differentiation and characterization of neural progenitors and neurons from mouse embryonic stem cells

우리는 마우스 배아 줄기 세포를 뉴런 혈통으로 배양하고 분화하기 위한 단계별 절차를 설명하고, 분화된 세포를 특징짓는 일련의 에세이를 따릅니다. 상기 E14 마우스 배아 줄기세포는 매달려 있는 낙하 방법을 통해 배아체를 형성하기 위해 사용되었고, 그 후 망막산에 의해 신경 전구 세포로 분화하도록 유도하고, 마지막으로 뉴런으로 분화하였다. 정량적 역전사 폴리머라제 연쇄 반응(RT-qPCR) 및 면역형광 실험은 신경 전조자와 뉴런이 각각 8일째와 12일째에 대응하는 마커(신경 전조자 및 뉴런용 신경필라멘트용 네신틴)를 나타낸다는 것을 밝혔다.
