我々のプロトコルは、関節軟骨に対する整形外科インプラント用金属の摺動を試験し、それによって特定の軟骨細胞の生合成活性に対する焦点インプラントまたはヘミ関節形成の影響を調査することを可能にし、この技術の主な利点は、軟骨ペアリングにおける機械的負荷の三種学的特性および生物学的影響の両方に関する包括的な洞察を提供することである。この技術は、膝の骨軟骨欠損または股関節形成術を治療するために焦点金属インプラントの移植のような外科的処置後の臨床所見を補完するかもしれない。手順を実証すると、ドナウ大学の私の研究室のポスドクであるクリストファー・バウアーです。プレート構成、垂直荷重機能、調整可能な負荷とスライド速度にシリンダを備えた市販の往復トリボメーターを使用します。さらに、液体セルは、潤滑液中のテストを行うために必要とされる。下のサンプルホルダーの骨軟骨シリンダーを、スライド方向に合わせたマーキングで固定します。まず、圧力測定フィルムを用いて軟骨系のモリブデンコバルトの接触圧力を決定します。圧力測定フィルムをインターフェースに置き、30秒間静電気を加え、初期接触圧力、接触サイズ、形状を決定します。コバルトクロムモリブデンシリンダーを上部のロードセルに取り付けます。液体セルにテスト液を加え、骨軟骨シリンダーを水没させ、金属軟骨スライディングインターフェースを覆います。通常の力ストロークやスライド速度で説明されているようなテストパラメータを設定し、テスト全体を通して維持されます。潤滑液に浸した関節軟骨に対して金属シリンダーの逆滑りを開始します。実験全体を通して摩擦係数を監視し、実験を終了する。望ましい試験期間の後、サンプルホルダーから骨軟骨プラグを取り除き、PBSですすいで、さらに生物学的分析ができるまで培地で保管します。試験の間、制御サンプルと試験溶液を室温で保管し、機械的荷重にさらされたサンプルと一緒に分析します。スケール上の24ウェルプレートを置き、それをゼロにします。オステオコンドラルプラグをPBSで洗い、ペトリ皿に入れます。その後、メスを使用して移植片から軟骨を2つの等分で切断し、接触領域が両方の軟骨片に均等に分配され、約半分を約1ミリメートル立方体に分けます。遺伝子発現解析のために後半を使用して、24ウェルプレートで調製した1つのウェルに細かく軟骨を移し、組織重量を決定し、各サンプルに対してこのプロセスを繰り返し、プレートの各ウェルに1ミリリットルの成長培地を加えます。各ウェルに500マイクロリットルのXTT溶液を加え、混合し、インキュベーション後4時間、37°Cでプレートをインキュベートし、5ミリリットルチューブに移します。各ウェルの軟骨組織にDMSOの500マイクロリットルを加えることによってテトラゾリウム製品を抽出し、室温で1時間連続撹拌を施し、DMSO溶液を除去し、以前に採取したXTT溶液で引き出します。各サンプルの100マイクロリットルを3分の1に96ウェルプレートに移します。プレートリーダーを使用して、波長492ナノメートルの吸光度と、690ナノメートルの基準波長を測定します。骨軟骨プラグから得られた軟骨組織の後半を小片に分けたRNAミンチを分離し、セラミックビーズと300マイクロリットルのライセンス緩衝液を含むチューブに組織を移す。1%βメルカプトエタノールを用いて、市販のライセートを使用して、各実行後20分間の冷却段階で6、500RPMを20秒間4回均質化する。各サンプルに20マイクロリットルのプロテイナーゼKと580マイクロリットルのRNAフリー水を加え、摂氏55度で30分間インキュベートします。サンプルを10,000倍Gで3分間遠心し、上清を1.5ミリリットルのチューブに移します。各チューブに0.5量の90%エタノールを加え、混合し、サンプル700マイクロリットルを2ミリリットルの回収チューブのRNA結合カラムに移し、8,000倍のGで15秒間遠心分離します。フロースルーを破棄し、残りのライセートに対して遠心化ステップを繰り返します。350マイクロリットルのバッファーRWをカラムに追加します。8,000回Gで15秒間遠心分離し、フロースルーを廃棄します。10マイクロリットルのDNAストック溶液と70マイクロリットルのバッファーRDDを混合します。溶液をRNA精製膜に加え、室温で15分間インキュベートし、350マイクロリットルのバッファーRWをカラムに加えます。遠心分離機をGの8,000倍で15秒間、フロースルーで500マイクロリットルのバッファRPEをRNA精製カラムに加え、遠心分離機を8時に廃棄し、 000回G 15秒間Gは流れを捨て、RNA精製カラムにさらに500マイクロリットルのバッファRPEを追加し、それを8,000倍Gで2分間遠心分離し、新しい1.5ミリリットルの回収チューブにカラムを入れ、30マイクロリットルを追加します。RNA遊離水は、チューブを8,000倍Gで1分間遠心し、市販キットを使用してcDNAを合成したRNAを全領域を混合してほのめかし、RNAサンプルを添加し、テキスト原稿に記載されているように反応とサーマルサイクラーを行う。RTQ PCRマスターミックスの9マイクロリットルを追加し、3重に各サンプルを持つ96ウェルプレートの各ウェルにcDNAの1マイクロリットルを追加すると、シーリングオイルでPCRプレートを輝き、遠心分離機でPCRプレートを877倍Gで4°Cで10分間、RTQ PCRと精度とサーマルサイクラーを行います。原稿の指示に従って。金属軟骨界面で接触面積と接触圧力を試験する前に、圧力測定フィルムを使用して確認する必要があります。次に、得られたインプリントを定義された接触に対する参照推論と比較することにより生理学的荷重条件を確認することができる低摩擦係数は、移動接触面積を有する少なくとも1時間維持することができる。細胞外マトリックス組成および構造は、サフランおよびO染色で決定することができる。サフランとO染色の強度は、プロテオグリカン含有量に比例します。プロテオグリカンの含有量は関節面上で変化するが、ベースラインサンプルの組織セクション全体で均一でなければならない、機械的負荷によって反作用することができるグリコサミノグリカンの抽出を示す。ウシ関節軟骨細胞の代謝活性は収穫部位から独立しているが、機械的負荷と共に増加を示す。軟骨特異的遺伝子の遺伝子発現レベルは、生理学的負荷条件とともに増加した。一方、同化遺伝子は静止した接触領域でアップレギュレートされています。この手順に従って、走査型電子顕微鏡を用いた軟骨ウェブ製品の測定や関節表面解析などの追加分析を行い、さらに様々な三重体組み合わせにおける関節軟骨の潤滑を最適化するよう努めています。
