Conpokal kombinerer konfokal mikroskopi med atomkraftmikroskopi ved hjelp av en sonde for å stikke prøveoverflaten. Selv om begge teknikkene er effektive individuelt, forenkler Conpokal fluorescenskon-lokalisering med mekanisk karakterisering. Den største fordelen med Conpokal-teknikken, er den nære samtidige dual-mikroskopien.
Confocal undersøker cytoskeletal og andre cellulære prosesser før og etter AFM-sondering, som leverer områdespesifikke mekaniske egenskaper. Denne teknikken er virkningsfull innenfor mekanobiologifeltet som for eksempel kan hjerneceller undersøkes under fysiologiske forhold for å undersøke elektriske impulser og krafttransaksjon. Før du begynner analysen, velger du en passende AFM-kantilever for ønsket datainnsamling og bruk hansker, AFM-monteringstrinnet, pinsett og en liten skrutrekker for å montere brikken i glassblokken.
Plasser AFM-brikken forsiktig på midten av glassblokken. Cantilever pluss en svært liten del av AFM-brikken skal være i den synlige, ikke-ugjennomsiktige delen av glassblokken. Bruk skrutrekkeren til å stramme skruen til brikken er tettsittende mot glassblokken, og bruk et objektiv for å kontrollere at brikken er riktig orientert.

Bruk lys feltbelysning og Z stepper motor kontrollpanel, sakte senke AFM chip til bunnen av glassfatet i trinn på 100 til 200 mikron for å unngå å krasje AFM spissen i petriskålen.
Finn de manuelle mikrometer som styrer X Y eller i vanlig bevegelse av AFM-hodet på instrumentplattformen. Etter hvert som skyggen fra AFM-cantilever blir mørkere, og formen blir skarpere, bruk de manuelle mikrometerene til å korrigere AFM-hodet og justere posisjonen til AFM-kantileveren innen synsfeltet. På dette tidspunktet kan det hende at oppslagstabellene må justeres.
Se etter en skygge som skal vises i mikroskopprogramvarevisningen, noe som indikerer at AFM-spissen nærmer seg bunnen av parabolen. Fortsett å bruke små trinn for å senke AFM-spissen til spissen for det meste er i fokus. Når laseren er i posisjon, bruk laserjusteringsskivene til å plassere laseren på baksiden av der AFM-spissen er plassert på kantileveren.
Laserjusteringspanelet skal vise et sumsignal som er større enn null volt. Flytt laseren, en liten avstand i alle retninger på AFM-kantileveren, til det maksimale sumsignalet oppnås mens det gjenstår på AFM-spissen. Når laserposisjonen er angitt, bruker du de manuelt kontrollerte avbøyningsskivene til å nulle den vertikale og laterale nedbøyningen.
Bruk de vertikale og laterale nedbøyningsknappene til å justere detektoren slik at målet er sentrert og det ikke er observert vertikal eller lateral nedbøyning i laserjusteringspanelet. Åpne Kalibrering-vinduet, og skriv inn all spesifikk informasjon om eksperimenter. Skift laserlysfilteret.
Før du kalibrerer, må du slå av den konfokale mikroskoplyskilden og lukke AFM-kabinettet for å dempe potensiell støy som kommer fra romlyset eller vibrasjonene. Trykk på Kalibrering-knappen for å la systemet kalibrere spissen automatisk. Når kalibreringen er fullført, vises stivheten til kantileveren og følsomheten i kalibreringspanelet.
Bruk deretter kommandoen Automatisert tilnærming til å senke tuppen til bunnen av prøvefatet og angi størrelsen på hudområdet, oppløsningen, settpunktet, Z-lengden og pikseltiden, før du trykker på avspillingsknappen for å begynne å skanne. For konfokal mikroskopavbildning i mikroskopprogramvaren, aktiver konfokale evner og velg laserlinjene som passer for fargestoffene som ble brukt til å flekke prøvene. Velg én eller flere laserlinjer for å opphisse og bilde disse funksjonene i prøven og sette gevinsten til en verdi som optimaliserer prøvefluorescensen, men begrenser mengden støy.
