Genetisch gemanipuleerde muismodellen worden vaak gebruikt voor het bestuderen van het mechanisme en de farmaceutische behandeling van menselijke ziekten13. Ctsk-Cre muizen zijn veel gebruikt voor functionele studies van osteoclasten6. De huidige studie beschreef de protocollen van de methoden om skeletfenotype te analyseren en de kritische genen te bestuderen die de osteoclastische activiteit in vivo controleren.

Skeletbot is het belangrijkste dragende orgaan van het menselijk lichaam en staat onder druk van zowel de interne als externe omgeving tijdens het lopen en sporten1. Gedurende iemands hele leven ondergaan botten voortdurend zelfvernieuwing, die wordt gecompenseerd door osteoblasten en osteoclasten. Het proces van osteoclasten die oude botten en osteoblasten die nieuw bot vormen, zuivert de homeostase en mechanische functie van het skeletsysteem2. Verstoring van het evenwicht kan botmetabole ziekten veroorzaken, zoals osteoporose. Osteoporose, die wordt veroorzaakt door overmatige osteoclastische activiteit, komt wereldwijd voor en veroorzaakt aanzienlijke economische verliezen voor de samenleving2,3,4. Volgens het beperkte aantal geneesmiddelen dat beschikbaar is voor osteoporosebehandeling en hun risico op bijwerkingen4, is het belangrijk om de details van osteoclastenvorming en -activiteit te onthullen.
Osteoclasten afgeleid van de monocyt/macrofaag hematopoietische afstamming hebben meerdere kernen (kunnen 2 tot 50 kernen hebben) en zijn groot (meestal groter dan 100 μm in diameter)2. Hoewel de exploratie van mechanismen en de screening van geneesmiddelen op osteoclastische aandoeningen sterk zijn verbeterd via in vitro osteoclastencultuur, maken de gecompliceerde organische reacties in vivo bewijs onmisbaar voor de gerichte therapie. Vanwege genetische en pathofysiologische overeenkomsten tussen muizen en mensen worden genetisch gemanipuleerde muismodellen vaak gebruikt voor het bestuderen van de mechanismen en de farmaceutische behandelingen van menselijke ziekten in vivo6. Het Cre-loxP-systeem is een veelgebruikte technologie voor het bewerken van muisgen en heeft onderzoekers in staat gesteld om genfuncties op een weefsel-/celspecifieke manier te onderzoeken5. Cathepsine K (CSTK) is een cysteïne protease uitgescheiden door osteoclasten die botcollageen kunnen afbreken8. Het is algemeen aanvaard dat CTSK selectief wordt uitgedrukt in volwassen osteoclasten; daarom worden Ctsk-Cre muizen beschouwd als een nuttig hulpmiddel voor functionele studies van osteoclasten en is gebruikt6.
De signaaltransducer en activator van transcriptie (STAT) familie is klassiek en zeer significant in immuniteit en kankerprogressie en ontwikkeling7,8. Van de zeven STATs is STAT3 naar verluidt het meest relevant voor bothomeostase9,10. Verschillende in vivo studies hebben gemeld dat specifieke inactivatie van STAT3 in osteoblasten de botvorming vermindert9,10. Niettemin is solide bewijs met betrekking tot de deelname van STAT3 aan osteoclastenvorming en botmetabolisme in vivo nog steeds beperkt. Onlangs hebben we in vivo bewijs geleverd met een osteoclastenspecifiek Stat3-deletiemuismodel (Stat3fl/fl; Ctsk-Cre, hierna Stat3Ctskgenoemd) dat STAT3 deelneemt aan osteoclastendifferentiatie en botmetabolisme11. In deze studie beschrijven we de methoden en protocollen die we gebruikten om de veranderingen in botmassa, bot histomorfologie en bot anabolisme en katabolisme van de Stat3Ctsk muizen te analyseren om de invloed van osteoclasten-specifieke STAT3 deletie op bothomeostase te bestuderen.

Transgene muismodellen zijn krachtig voor het begrijpen van de kritische genen die osteoclastendifferentiatie en -activiteit beheersen, en voor het bestuderen van mechanismen en farmaceutische behandelingen van osteoporose. Cathepsine K (Ctsk)-Cre muizen zijn veel gebruikt voor functionele studies van osteoclasten. De signaaltransducer en activator van transcriptie 3 (STAT3) is relevant bij bothomeostase, maar de rol ervan bij osteoclasten in vivo blijft slecht gedefinieerd. Om het in vivo bewijs te leveren dat STAT3 deelneemt aan osteoclastendifferentiatie en botmetabolisme, hebben we een osteoclastenspecifiek Stat3-deletiemuismodel gegenereerd (Stat3 fl/fl ; Ctsk-Cre) en analyseerde zijn skeletfenotype. Micro-CT-scanning en 3D-reconstructie impliceerden een verhoogde botmassa bij de voorwaardelijke knock-outmuizen. H&E-kleuring, calceïne en alizarine rode dubbele kleuring en tartraatresistente zuurfosfatase (TRAP) kleuring werden uitgevoerd om het botmetabolisme te detecteren. Kortom, dit protocol beschrijft enkele canonieke methoden en technieken om skeletfenotype te analyseren en de kritische genen te bestuderen die de osteoclastenactiviteit in vivo controleren.

Met behulp van dit protocol werden osteoclastenspecifieke Stat3-deletiemuizen gegenereerd om de invloed van STAT3-deletie op osteoclastendifferentiatie te bestuderen. Stat3Ctsk muizen en hun wildtype (WT) nestgenoten werden gefokt en gehouden na genotypering. Beenmergmafagen werden geïsoleerd en gekweekt tot osteoclasten, en STAT3-deletie bij Stat3Ctsk-muizen werd aangetoond (figuur 1).
