La resonancia magnética funcional (fMRI) es un método no invasivo para medir las respuestas hemodinámicas 1,2,3,4,5,6,7,8,9, por ejemplo, la señal de flujo y volumen sanguíneo dependiente del nivel de oxígeno en sangre (BOLD), asociada con la actividad neuronal en el cerebro. En estudios con animales, las señales hemodinámicas pueden ser afectadas por la anestesia 10, el nivel de estrés de animales despiertos11, así como los posibles artefactos no fisiológicos, por ejemplo, pulsaciones cardíacas y movimientos respiratorios12,13,14,15. Aunque se han desarrollado muchos métodos de posprocesamiento para proporcionar un análisis retrospectivo de la señal de fMRI para la dinámica funcional relacionada con la tarea y el estado de reposo y el mapeo de conectividad16,17,18,19, existen pocas técnicas para proporcionar una solución de mapeo de la función cerebral en tiempo real y lecturas instantáneas en el cerebro animal 20 (la mayoría de las cuales se utilizan principalmente para el mapeo del cerebro humano 21, 22,23,24,25,26,27). En particular, este tipo de método de mapeo de fMRI en tiempo real falta en los estudios con animales. Es necesario establecer una plataforma de resonancia magnética funcional para permitir la investigación de las etapas fisiológicas dependientes del estado cerebral en tiempo real y proporcionar un paradigma de estimulación de biorretroalimentación en tiempo real para estudios funcionales del cerebro animal.
En el presente trabajo, ilustramos una configuración experimental de fMRI en tiempo real con las macrofunciones personalizadas del software de consola de MRI, demostrando el monitoreo en tiempo real de las respuestas evocadas BOLD-fMRI en la corteza somatosensorial de la pata anterior primaria (FP-S1) de las ratas anestesiadas. Esta configuración en tiempo real permite la visualización de la activación cerebral en curso en mapas funcionales, así como cursos de tiempo individuales de una manera voxel-wise, utilizando el software de análisis de neuroimagen existente, Análisis de NeuroImágenes Funcionales (AFNI)28. real-time fmri brain mapping in animals

Las respuestas dinámicas de fMRI varían en gran medida de acuerdo con las condiciones fisiológicas de los animales, ya sea bajo anestesia o en estados despiertos. Desarrollamos una plataforma de resonancia magnética funcional en tiempo real para guiar a los experimentadores a monitorear las respuestas de resonancia magnética funcional instantáneamente durante la adquisición, que se puede utilizar para modificar la fisiología de los animales para lograr las respuestas hemodinámicas deseadas en los cerebros de los animales. La configuración de fMRI en tiempo real se basa en un sistema de resonancia magnética preclínica de 14.1T, lo que permite el mapeo en tiempo real de las respuestas dinámicas de fMRI en la corteza somatosensorial primaria de la pata anterior (FP-S1) de ratas anestesiadas.
