פרוטוקול זה משמש כדי לחקור כמותית את ההרכבה והמבנה של קטעים ראשוניים אקסון של נוירונים היפוקמפוס חסר AIS מורכב מראש בשל היעדר ankyrin-G ענק. כדי להתחיל, לנתח שישה עד שמונה היפוקמפי לאחר הלידה, גורים בני יום אחד עם מדיום HBSS בצלחת פטרי. קוצצים את ההיפוקמפי במספריים מנותבים לחתיכות קטנות יותר, ומעבירים אותם מהצלחת לצינור של 15 מיליליטר.לשטוף את ההיפוקמפי פעמיים עם חמישה מיליליטר של HBSS, ולהשאיר אותם 4.5 מיליליטר של HBSS לאחר הכביסה. מוסיפים 0.5 מיליליטר של 2.5% טריפסין ל-4.5 מיליליטר של HBSS, ומדגרים באמבט מים של 37 מעלות צלזיוס במשך 15 דקות, הופכים את הצינור כל חמש דקות. היפוקמפי מתעכל היטב צריך להיות דביק וליצור אשכול.במידת הצורך, להאריך את העיכול במשך חמש דקות נוספות. לשטוף היפוקמפי עם HBSS שלוש פעמים במשך חמש דקות לכל לשטוף. לאחר הכביסה, מוסיפים שני מיליליטר של HBSS ופיפט ההיפוקמפי למעלה ולמטה עם פיפטה פסטר 15 פעמים.לתמחר את הרקמה עם פיפט פסטר מלוטש 10 פעמים, ולהניח את הצינור במשך חמש דקות עד שכל הגושים להגדיר לתחתית. חזור על הטריטורציה עם הנתחים הנותרים עד שרובם ייעלמו. לאחר מכן, השתמש טיפ פיפטה מיליליטר אחד בעדינות להעביר את supernatant המכיל את הנוירונים מנותקים מנות ציפוי.מוסיפים אותו ישירות למדיום הציפוי המדגר מראש ומנערים את הצלחת בעדינות. שעתיים עד ארבע שעות לאחר הזריעה, בדקו את מנות הציפוי במיקרוסקופ קל. רוב הנוירונים היו צריכים להיצמד לתעודת הכיסוי.בעזרת מלקחיים עדינים, הופכים את החלקות הכיסוי למנות מאכילות תאי הגלייה המכילות מדיום תרבות עצבית מותנה מראש כאשר צד נקודות השעווה פונה כלפי מטה. נוירונים יכולים לגדול במנות מאכיל תאי גליה עד חודש. להאכיל נוירונים כל שבעה ימים עם מיליליטר אחד של מדיום תרבות עצבית טרי.ביום השלישי במבחנה, להפוך את הכיסוי מחליק עם צד נקודה שעווה פונה כלפי מעלה לתוך צלחת הזנת תאים גליה עם מדיום תרבות עצבית מותנית. יש לערבב 0.25 מיקרוגרם של דנ"א CRE BFP עם 0.5 מיקרוגרם של דנ"א של Ankyrin-GFP בצינור של 1.7 מיליליטר כדי להחדיר ארבע תלושי כיסוי. הוסף 100 מיקרוליטר אופטימלי.ואז לערבב את הצינור ונחו על מתלה. אם רק CRE BFP מודבק, עמוד השדרה הפלסמי של GFP משמש להתאמת הכמות הכוללת של ה- DNA. מערבבים שלושה מיקרוליטרים של מגיב transfection עם 100 microliters של אופטימלי בצינור חדש 1.7 מיליליטר ולהדגיר את הצינור במשך חמש דקות בטמפרטורת החדר.לאחר מכן, מערבבים 100 מיקרוליטרים של הדנ"א המעורב הקודם עם 100 מיקרוליטרים של תערובת מגיבה טרנס-פקציטית, ומשאירים אותו למשך חמש עד 10 דקות על מתלה. מוסיפים 50 מיקרוליטרים של תערובת הדנ"א מעל כל תלוש כיסוי על ידי החדרת הקצה ממש מתחת למדיום מבלי לגעת בכיסוי. פיפטה לאט כדי למנוע הפצת תערובת ה-DNA.לאט לאט מחזירים את המנה לאינקובטור ומשאירים אותה שם למשך 30 עד 45 דקות. הופכים את המכסה בחזרה לצלחת מאכיל הגלישה הביתית כשצד נקודה שעווה פונה כלפי מטה ומחזירים את המנה לאינקובטור. לכימות AIS, פתח את תמונות סדרת Z בפיג'י וצור הקרנה מרבית של תמונות.לאחר חיסור אות הרקע הריק של החלקת הכיסוי מהתמונה, צייר קו לאורך ה- AIS וודא שרוחב הקו מכסה באופן מלא את ה- AIS. הפעל את הקו לפני שאות ה- AIS מופעל מעל הרקע והפסק לאחר שהוא יורד לרקע. מדוד את עוצמת הפיקסל הממוצע לאורך הקו וייצא אותו לגיליון אלקטרוני.לאחר מכן, הפעל סקריפט MATLAB כדי ליצור את הדמות הסופית. הניסוי צריך לכלול Cre-BFP רק transfection כמו שליטה שלילית, Cre-BFP בתוספת 480 קידוד ankyrin-G cotransfection כמו שליטה חיובית, ומצב שאינו transfected כמו בקרת טכניקה. בשליטה Cre-BFP רק, נוירונים transfected חסר הצטברות של סמני AIS, כולל ankyrin-G, בטא 4 spectrin, נוירופסין, מתח מגודר ערוצי נתרן.לעומת זאת, Cre ו 480 קילודאלטון ankyrin-G שיתוף transfected נוירונים יש מורכבים באופן מלא AIS, אשר מודגם על ידי נוכחות של סמני AIS. חשוב לאשר את איכות התרבות בהשוואה למנות שאינן מודבקות. נוירונים לא בריאים נוטים להראות מבנה AIS חריג, כמו AIS הופסק או חוץ רחמי.כדי להעריך כיצד מוטציה בהפרעה נוירו-התפתחותית אנושית ankyrin-G משפיעה על הרכבת AIS, עוצמת AIS הממוצעת הותוותה מהסומה לאקסון הדיסטלי. חלבונים מועשרים AIS בדרך כלל מראים עלייה מהירה של אות מן האקסון proximal וירידה איטית של אות אקסון דיסטלי. כאשר הוא מיושר עם ה- AIS שאינו מודבק, עקומת המוטנטים רחבה יותר ושיא העקומה נמוך יותר, דבר המצביע על שינוי מבנה של AIS.סוג הפרא ankyrin-G אסף AIS מיושר באופן הדוק עם אחד שאינו מודבק.
