Этот протокол описывает, как вводить G-actinRed, чтобы визуализировать сборку внутриядерных актин-стержней в термечных эмбрионах, которые проходят неопругиваемый actin Stress Response (ASR)1. Протокол микроинъекций G-actinRed в живые эмбрионы дрозофилы был первоначально разработан для изучения динамики нормальных цитоплазмических актиновыхструктур 17 во время построения тканей. imaging intranuclear actin rods in live heat stressed drosophila embryos

Целью данного протокола является визуализация внутриядерных актин-стержней, которые собираются в живых эмбрионах drosophila melanogaster после теплового стресса. Actin стержней являются отличительной чертой сохраняется, неудобоваемые Actin стресс ответ (ASR), который сопровождает человеческие патологии, в том числе нейродегенеративных заболеваний. Ранее мы показали, что ASR способствует морфогенеза неудачи и снижение жизнеспособности развивающихся эмбрионов. Этот протокол позволяет продолжить изучение механизмов, лежащих в основе сборки актин стержня и ASR в модельной системе, которая очень податлима для визуализации, генетики и биохимии.
