immunofluorescence imaging of dna damage and repair foci in human colon cancer cells

本稿の目的は、ホウ素中性子捕捉療法(BNCT)で用いられる中性子ガンマの混合ビームによって誘導される放射線誘発DNA損傷応答を研究するための免疫蛍光顕微鏡を実施するためのステップバイステップのプロトコルを提供することにある。具体的には、DNA二本鎖切断(DNA-DSB)に特異的な抗体を用いて病巣として可視化できる、修復タンパク質活性化の検出に提案された方法論が適用される。DNA修復病巣は、中性子混合ビームを照射した後の大腸癌細胞(HCT-116)における免疫蛍光によって評価した。DNA-DSBは最も遺伝子毒性の病変であり、哺乳類細胞では、非相同のエンド結合経路(NHEJ)および相同組換え修復(HRR)の2つの主要経路によって修復される。γ-H2AXのような放射線生物学で一般的に使用されるマーカーについては、病巣の周波数は、免疫化学的染色、53BP1はDNA-DSB数に関連しており、DNA-DSBの誘導および修復を監視するための効率的で敏感なマーカーと考えられている。γ-H2AX病巣が修復タンパク質を引き付け、DSB付近の修復因子の濃度が高いことが確立されました。細胞レベルでのDNA損傷をモニタリングするために、NHEJ経路からのDNA-PKcs代表的な修復タンパク質病巣およびHRR経路からのRad52の存在に対する免疫蛍光分析が計画された。NHEJおよびHRR経路からの修復因子に特異的な抗体を用いた放射線誘発DNA損傷応答の検出および放射線誘発病巣(RIF)の観察のための信頼できる免疫蛍光染色プロトコルを開発し、導入しました。提案された方法論は、中性子混合ビーム放射の場合に高度に活性化される修復タンパク質の調査に使用することができ、それによって修復経路の優位性を示す。
