ここで紹介する手法は、デスクトップ SEM の視覚化のための横断的な米の断面を準備するための、高速でシンプルで鋭いアプローチを表します。この断面化技術により、胚乳細胞の形状、大きさ、パターン、化合物顆粒、デンプン形態の迅速な観察が可能になります。胚精子のフェノタイピングや胚芽スクリーニングの目的のために、米カーネル4、23、24の全断面を得ることが重要である。メスの刃の圧力が粉々になったり粉々になったりするのを防ぐために、ピペットチップ内にカーネルを完全に挿入することが最も重要です。「望遠鏡」の集合体が適切に組み立て済みである限り、サンプルは、典型的な実験室の設定で既に手に入っている材料を使用して、15秒以内に可視化のために調製することができる(表2)。この技術は、楕円体の種子の断面に適用可能です 直径約4ミリメートルの最も広い点で.モデル草の種はブラキ表ディスタキオン(図S2A)も同様に切り離すことができますが、環状に囲まれたままにしないでください。小麦のような大きな種子は、容易に骨折し、断面化時に注意を必要とします(図S2B)。
ただし、ここで示す手法には、いくつかの制限があります。この技術を用いて得られたセクションは、光が通過するのに十分な薄さではなく、明視野(米カーネルセクション25の最大サンプル厚さ500 μm)や透過電子顕微鏡(TEM)(500 nm最大サンプル厚さ26)のような透過光ベースの顕微鏡アプローチに対して使用することを禁止する).また、断面「マトリックス」としてピペットチップを使用すると、この技術を使用して切り離すことができる種子のサイズも制限されます。米とは非常に異なる種に対してこの技術を適応させるためには、さらなるトラブルシューティングが必要であり、「マトリックス」のサイズは、購入可能なピペットチップのサイズによって制限されます。
この技術が提供するもう一つの明確な利点は、チョーク状の表現型の米のカーネルから作り出すことができるサンプルの質である。松島の研究でさえ、比較を目的として本研究で再現されたチョーク状表現型4の特定の方法を用いて断面を得ることは困難であると認めたことは注目に値する(図1S)。その場合、チョーク状の米サンプルを化学的に固定し、切り離し用の樹脂に埋め込む必要が生じるようになった。この新しい技術は、デスクトップSEMイメージングと組み合わせて、研究者が固定化サポートなしでより一貫性のある顕微鏡用の米カーネルの横断セクションを簡単に準備することを可能にする(表3)。
フェノミクスとメタボロミクスの新時代には、種子中のデンプンの機能と重要性をよりよく理解するために、変異原性線とトランスポゾンタグ付きライブラリを監視することが重要です。さらに、国際ライスジーンバンクは、130,000米のアクセスを27以上保持しています。ここで示したような迅速な種子のフェノタイピング技術は、栄養の質28のための分類とサンプリングを促進するだろう。最後に、この技術は、気候変動の影響を侵食することに照らして有用である可能性があります。穀物充填時の季節的な高温ストレスは、すでにチョークの主な原因として同定されていたが、最近の研究は、米収量7、29のチョーク性の増加に世界的な気温の上昇に関与している。このような迅速な胚乳型のフェノタイピングは、世界的な温度の上昇の影響の広範な農業イメージを提供するのに役立つかもしれない。

デンプン顆粒(SG)は、植物種によって異なる形態を呈し、特にポアセ科1,2の胚乳では特に。子宮内精子表現型は走査型電子顕微鏡分析を用いてSG表現型に基づく遺伝子型を分類するために使用することができる。SGは、走査型電子顕微鏡(SEM)を用いて、カーネルをスライスし、子宮内精子細胞壁2を掻き取ることによって可視化することができる。
この技術の目的は、高速SEM分析のためだけに、簡単に横米カーネルセクションを準備することです。この技術の開発は、最小限の装置を使用して可視化する直前にサンプルをSEM顕微鏡用に調製する迅速な断面アプローチの必要性によって動機づけられました。

穀類種子の断面は、パラフィンワックスまたは別の固体マトリックス4、8、9、10に埋め込まれたサンプルに続く化学的固定法および断面を使用して伝統的に調製されてきた。2010年には、複雑で時間のかかる米のカーネルサンプル調製4を避ける方法として、松島法が導入されました。この方法では、切り捨てられたピペットチップに殻を押した米カーネルを挿入する必要があります。先端はブロックトリマーによって静止し、薄い部分的な胚乳セクションは手持ち型のカミソリの刃を使用して収穫される。2016年に開発された別の迅速な技術は、チョーク品種10を含む多種多様な乾燥種子の薄い全体の切り分けを可能にしました。これらの方法は、ここで提示された迅速な技術の開発を動機づけました。

