1. C. クィケファシアトゥス コロニー飼育

</ol>6. マイクロインジェクションのセットアップ

	<li>一度に約20個の卵を注入し、停止して胚の回復手順を実行します。 注:注射中に、針が詰まったり壊れたりする可能性が高くなります。詰まりが発生した場合は、注射液が針を流れる不足によって判断される可能性があり、電子マイクロインジェクターのクリーン機能で針を掃除するか、針を再びベベリングしてみてください。どちらのステップもうまくいかない場合は、準備された卵が湿ったままであることを確認しながら、すぐに新しい針に変更します。</li>
</ol>8. 胚の回復と孵化
<ol><li>卵を少なくとも5分間邪魔しません。20分以内に、きれいなペイントブラシで軽くブラッシングしてハロカーボンオイルを慎重に取り除きます(図2H)。</li>
	<li>卵をペイントブラシでやさしく持ち上げ、二重蒸留水のカップに入れます(図2I)。水面に卵を保管するように注意してください。</li>
	<li>卵が孵化するために毎日7日間チェックしてください(図2J)。</li>

Culex蚊はいかだに卵のグループを産むので、卵を集めるのはかなり簡単です。卵は、ペイントブラシでいかだ全体をすくうことによって単に収集することができます。卵の分離はより複雑ですが、正常な注射には不可欠です。卵は、ペイントブラシや鉗子で卵の間に穏やかな下降圧力をかけることで、いかだから慎重に分離することができます。いくつかの練習では、複数のユーザーが確実に卵のいかだから単一卵を分離することができました。各卵を分離した後、卵は両面付き粘着テープ上で同じ向きに整列し、マイクロインジェクション中に卵を安定させる。卵は先細りの後端に注入され、ハロカーボンオイルの添加は、操作中に卵を湿らせた保ちます。
マイクロインジェクション中の胚の損傷を制限するには、注射針が適切な強度を持ち、適切な角度でベベブする必要があります。50°の角度で10秒間ベベにされたアルミノケイ酸針は最良の結果を得ましたが、ボロシリケート針とクォーツ針も機能しますが、耐久性が低く、より高価である可能性があります。さらに、適切な針のベベルはCas9およびsgRNA混合物のよりよい流れを促進し、胚へのより容易な浸透のためのより鋭いポイントを作成する。多くの場合、ベベルは詰まった針を交換するのに時間と労力を費やすのではなく、詰まった針を素早く固定する効率的な方法でもあります。
注射後、卵は少なくとも5分間邪魔されず、ハロカーボンオイルはきれいな絵筆で軽くブラッシングしてすぐに取り除かれます。ハロカーボン油の除去後、注入された卵は、通常、注射後3日以内に発生する孵化するために水に入れることができます。練習と卵の十分な数で、このプロトコルは 、C.キンケファシアトゥス で一貫した体細胞および生殖細胞系列突然変異を達成することができ、他の Culex 蚊に容易に適応可能であるべきであるほど汎用性があります。

以前に公開された実験では、この方法は、暗い目の色素沈着の発達に重要な遺伝子の体細胞性および生殖細胞系列突然変異を正常に生成するために使用された、白色(CPIJ005542)30(表1)。CRISPR/Cas9は、注入された個体のプパルステージ目における色素沈着の喪失をスクリーニングすることによって採点した(G0)。体細胞変異は一般に、一部の細胞が変異表現型を有するが、その一部が変異型であるモザイク表現型として存在していた。例えば、白色遺伝子の体細胞変異は、色素沈着を欠く表現型を有するオマチディアと、野生型色素性表現型30との混合物をもたらした。しかし、白い遺伝子の生殖系列変異があった場合、次の世代(G1)は完全な白目表現型を受け継いだ。生殖細胞系列突然変異率は、モザイクG0個体を交交し、完全に白い目の子孫に対して得点することによって決定した(G1)。これらの実験結果は、胚生存率が64%から82%、ならびに37%〜57%の体細胞性突然変異誘発率および>61%の生殖細胞系変異生成率をもたらした(表1)。同じ遺伝子内の複数の遺伝子を標的とするsgRNAを多重化することにより、体細胞および生殖細胞系列突然変異誘発率は86%まで上方に増加した(表1)。さらに、多くの世代のために、白い突然変異体の実行可能なホモ接合株は、実験室で正常に保持されています。
