開発中マウス胚盤胞1,2の内細胞塊からの初の導出以来、マウス胚性幹細胞(mESC)は幹細胞自己再生と分化を研究する強力なツールとして用いられてきた3。さらに、mESC分化を研究することは、神経変性疾患などの疾患の治療における幹細胞ベース療法における効率と安全性を向上させる可能性のある分子機構の多大な理解につながる4。動物モデルと比較して、このin vitroシステムは、実際と評価におけるシンプルさ、動物とは対照的に細胞株を維持する際の低コスト、および遺伝子操作における相対的な容易さなど、多くの利点を提供する。しかし、分化された細胞タイプの効率と品質は、多くの場合、mESCの異なるラインと分化方法5、6の影響を受けます。また、分化効率を評価する従来のアッセイは、堅牢性に欠ける選択されたマーカー遺伝子の定性的検査に依存しているため、遺伝子発現の世界的な変化を把握することができません。
ここでは、神経分化の系統的評価にアッセイの電池を使用することを目指しています。選択したマーカーとRNA-seqに対する従来のインビトロ分析を用いて、分化効率とこのプロセス中の転写変化を測定するためのプラットフォームを確立します。既に確立されたプロトコル7に基づいて、吊り下げ低下技術を通じて胚体(EB)を生成し、続いて上端菌量のレチノイン酸(RA)を用いて神経前駆細胞(NPC)を生成し、その後神経誘導培地でニューロンに分化した。分化の効率を調べるために、従来のRT-qPCRおよび免疫蛍光(IF)アッセイに加えて、RNA-seqおよびフローサイトメトリーを実施した。これらの分析は、ステージ固有の分化の進行を包括的に測定します。

differentiation and characterization of neural progenitors and neurons from mouse embryonic stem cells

マウス胚性幹細胞を神経系統に分化し、次に分化した細胞を特徴付ける一連のアッセイを行うステップバイステップの手順について述べています。E14マウス胚性幹細胞を使用して吊り下げドロップ法を介して胚体を形成し、次にレチノイン酸によって神経前駆細胞に分化し、最終的にニューロンに分化した。定量的逆転写ポリメラーゼ連鎖反応(RT-qPCR)および免疫蛍光実験により、神経前駆物質およびニューロンが、8日目と12日目の分化後に対応するマーカー(神経前駆物質用のネチンとニューロンの神経フィラメント)をそれぞれ示すことが明らかになった。 Sox1 プロモーター主導のGFPレポーターを発現するE14ラインでのフローサイトメトリー実験は、8日目の細胞の約60%がGFP陽性であることを示し、この段階で神経前駆細胞の分化が成功したことを示した。最後に、RNA-seq分析を使用して、世界的な転写変化をプロファイリングしました。これらの方法は、神経細胞の分化中の細胞の同一性遷移を調節する際の特定の遺伝子および経路の関与を分析するのに有用である。
