该技术的目的是轻松制备仅用于快速SEM分析的横向稻仁切片。该技术的发展是由于需要快速横截面方法，即在使用最少的设备进行可视化之前立即为SEM显微镜准备样品。
该技术涉及将去壳的米粒插入移液器尖端以完全固定。这在横截面白垩米仁表型时尤其重要，这些表型易碎，在压力下容易破碎3。白垩是大米中不受欢迎的质量，因为它会影响内核的外观，并导致内核在抛光和碾磨过程中容易破裂3。白垩度表现为可以用肉眼观察到的内核横截面中的不透明区域;在微观层面上，白垩的特征是小的，松散的淀粉颗粒。白垩的原因可以是遗传4，5或环境6，7。
谷物种子横截面传统上是使用化学固定方法制备的，并在样品嵌入石蜡或其他固体基质4，8，9，10之后切片。2010年，松岛法被引入，作为避免复杂和耗时的稻仁样品制备方法4。该方法涉及将去壳的米粒插入截断的移液器尖端。尖端由块修剪器固定，并使用手持剃须刀片收获薄的局部胚乳部分。2016年开发的另一项快速技术允许对各种干燥种子进行薄整切片，包括白垩品种10。这些方法推动了这里介绍的快速技术的发展。

野生型日本（图2A）和 ssg1 部分（图2B）在三种放大倍率下进行了检查：260x，920x和4200x。该技术允许制备足够质量的切片以观察整个胚乳细胞（图3A），复合淀粉颗粒（图3B）和单个亚颗粒（图3C）。去壳的果仁比抛光的核需要更长的时间来处理，因为在切片之前需要通过磨损去除干燥的壳。白垩核的加工时间也比抛光的半透明内核更长，因为在切片过程中必须注意不要破坏内核。适当制备的稻块应约0.9毫米厚（表1），胚乳（图1N）和完整的果皮和芦荟层（图1O）的破碎最小或没有破碎。切片时手术刀在移液器吸头上放置不当会导致"碎裂"切片（图1P）。类似地 ，ssg1 最佳横向截面的明场图像（图1Q）显示完整的胚乳，果层和aleurone层完好无损，可用于可视化（图1R）。如果唯一的目的是观察SG，则破碎的白垩核部分（图1S）可能仍可用于可视化，但胚乳细胞模式将不可见。损坏的部分可能难以处理以进行分析。在野生型日本人中观察到更多的胚乳细胞壁剪切，因为细胞比 ssg1 内核更紧密地包装并且更不易碎。在 ssg1 切片中没有观察到胚乳细胞的剪切，并且复合淀粉颗粒完好无损。
