このプロトコルは、巨大なアンキリンGがないために組み立てられたAISを欠いている海馬ニューロンの軸索初期セグメントの組み立てと構造を定量的に研究するために使用される。まず、出生後6~8匹の海馬、ペトリ皿にHBSS培地を持つ1日齢の子犬を解剖する。解剖はさみで海馬を小さく刻み、皿から15ミリリットルのチューブに移します。5ミリリットルのHBSSで海馬を2回洗い、洗浄後に4.5ミリリットルのHBSSに残します。2.5%トリプシンの0.5ミリリットルを4.5ミリリットルのHBSSに加え、37°Cのウォーターバスで15分間インキュベートし、5分ごとにチューブを反転させます。よく消化された海馬は粘着性になり、クラスターを形成する必要があります。必要に応じて、消化をさらに5分間延長します。HBSSで海馬を1回5分間3回洗います。洗浄後、HBSSを2ミリリットル加え、パスツールピペットでカバカンピを上下にピペットします。火でティッシュを磨いたパスツールピペットを10回トリチュレートし、すべての塊が底にセットされるまで5分間チューブを休ませます。残りのチャンクの大部分が消えるまで、分割を繰り返します。次に、1ミリリットルのピペットチップを使用して、解体したニューロンを含む上清をめっき皿にそっと移します。事前インキュベートされためっき媒体に直接加え、プレートを軽く振ります。播種後2~4時間、めっき皿を軽顕微鏡で確認します。ニューロンの大部分は、カバースリップに取り付けられているはずです.細かい先端鉗子を使用して、カバースリップを、前もって調整された神経細胞培養培地を含むグリア細胞フィーダーディッシュに反転させ、ワックスドット側を下向きにします。ニューロンは、グリア細胞フィーダー皿で最大1ヶ月間成長することができます。新鮮な神経細胞培養培地の1ミリリットルで7日ごとにニューロンを供給します。インビトロで3日目に、ワックスドット側を上に向けてカバースリップを、調節された神経細胞培養培地を備えたグリア細胞フィーダー皿に反転させます。CRE BFP DNAの0.25マイクログラムと0.5マイクログラムのアンキリンGFP DNAを1.7ミリリットルチューブに混ぜて、4つのカバースリップをトランスフェクトします。最適な100マイクロリットルを加えます。その後、チューブを混合し、ラックの上に置いた。CRE BFPのみがトランスフェクションされた場合、GFPプラスミックバックボーンを使用してDNAの総量に一致します。新しい1.7ミリリットルチューブに最適な100マイクロリットルのトランスフェクション試薬を3マイクロリットル混合し、室温で5分間チューブをインキュベートします。次に、以前混合したDNAの100マイクロリットルを100マイクロリットルのトランスフェクション試薬ミックスと混合し、ラックに5〜10分間放置します。カバースリップに触れることなく、先端を媒体のすぐ下に挿入して、各カバースリップの上にDNAミックスの50マイクロリットルを追加します。ピペットはゆっくりとDNAミックスを拡散しないようにします。ゆっくりと皿をインキュベーターに戻し、30〜45分間そこに置いておきます。カバースリップをホームグリアフィーダー皿に戻し、ワックスドット側を下に向け、皿をインキュベーターに戻します。AIS の定量化の場合は、フィジーで Z シリーズ画像を開き、画像の最大投影を生成します。空のカバースリップの背景信号を画像から引いた後、AIS に沿って線を引き、線の幅が AIS を完全に覆っていることを確認します。AIS 信号がバックグラウンドの上に上がる前にラインを開始し、バックグラウンドにドロップした後で停止します。線に沿って平均ピクセルの強度を測定し、スプレッドシートにエクスポートします。次に、MATLAB スクリプトを実行して、最終的な図を生成します。実験には、陰性対照としてトランスフェクションのみを含むCre-BFP、正の対照としてCre-BFPプラス480キロダルトンアンキリンGコトランスフェクション、および技術制御として非トランスフェクション状態を含める必要があります。Cre-BFPのみのコントロールでは、トランスフェクトされたニューロンは、アンキリンG、β4スペクトリン、ニューロファシン、および電圧ゲートナトリウムチャネルを含むAISマーカーの蓄積を欠いている。対照的に、Creと480キロダルトンアンキリン-G共トランスフェクションニューロンは、AISマーカーの存在によって実証されたAISを完全に組み立てています。非トランスフェクションされた料理との比較を通じて、培養の質を確認することが重要です。不健康なニューロンは、中止または異所AISのような異常なAIS構造を示す傾向がある。アンキリン-Gヒト神経発達障害突然変異がAIS集合体に与える影響を評価するために、平均AIS強度を相馬から遠位軸索にプロットした。AISの富化タンパク質は、通常、近位軸索からのシグナルの速い増加と遠位軸索への信号のゆっくりとした減少を示す。非トランスフェクトAISと整列すると、変異曲線は広く、曲線のピークは低く、AISの構造変化を示唆している。野生型アンキリンGは、非トランスフェクトされたものと密接に一致するAISを組み立てた。