デンプン顆粒(SG)は、植物種、特に ポアセ科 の胚乳に応じて異なる形態を示す。子宮内精子表現型は走査型電子顕微鏡(SEM)解析を用いてSG形態型に基づく遺伝子型を分類するために使用することができる。SGは、カーネル(pericarp、アローロン層、および子宮内精子)をスライスし、オルガネラ属の内容物を露出させることによって、SEMを使用して可視化することができる。現在の方法では、米のカーネルをプラスチック樹脂に埋め込み、ミクロトームを使用して切り離すか、切り捨てられたピペットチップに埋め込み、カミソリの刃を使用して手で切り離す必要があります。前者の方法は特殊な設備を必要とし、時間がかかり、後者は米の遺伝子型に応じて新しい問題を導入します。チョーク米の品種は、特に、彼らの子宮内精子組織の不可解な性質のために、このタイプの断面に問題を提起する。ここでは、ピペットの先端とメスの刃のみを必要とする顕微鏡用の半透明およびチョーク米カーネルセクションを準備するための技術です。ピペットチップの範囲内でセクションを準備すると、米カーネルの胚乳が粉々になり(半透明または「膜状」の)、崩れ(チョーク状のフェノタイプの場合)を防ぐことができます。この手法を用い、無傷のSGの細胞パターニングや構造を観察できる。

野生型の日本裸(図2A)と ssg1 セクション(図2B)を、260x、920x、4200xの3つの倍率で調べた。この技術は、十分な質のセクションを作製し、全ての胚乳細胞(図3A)、化合物デンプン顆粒(図3B)、および個々のサブ顆粒(図3C)を観察する。Husked カーネルは、切断前に摩耗によって除去する必要があるため、磨かれたカーネルよりも処理に時間がかかります。チョークカーネルはまた、切断中にカーネルを粉砕しないように注意しなければならないので、研磨された半透明のカーネルよりも処理に時間がかかります。適切に準備された米の部分は、約0.9mmの厚さ(表1)で、子宮内精子(図1N)と無傷の腹膜およびアローロン層の粉砕を最小限に抑える必要があります(図1O)。切り離し時にピペットチップにメスを不適切に配置すると、セクションが欠けする可能性があります(図1P)。同様に 、ssg1 の最適な横切りセクションの明るいフィールド画像(図1Q)は、無傷の子宮内精子、包皮、およびアローロン層をそのまま可視化し、可視化可能に示した(図1R)。SG を観察するだけなら、壊れたチョークカーネルセクション(図1S)は、まだ可視化に使用できるかもしれませんが、子宮内精子細胞パターンは見えません。壊れたセクションは、解析のために扱うのが難しい場合があります。細胞は ssg1 カーネルよりもしっかりと詰め込まれ、難易度が低いので、胚乳細胞壁のより多くの剪断が野生型の日本裸で観察された。 ssg1 セクションでは、胚芽細胞の剪断は認められず、化合物デンプン顆粒はそのままである。
図S1は、米のカーネルを切除する「望遠鏡」技術を用いた結果の信頼性を示しています。半透明のカーネル生産者として識別された米線 - 野生型耐性デンプン(RS)ハイブリッドラインXieyou 7954(オリザサティバL.インディカ)12、13、14(図S1A)とコバルト生成変異体RS11113、15(図S1B)は、光がステレオ顕微鏡を使用して見えるセクションを生成しました。対応するSEM画像は、これらのラインが「正常な」米の胚乳能を生成することを明らかにしました:しっかりと詰まった、多面体デンプン顆粒。チョーク状カーネル生産者は、市販品Yi-Tang16(図S1C)およびRS4 13、RS111115の変異体(図S1D)、白色、不透明なカーネル切片を示した。対応するSEM画像は、野生型半透明RS背景線と比較して著しく異なる形態を表示しました:デンプン顆粒は丸く、緩く詰め込まれました。野生型Xiushui11(オリザサティバL.ssp.ジャポニカ)とその変異体であるKMD1(Kemingdao1)は、昆虫の閉食を阻害するCry1Ab遺伝子を発現するKMD1(Kemingdao1)17、18、19(図S1F)は、半透明RS線に類似した切片および子宮内精子形態を呈した。
ここで紹介する技術は、表現型分析のためのチョーク状の米核のサンプルを調製するのに最適であるが、半透明の米核表現型20を切り離す利点を提供する:上記からの圧力を用いてサンプルをスライスすると、子宮内胚乳の粉砕および脱臼のリスクを低減する。サンプルは数秒以内に簡単に準備できます(表2)。この技術を用いて複数の遺伝子型を分析し、その有効性をテストした(表3)。図S2に示すように、この技術は他の種の種子に適用することができる。モデルモノコットブラキゲッテディスディスタキオンは、B-顆粒デンプン21のみを含む非常に硬い種子を産生し、これは、純粒状肉粒22に柔らかさを与えるタンパク質であるプロイヌリンAを欠いている。それでも、そのまま横断面を得ることが可能であった(図S2A)。軟白冬小麦(SWWW)から無傷の横切れ部を得るは困難であったが、実行することができる(図S2B)。SWWW種子は、B.ディタキオン種子や米のカーネルと比較すると、プロイヌリンAが高く、大きい。望遠鏡の集合体を使用して切除すると、これらの種子は頻繁に崩壊します。
