Vi har presentert en modell for kontinuerlig video-EEG-overvåking hos nyfødte mus under hypoksiske-iskemiske anfall. Videoanalyse i forbindelse med EEG tillater karakterisering av anfallssemologi. Analyse av EEG muliggjør utvinning av kraftspektrogrammer og bakgrunnsamplitudeanalyse.

Hypoksisk iskemisk encefalopati (HIE) er en tilstand som påvirker 1,5 av 1000 nyfødte årlig og er den vanligste årsaken til nyfødte anfall1,2. Spedbarn som overlever er i faresonen for ulike nevrologiske funksjonshemninger som cerebral parese, intellektuell funksjonshemming og epilepsi3,4,5.
Dyremodeller spiller en kritisk rolle i å forstå og undersøke patofysiologien til hypoksi iskemi og neonatale anfall6,7. En modifisert Vannucci-modell brukes til å indusere hypoksi iskemi (HI) på postnatal dag 10 (p10)7,8. Musevalper i denne alderen oversetter nevrologisk grovt til full sikt menneskelig neonate9.
Kontinuerlig overvåking av videoelektroencefalografi (EEG) som brukes i forbindelse med denne skademodellen, gir mulighet for ytterligere forståelse og karakterisering av neonatale hypoksiske iskemiske anfall. Tidligere studier har brukt ulike metoder for å analysere neonatale anfall hos gnagere, inkludert videoopptak, begrensede EEG-opptak og telemetri EEG-opptak10,11,12,13,14,15,16. I det følgende manuskriptet diskuterer vi i dybden prosessen med å spille inn kontinuerlig video EEG i musevalper under hypoksi-iskemi. Denne teknikken for kontinuerlig video EEG-overvåking i neonatale musevalper kan brukes på en rekke sykdoms- og anfallsmodeller.

Hypoksi iskemi er den vanligste årsaken til nyfødte anfall. Dyremodeller er avgjørende for å forstå mekanismene og fysiologien som ligger til grunn for neonatale anfall og hypoksi iskemi. Dette manuskriptet beskriver en metode for kontinuerlig overvåking av videoelektroencefalogram (EEG) hos nyfødte mus for å oppdage anfall og analysere EEG-bakgrunn under hypoksi iskemi. Bruk av video og EEG i forbindelse med beskrivelse av anfallssemologi og bekreftelse av beslag. Denne metoden tillater også analyse av kraftspektrogrammer og EEG-bakgrunnsmønstertrender over den eksperimentelle tidsperioden. I denne hypoksi iskemimodellen tillater metoden EEG-opptak før skade for å oppnå en normativ basislinje og under skade og gjenoppretting. Total overvåkingstid er begrenset av manglende evne til å skille valper fra moren i mer enn fire timer. Selv om vi har brukt en modell av hypoksiske-iskemiske anfall i dette manuskriptet, kan denne metoden for neonatal video EEG-overvåking brukes på forskjellige sykdoms- og anfallsmodeller hos gnagere.
