低流量のデジタル気化器は、一般的に、揮発性麻酔薬をキャリアガスの流れに直接投与するためにシリンジポンプを利用します。動物の福祉勧告に従って、動物は麻酔を必要とする処置の間に温められ、監視される。一般的な麻酔および生理学的モニタリング装置はガスタンク、麻酔気化器および立場、加温制御装置およびパッド、機械換気装置および脈拍の酸化計を含んでいる。また、データ収集や機器ソフトウェアの実行には、コンピュータが必要です。小さなスペースやフィールドワークを行う場合、限られたスペースですべての機器を構成することは困難な場合があります。
このプロトコルの目的は、圧縮酸素と室内空気の両方を使用した低流量デジタル気化器の使用に関するベストプラクティスを示すとともに、統合された機械式人工呼吸器、パルスオキシメータ、および高い範囲の麻酔と齧歯類に理想的な生理学的モニタリングスイートとして遠赤外線の温暖化を示すことです。

生後10週、雄、野生型C57Bl6jマウスの重さ25.41±0.8gがこの研究に使用された。マウスは麻酔を受け、鼻コーンで維持されるか、挿管され、心拍数と酸素飽和度を監視しながら1.5〜2.5%のイオブルランを備えた統合された機械人工呼吸器上で維持された。動物はマイクロアイソレーションケージでグループ収容され、ボトルで標準的なげっ歯類のチョウと水への自由なアクセスを提供しました。
心拍数とSpO2は、パルスオキシメトリーを介してメンテナンス中に監視した(図5、図6、図7、)。体温は、赤外線加熱パッドとヒートランプを介して36.5〜37.5°Cに維持された。換気された動物は統合された鼻のコーンと挿管の立場を通して挿管のプロシージャの間にイオブルランの連続的な配達を受けた。各マウスは、15分間、141 mL/minの室内空気(RA)または酸素(O2)を超えない低流量で、鼻コーン上で正常に換気または維持されました。動物の心拍数と血中酸素飽和度は安定したままで、すべての群の測定に有意な変化はほとんどなかった。SpO2は全群で82~99%の間にとどまり、体温は36.5~37.5°Cの間に維持された。 パルスオキシメーターの位置と体温の両方がSpO2の測定に影響を与えるのを観察しました。パルスオキシメータからの無効な読み取りを観察した場合、センサーの配置と加熱レベルを調整して、コア体温を安定に保ちます。
図 5 、図 6、図7のデータの重要性を判断するために、ボンフェローニ補正を伴う双方向の ANOVA が実行されました。0.05 未満のp値は有意であると考えられた。
