Malaria er forårsaget af Plasmodium parasitter og overføres til deres hvirveldyr værter via infektiøse bid af kvindelige Anopheles myg. Ifølge Verdenssundhedsorganisationens (WHO) rapport fra 2019 var der anslået 405.000 dødsfald fra i alt 228 millioner tilfælde af malaria1. De fleste malariarelaterede dødsfald var koncentreret i den afrikanske region, især blandt børn under fem år. Mens den samlede forekomst af malaria er faldet globalt fra 2010, i de seneste år tilbagegang har plateaued og yderligere kontrol strategier er presserende behov for at fjerne sygdommen.
Cykliske aseksuelle blod stadier af malaria parasitter forårsage sygdom patogenese og en lille delmængde af disse differentiere sig til kvindelige og mandlige gametocytter. Plasmodium falciparum gametocytter er unikke i naturen, da de tager 7-10 dage at udvikle gennem fem morfologisk forskellige stadier. Umodne gametocytter fra fase I til IV er afsondret i knoglemarvsparenkym og forbliver stort set fraværende fra perifer cirkulation2,3,4,5. Erythrocytter inficeret med modne fase V gametocytter frigives i blodbanen og cirkulerer frit for at blive taget op af myg. En gang inde i myg midgut aktiveres gametocytter gennem en ændring i temperatur og eksponering for midgut-miljøet til kvindelige og mandlige gameter og begynder udviklingen af mygstadiet, som kulminerer med de infektitive stadier af sporozoitter i mygspytkirtlerne6,7.
Siden Trager og Jenson8 beskrev en standardiseret metode til kultur P. falciparum, undersøgelser af de aseksuel blod faser har langt fremskreden. Manglen på et pålideligt kultursystem for seksuelle stadier har imidlertid gjort det vanskeligt at studere P. falciparum gametocytter, transmissionsbiologi og myggestadier. I de senere år er der blevet offentliggjort flere metoder , som har hjulpet laboratorier med at etablere vildtatcytkulturer9,10,11,12. Dette manuskript beskriver standardiserede og pålidelige protokol til kultur P. falciparum gametocytter, der kan repræsentere en værdifuld ressource for malaria forskning samfund. Malaria er fortsat et af de vigtigste folkesundhedsproblemer, der forårsager betydelig sygelighed og dødelighed. Malaria er en mygbåren sygdom, der overføres gennem en smitsom bid fra den kvindelige Anopheles myg. Malaria kontrol vil i sidste ende stole på et væld af tilgange, som omfatter måder at blokere transmission til, gennem og fra myg. For at studere myg stadier af malaria parasitter i laboratoriet, har vi optimeret en protokol til kultur meget smitsomme Plasmodium falciparum gametocytter, en parasit fase kræves til overførsel fra den menneskelige vært til myg vektor. P. falciparum gametocytter modnes gennem fem morfologisk forskellige trin, hvilket tager cirka 1-2 uger. Gametocyte kultur beskrevet i denne protokol er afsluttet i 15 dage og er smitsom for myg fra dag 15-18. Disse protokoller blev udviklet for at opretholde en kontinuerlig cyklus af infektion kompetente gametocytter og for at opretholde uafbrudt forsyning af myg stadier af parasitten. Her beskriver vi metoden for gametocytkultur og hvordan man inficerer myg med disse parasitter ved hjælp af glasmembranfødere.

Her præsenterer vi resultater fra en række membran feeds ved hjælp af P. falciparum NF54 gametocyte kulturer genereret ved hjælp af protokollen ovenfor (se (Figur 5). Gametocyte kultur blev indledt med ca. 0,5% blandet fase aseksuel kultur på dag 0, som voksede til et højdepunkt parasitmia på ca. 15% af dag 4 og dag 5.
