialmente desenvolvido para estudar a dinâmica das estruturas normais de actina citoplasmática17 durante eventos de construção de tecidos. Nesses estudos, descobrimos que a injeçãovermelha G-actin não afetou negativamente os processos de desenvolvimento precoce no embrião, incluindo citocina ou gastraculação17,18. Em seguida, modificamos o protocolo, adaptando o manuseio de embriões e a injeçãovermelha G-actin para permitir a imagem de hastes de actina em embriões estressados pelo calor submetidos ao ASR1. Outros métodos além da injeção de G-actinRed podem ser usados para visualizar actina em embriões. Esses métodos dependem da expressão de proteínas fluorescentes (FPs) marcadas para atuar ou para domínios de proteínas de ligação de actina, como Utrophin-mCherry, Lifeact, F-tractin-GFP e Moesin-GFP (revisada em19). No entanto, o uso dessas sondas FP requer cautela, pois podem estabilizar ou interromper algumas estruturas de actina, não rotular igualmente todas as estruturas actin20, e no caso de actin-GFP, são altamente superexpressas – problemáticas para a análise da montagem de haste que não só é dependente do estresse, mas também atua na concentração dependente1. Assim, g-actinred é a sonda preferida para estudos de vara em embriões de mosca, e o grande tamanho do embrião permite sua fácil injeção.
O fluxo de trabalho deste protocolo é semelhante a outras técnicas de microinjeção bem estabelecidas que têm sido utilizadas para injetar proteínas, ácidos nucleicos, drogas e indicadores fluorescentes nos embriões de Drosophila 21,22,23,24,25,26,27. No entanto, após a microinjeção do G-actinRed aqui, os embriões são expostos a um leve estresse térmico para induzir o conjunto de hastes de actina ASR e intranuclear. Para laboratórios com acesso a moscas e uma plataforma de injeção, este método deve ser facilmente implementável e adaptável para linhas específicas de estudo em relação ao ASR, incluindo sua indução por diferentes estresses ou modulação em diferentes origens genéticas.

imaging intranuclear actin rods in live heat stressed drosophila embryos

O objetivo deste protocolo é visualizar hastes de actina intranucleares que se reúnem em embriões de melanogaster Drosophila vivos após estresse térmico. As hastes actina são uma marca registrada de uma Resposta ao Estresse Actin (ASR) conservada e indutível que acompanha patologias humanas, incluindo doenças neurodegenerativas. Anteriormente, mostramos que o ASR contribui para falhas de morfogênese e viabilidade reduzida do desenvolvimento de embriões. Este protocolo permite o estudo contínuo dos mecanismos subjacentes à montagem da haste actin e do ASR em um sistema modelo altamente favorável à imagem, genética e bioquímica. Os embriões são coletados e montados em uma tampa para prepará-los para injeção. A actina globular conjugada por rhodamina(G-actin Red) é diluída e carregada em uma microacela. Uma única injeção é feita no centro de cada embrião. Após a injeção, os embriões são incubados a temperatura elevada e as hastes de actina intranuclear são então visualizadas por microscopia confocal. A recuperação da fluorescência após experimentos de fotobleaching (FRAP) pode ser realizada nas hastes de actin; e outras estruturas ricas em actina no citoplasma também podem ser imagens. Descobrimos que o G-actinRed polimeriza como g-actina endógeno e não interfere, por si só, no desenvolvimento normal de embriões. Uma limitação deste protocolo é que deve-se tomar cuidado durante a injeção para evitar ferimentos graves no embrião. No entanto, com a prática, injetar G-actinRed em embriões de Drosophila é uma maneira rápida e confiável de visualizar hastes de actina e pode ser facilmente usada com moscas de qualquer genótipo ou com a introdução de outros estresses celulares, incluindo hipóxia e estresse oxidativo.

Um fluxo de trabalho esquemático de manuseio de embriões é retratado na Figura 1, e um cronograma para um experimento típico é apresentado na Tabela 1. Uma estimativa para um bom resultado experimental é que para cada 10 embriões injetados, pelo menos metade dos embriões vistos estará no estágio correto de desenvolvimento, não danificado, e exibirá um ASR robusto com estresse térmico a 32 °C.
