B型肝炎は20億人以上の人々の生活に影響を与え、公衆衛生に対する大きな脅威の1つです。世界中で約2億5,700万人が慢性B型肝炎ウイルス(HBV)に感染しており、社会に大きな負担を与えています。肝細胞は、腎臓や精巣などの非肝組織におけるHBVおよび他の細胞に感染した唯一の細胞ではなく、このウイルス22、33にも感染している。現在、HBV感染細胞モデルは、非肝細胞系モデルの数が少ないヒト肝細胞に限定されている。これは、HBV感染および非肝組織のHBV関連病理の研究を妨げる。ここでは、非肝細胞293T細胞および肝細胞におけるHBV感染を研究するためのプロトコルを提示する。

HepG2.2.15細胞は、HepG2線に由来し、感染性HBVを慢性分泌し、ならびにサブウイルス粒子を培地66,77に分泌する。シクロスポリンA(CsA)は、異種移植片拒絶反応の抑制のために臨床的に用いられる免疫抑制剤である。研究は、CsAがNTCPのトランスポーター活性を阻害し、NTCPの大きなエンベロープタンパク質8への結合を遮断することによって培養肝細胞へのHBVの侵入を阻害することを示している。
HepG2-NE細胞は陽性対照として用いたのに対し、CsA処理細胞は陰性対照として用いた。陽性対照群と陰性対照群と比較することで、HBV感染においてどの宿主遺伝子が重要な役割を果たすかを調べることができる。さらに、HBV感染のこのモードを介して、HBV感染に関与する他の新しいメカニズムまたは宿主遺伝子を見つけることもできます。

Modeling Hepatitis B Virus Infection in Non-Hepatic 293T-NE-3NRs Cells
