1. 材料准备

</ol>4. 乌特卡的绝育和孵化

</ol>5. 维持侏罗万象仙女
<ol><li>在层压流罩中，无菌地将灭菌大鼠的颗粒转移到准备好的 BHI 烧瓶（从步骤 1.3.3 开始）与无菌钳子。作为不育检查，将烧瓶放在 30 °C 孵化器中的二次容器中 24 小时，如果出现生长，则不使用。</li>

</ol>6. 不育质量控制
<ol><li>从 BHI 烧瓶中取出一个仙女，通过限制片段长度多态性 （RFLP） 来牺牲对侏儒状态进行文化独立的质量控制检查。为此，将仙女倒入无菌离心管（类似于从步骤 5.1 的仙女转移），或将无菌木制施用器放入烧瓶中，等待仙女开始攀爬，然后将其转移到离心管。</li>
	<li>在离心管中的仙女中加入 0.5 mL 的 1 倍磷酸盐缓冲盐水 （PBS），然后用无菌微虫均质，直到所有大块碎裂。漩涡很好。</li>

虽然在没有阳性培养结果的情况下，该测试尚未确定污染，但这一步骤在关键实验期间已定期进行，以排除存在污染氧敏感或挑剔的微生物。与标准/非星体昆虫相比，侏罗纪蟑螂的生长速度较慢。