Juster laserkraften for å unngå mettede piksler samtidig som du maksimerer det dynamiske området og setter pinholestørrelsen til én områdeenhet for å maksimere oppløsningen for den optiske snittingen. Juster om nødvendig verdiene basert på prøvelystheten. Hvis du vil angi pikselbotid, begynner du med omtrent to mikrosekund oppholdstid, og justerer for å gjenspeile prøvelysheten etter behov.
Hvis du vil velge pikselstørrelsen for det valgte målet, lar du instrumentet beregne pikselstørrelsen via Nyquist-alternativknappen og det valgte antallet piksler i bildet. Deretter velger du alternativet Skann og starter datainnsamlingen. Bruk fokusknappen til å finne fokusplanet som representerer prøvefunksjonene i sin klareste form, Skann-knappen for å starte datainnsamling og Capture-knappen for å ta et todimensjonalt bilde.
Bruk panelet som styrer pikselstørrelsen via Nyquist-alternativet, reduser områdestørrelsen på skanningen for å bare omslutte en enkelt celle. Aktiver oppsamlingsverktøyet og bruk et alternativ fra bunn til topp, med bare laserlinjen som lyser opp en funksjon i prøven tydeligst, sett start- og sluttplanene for volumet som skal måles ved hjelp av den foreslåtte avstanden mellom planene. Gi filen et navn, og lagre den i den tilknyttede mappen.
Alternativt, hvis vår priori kunnskap om prøvetykkelsen eksisterer, velg den lyseste, skarpeste, midtre plan ved å justere fokus for å definere volumet og sette toppen for å ha prøvetykkelsen over det midterste planet, og bunnen for å ha tykkelsen under det midterste planet, og velg deretter riktig trinnstørrelse. Kjør oppkjøpet ved å trykke på Kjør nå-knappen. Når anskaffelsen er fullført, lagrer du filen i den aktuelle mappen.
På slutten av eksperimentet, bruk hansker mens du fjerner glassblokken og plasserer den i monteringsstasjonen. Bruk pinsett med gummigrep for å fjerne AFM-chipen forsiktig og plasser den brukte spissen tilbake på plasseringen av oppbevaringsboksen orientert av tilgang for å betegne at den har blitt brukt. For fremtidig referanse, legg merke til hvilket tips som ble brukt i eksperimentet.
Her vises en representativ AFM-skanning over en levende HEK-celle. Høyden for denne spesielle HEK-cellen var rundt 10 mikrometer som demonstrert av linjeskanningen. Her kan et eksempel på en dårlig skanning på grunn av et feil AFM-tipsvalg observeres.
I dette bildet dukket svarte piksler opp på toppen av cellen som indikerer at AFM PA-sonen var utenfor rekkevidde på grunn av en stor cellehøyde. Slutten av AFM-cantilever vises også på bildet på grunn av spissens forskyvning kombinert med utilstrekkelig spisshøyde sammenlignet med cellehøyden. Disse artefaktene i AFM-bildet indikerer at en annen AFM-spiss burde ha blitt valgt for å bilde cellen.
Tre fargekonokalavbildning kan utføres. For eksempel, for å visualisere cellekjernen, mikrotubuli og lipofil membran. Analyse av spissens innrykk i kombinasjon med nano mekanisk modell, tillater generering av et moduluskart over overflaten.
Her vises en tilsvarende 3D-projeksjon av laserkonferanse Z-stakken. AFM-skanninger og målte moduluskart kan også anskaffes for mikrober, som observert i disse representative analysene av streptokokker mutans bakterie. Som demonstrert kan en bedre oppløsning oppnås i denne skalaen med AFM, enn med tradisjonell konfokal mikroskopi.
Adhesive force mapping er en teknikk utført gjennom conpokal for å visualisere molekylære interaksjoner. For eksempel, for å studere modulering av celleoverflatemolekyler for å observere bindende kinetikk. Conpokal innleder en ny vei for å utforske strukturfunksjonsrelasjoner i medisinsk mikrobiologi.
For eksempel kan fysiske og kjemiske hendelser og peptidoglykanlaget knyttes til antibiotikaresistens.
Presentert her er protokollen til Conpokal teknikken, som kombinerer konfokal og atomkraft mikroskopi i en enkelt instrument plattform. Conpokal gir samme celle, samme region, nær samtidig konfokal avbildning og mekanisk karakterisering av levende biologiske prøver.
