biomimetic replication of root surface microstructure using alteration of soft lithography

要形成根面微观结构复制，必须选择根进行成型。我们在土壤中种植番茄植物，使得利用根系的天然根部极具挑战性。从根系中去除土壤可能很困难，此外，根系根系的根部是脆弱的，在成型尝试时可能会断裂。因此，我们建议首先使用更严格的根，在实验室中建立协议。图1A描述了这种根的形成。植物根系在植物生长3周后被移除，无根植物在水中放置约一周，直到从茎中产生出的生根。这些根可用于在协议建立期间的复制。一旦协议建立良好，需要一个更逼真的根面结构。在这里，我们建议避免根生长在土壤中，因为完全去除土壤在极具挑战性。相反，我们建议使用发芽根，提供基因特异性植物的根面微观结构的宝贵信息。图1B中描述了这种根的生长。种子放在湿滤纸上，在25°C下孵育。大约 5 天后，过滤纸保持湿润，发芽的根足够长，可以复制。这些根比先前建议的根更脆弱，需要更微妙的照顾。
根面微观结构副本的生产是一个两步过程。在第一步中，自然根被模塑成聚氨酯基模具（负仿）。此步骤的优点是，聚氨酯模具的所有材料都在准备中，根部在最接近尾声时放在准备好的溶液上，10分钟暴露在紫外线下。因此，生物组织不会暴露在恶劣条件下太久，可以在过程结束时轻轻处理。如果遵循所有协议步骤，则生成一个良好的负副本。此副本将显示根面的细胞结构以及表示根毛位置的孔（图 2A）。如果未遵循协议中某些关键步骤，则该过程将失败。其中一个步骤是将根放在聚氨酯溶液上，然后固化。根必须非常轻轻地放置，以避免其淹没在聚氨酯溶液中。这种淹没，任何部分的根，将导致根在硬聚合物的陷阱，没有能力去除它。如果发生此类事件，根根在固化后将保留在负副本中（图 2B）。另一个关键步骤是紫外线的固化时间。推荐的固化时间是8\u201210分钟。超过10分钟将导致极硬的聚氨酯模具，使得在不破坏聚氨酯模具中去除根部是不可能的。根的破损有时可以肉眼可见，例如，当一大块破损时（图2C，顶部，标有紫色箭头）。然而，有时小根片留在材料中，这是很难发现肉眼和显微镜必须使用（图2C，底部，标有紫色箭头）。我们建议在协议延续之前，用显微镜仔细检查聚氨酯负复制体，以确保不存在残留根。
一旦聚氨酯负复制品被准备好;许多材料可用于制备正副本。正复制品的制备，使用聚氨酯负复制品作为模具，是直截了当的，完全取决于聚氨酯负复制品的质量。为了生成正副本，我们同时使用了PDMS（在软光刻领域（图3A）和乙基纤维素（12.18中）作为更好地模仿主要由纤维素构成的根面特性的材料（图3B）。PDMS 副本的 SEM 图像非常清楚地显示根毛。头发在伸长区，在那里他们开始出现。因此，根毛的长度沿根面变化，因为它们变长，很像在自然根（图3A）。乙基纤维素产生的薄膜比PDMS更硬、更不灵活。因此，从负模中去除它需要更多的注意。然而，一些毛发和表面微观结构在光显微镜下可见（图3B）。我们使用这两种材料生成正副本，但是，任何可以形成薄膜的材料都将是正复制品的候选材料，使用聚氨酯负复制品。
