Skelettknochen ist das Haupttragende Organ des menschlichen Körpers und steht unter Druck sowohl der inneren als auch der äußeren Umgebung während des Gehens und der Übung1. Das ganze Leben über gehen die Knochen kontinuierlich durch Selbsterneuerung, die durch Osteoblasten und Osteoklasten ausgeglichen wird. Der Prozess der Osteoklasten, die alte Knochen und Osteoblasten, die neue Knochen bilden, beseitigt, behält die Homöostase und mechanische Funktion des Skelettsystems2. Störungen im Gleichgewicht können Knochenstoffwechselerkrankungen wie Osteoporose auslösen. Osteoporose, die durch übermäßige osteoklastische Aktivität verursacht wird, ist weltweit verbreitet und verursacht erhebliche wirtschaftliche Verluste für die Gesellschaft2,3,4. Nach der begrenzten Anzahl von Medikamenten für osteoporose Behandlung und ihr Risiko von Nebenwirkungen4, Ist es wichtig, die Details der Osteoklastbildung und Aktivität zu enthüllen.
Osteoklasten, die aus der hämatopoetischen Abstammung monozyt/makrophage abgeleitet sind, haben mehrere Kerne (können 2 bis 50 Kerne haben) und sind groß (in der Regel größer als 100 m im Durchmesser)2. Obwohl die Erforschung von Mechanismen und das Screening von Medikamenten auf osteoklastische Störungen durch in vitro Osteoklastkultur weitgehend verbessert wurden, machen die komplizierten organischen Reaktionen in vivo Beweise für die zielgerichtete Therapie unverzichtbar. Aufgrund genetischer und pathophysiologischer Ähnlichkeiten zwischen Mäusen und Menschen werden gentechnisch veränderte Mausmodelle häufig zur Untersuchung der Mechanismen und der pharmazeutischen Behandlung menschlicher Krankheiten in vivo6verwendet. Das Cre-loxP-System ist eine weit verbreitete Technologie zur Mausgenbearbeitung und hat es Forschern ermöglicht, Genfunktionen gewebe-/zellspezifisch zu untersuchen5. Cathepsin K (CSTK) ist eine Cystein-Protease, die von Osteoklasten abgesondert wird, die Knochenkollagenabbauenkönnen 8 . Es ist allgemein anerkannt, dass CTSK selektiv in reifen Osteoklasten exprimiert wird; Daher gelten Ctsk-Cre-Mäuse als nützliches Werkzeug für funktionelle Studien von Osteoklasten und wurden6verwendet.
Der Signalgeber und Aktivator der Transkriptionsfamilie (STAT) ist klassisch und sehr signifikant in der Immunität und Krebsprogression und -entwicklung7,8. Unter sieben STATs ist STAT3 als die relevanteste für die Knochenhomöostase9,10. Mehrere In-vivo-Studien haben berichtet, dass die spezifische Inaktivierung von STAT3 bei Osteoblasten die Knochenbildungverringert 9,10. Dennoch sind solide Beweise für die Beteiligung von STAT3 an der Osteoklastbildung und am Knochenstoffwechsel in vivo nach wie vor begrenzt. Kürzlich haben wir in vivo-Beweise mit einem osteoklastspezifischen Stat3-Löschmausmodell (Stat3fl/fl; Ctsk-Cre, im Folgenden Stat3Ctskgenannt,dass STAT3 an der Osteoklastdifferenzierung und dem Knochenstoffwechsel11beteiligt ist. In der vorliegenden Studie beschreiben wir die Methoden und Protokolle, mit denen wir die Veränderungen der Knochenmasse, der Knochenhistomorphologie und des Knochenanabolismus und katabolismus der Stat3Ctsk-Mäuse analysiert haben, um den Einfluss der osteoklastspezifischen STAT3-Deletion auf die Knochenhomöostase zu untersuchen.

Transgene Mausmodelle sind mächtig für das Verständnis der kritischen Gene, die osteoklastische Differenzierung und Aktivität steuern, und für das Studium von Mechanismen und pharmazeutischen Behandlungen von Osteoporose. Cathepsin K (Ctsk)-Cre Mäuse wurden weit verbreitet für funktionelle Studien von Osteoklasten verwendet. Der Signalgeber und Aktivator der Transkription 3 (STAT3) ist bei der Knochenhomöostase relevant, aber seine Rolle bei Osteoklasten in vivo bleibt schlecht definiert. Um den in vivo-Beweis dafür zu liefern, dass STAT3 an der Osteoklastdifferenzierung und dem Knochenstoffwechsel beteiligt ist, haben wir ein osteoklastspezifisches Stat3-Löschmausmodell (Stat3 fl/fl; Ctsk-Cre) und analysierte seinen Skelett-Phänotyp. Mikro-CT-Scanning und 3D-Rekonstruktion implizierten eine erhöhte Knochenmasse in den bedingten Knockout-Mäusen. H&E-Färbung, Calcein und Alizarin rote Doppelfärbung, und tartrate-resistente Säurephosphatase (TRAP) Färbung wurden durchgeführt, um Knochenstoffwechsel zu erkennen.
