암은 마우스 모델이 큰 유용성을 갖는 분야 중 하나입니다. 암의 유전자 마우스 모델은 세포가 발암성 형질전환을 겪기 위해 종양유전자 또는 종양 억제 유전자의 발현 조절에 단독으로 또는 조합하여 의존합니다. 마우스에 원발성 또는 확립 된 종양 세포주를 주입하는 것도 수행됩니다. 인간 또는 생쥐를 포함한 다른 동물 종의 세포주 또는 조직의 도입은 생체 내에서 가장 널리 사용되는 암 모델로 남아 있습니다. 면역이 저하된 마우스에서 이종 종(이종 이식편)의 세포 및 조직을 사용하는 것이 가장 일반적으로 시행된다2. 그러나, 숙주와 수용자가 모두 동일한 종인 동종이식 종양 세포 또는 조직의 사용은 동종 시스템에서 동일한 숙주 마우스 균주와 결합될 때 온전한 면역계와의 상호작용을 허용한다3.
원발성 골 종양 또는 고형 종양으로 인한 골 전이는 고통스러운 골용해성, 조골용해성 병변, 또는 혼합성 골용해성/조골아세포 병변을 초래한다 3,4. 이 종양은 뼈 구조를 손상시켜 병적 골절의 위험을 증가시키고 환자에게 제한된 치료 옵션을 남깁니다. 원발성 뼈 종양은 먼 장기로 전이되며 일부 유형은 다른 골격 부위로 퍼질 수 있습니다. 유방암 환자에서 뼈는 첫 번째 전이의 가장 흔한 부위이며 전이성 질환의 가장 빈번한 첫 번째 부위입니다 5,6. 또한, 파종성 종양세포(disseminated tumor cell, DTCs)는 다른 장기의 전이를 진단하기 전에 골수에 존재하며, 전이의 진행을 예측한다7. 따라서 뼈에 존재하는 암세포는 궁극적으로 다른 장기 시스템으로 전이되는 세포의 원천이라고 믿어집니다. 고형 종양 전이가 주로 폐와 림프절에서 전이가 발생하는 많은 마우스 모델이 존재하며, 종양 유형 및 주사 기술에 따라 잠재적으로 다른 장기 시스템이 발생할 수 있다3. 그러나, 뼈 전이의 마우스 모델은 마우스가 원발성 종양 부담 또는 다른 장기로의 전이로부터 조기 제거 기준에 도달하기 전에 신뢰할 수 있고 재현성 있게 부위 특이적 골격 전이를 생성하고 뼈 전이를 진행하는 것이 부족하다. 우리는 생쥐의 근위 경골에 고형 종양 동종이식편을 외과적으로 이식하는 원발성 골종양 골육종의 모델을 보고했다8. 뼈 종양은 생쥐의 100 %에서 형성되었고 88 %는 폐 전이를 일으켰습니다. 이 전이 발생률은 사람(~20-50%)에서 임상적으로 일반적으로 보고되는 것보다 높지만 폐가 골육종의 가장 흔한 전이 부위이기 때문에 큰 관심을 끌고 있습니다 9,10,11. 이 모델은 원발성 골 종양을 모델링하는 데 유리하지만 유방, 폐, 전립선, 갑상선, 간, 신장 및 위장관 종양과 같은 다른 골성 고형 종양의 뼈 전이를 모델링하는 데에도 큰 유용성을 가지고 있습니다.
