In dit document hebben we een protocol opgesteld voor het efficiënt isoleren en kweken van zuivere en levensvatbare primaire menselijke NK-cellen uit perifeer bloed. We hebben ook de voorwaarden geoptimaliseerd voor de meting van de metabolische activiteit van deze NK cellen beoordeeld door zuurstofverbruik en extracellulaire verzuring met behulp van een extracellulaire flux analyzer. Natural Killer (NK) cellen zijn aangeboren lymfocyten die anti-tumor en anti-virale reacties bemiddelen. NK-cellen bestaan uit 5-15% van alle lymfocyten in menselijk perifeer bloed, en kunnen ook worden gevonden in milt, lever, beenmerg en lymfeklieren. NK-cellen drukken geen polymorfe clonotyp receptoren uit, zoals T-celreceptoren (TCR) of B-celreceptoren (BCR). De activering van hun cytolytische functies daarentegen wordt ingegeven door de betrokkenheid van receptoren die onveranderlijke liganden herkennen op het oppervlak van een doelcel1,2.
Rustende menselijke NK cellen geïsoleerd uit perifeer bloed kan overleven voor meerdere dagen in cultuur medium aangevuld met menselijk serum. Activering van NK-cellen door cytokinen zoals IL-15 of IL-2 drijft de cellen tot proliferatie en tot een toename van hun moordvermogen, onder andere effecten3,4,5. Verschillende studies hebben een directe correlatie aangetoond tussen NK-celactivering en veranderingen in hun metabolische activiteit6. Deze metabolische veranderingen zijn bestemd om te voldoen aan de specifieke eisen van de cellen in termen van energie en biosynthese.
Aërobe cellen en organismen verkrijgen energie door middel van een reeks chemische reacties die de katabolisme en oxidatie van koolhydraten, vet en eiwitten te betrekken. Door een combinatie van glycolyse, de tricarboxylzuurcyclus (TCA) en oxidatieve fosforylatie voldoen eukaryotische cellen aan de meerderheid van hun ATP-vraag en verkrijgen ze tussenproducten die nodig zijn als bouwstenen voor macromolecules die essentieel zijn voor celgroei en proliferatie. Het proces van glycolyse (figuur 1A) begint met de invoer van glucose in de cel. In de cytosol wordt glucose gefossyleerd en omgezet in pyruvaat (met een nettoproductie van 2 moleculen ATP per glucosemolecuul), die kunnen worden gereduceerd tot lactaat of getransporteerd naar de mitochondriën om te worden omgezet in Acetyl-CoA en de TCA-cyclus binnen te gaan. De TCA-cyclus blijft fietsen gevoed met meer moleculen van Acetyl-CoA en produceert CO2 (die uiteindelijk buiten de cel zal verspreiden en, door te reageren met H2O in het medium, zal genereren koolzuur dat zal leiden tot de verzuring van het medium) en NADH, het molecuul belast met het doneren van elektronen aan de elektronentransportketen (ETC). De elektronen reizen door verschillende eiwitcomplexen en worden uiteindelijk geaccepteerd door zuurstof. Deze complexen (I, III en IV) pompen ook H+ van de mitochondriale matrix in de intermembrane ruimte. Als gevolg van de elektrochemische gradiënt gegenereerd, zal de H+ opnieuw in de matrix via de complexe V (ATP-synthase), het investeren van de potentiële energie verzameld in de generatie van ATP.
Zowel glycolyse als mitochondriale ademhaling kunnen op verschillende punten worden geblokkeerd met behulp van remmers. De kennis en het gebruik van deze remmers was de basis voor de ontwikkeling van de extracellulaire fluxtest. Door twee eenvoudige parameters in real time te meten, zoals pH en zuurstof, leidt de extracellulaire flux analyzer de snelheid van glycolyse en mitochondriale ademhaling af in een 96-put plaat. De glycolysestresstest wordt uitgevoerd in een basaal medium zonder glucose (figuur 1B)7. De eerste metingen van de extracellulaire verzuring (ECAR) zijn indicatief voor glycolyse-onafhankelijke verzuring. Het wordt aangeduid als niet-glycolytische verzuring en correleert met CO2 geproduceerd door de TCA die, zoals eerder uitgelegd, combineert met H2O in het medium om H+ (TCA-gekoppelde ECAR) te genereren. De eerste injectie is glucose om glucosegebruik te induceren en glycolyse te stimuleren. De tweede injectie combineert zowel rotenone, een Complexe I-remmer, en antimycine A, een Complexe III-remmer samen, om de ETC. Cellen te blokkeren reageren op deze dramatische afname van mitochondriale ATP-productie door het activeren van glycolyse om cellulaire ATP-niveaus te handhaven, en dit vertegenwoordigt de hoeveelheid glycolyse die niet wordt gebruikt door de cel in de basale toestand, maar mogelijk kan worden aangeworven als reactie op de toename van de ATP-vraag (compenserende glycolyse). De derde injectie is de glucose analoge 2-Deoxyglucose (DG), die concurreert met glucose als substraat voor het enzym hexokinase. Het product van deze fosforylatie, 2-deoxyglucose-6-fosfaat kan niet worden omgezet in pyruvaat, en daarom glycolyse wordt geblokkeerd, waardoor de ECAR tot zijn minimum wordt beperkt. De op dit punt gemeten ECAR omvat andere bronnen van extracellulaire verzuring die niet worden toegeschreven aan glycolyse of ademhalingsactiviteit, evenals eventuele resterende glycolyse die niet volledig door 2-DG (post 2-DG-verzuring) wordt geremd.
De mitochondriale stresstest wordt uitgevoerd in een medium met glucose (figuur 1C)8. De eerste metingen van het zuurstofverbruik (OCR) komen overeen met de basislijn van mitochondriale ademhaling (basale ademhaling). De eerste injectie is oligomycine, die de terugkeer van protonen via de ATP-synthase (complex V) remt, de ATP-synthese blokkeert en zo snel het mitochondriale membraan hyperpolariseert, wat verdere protonpompen door ademhalingscomplexen voorkomt en leidt tot een afname van OCR. De vergelijking tussen de basisinademing en de waarde die wordt gegeven door toevoeging van oligomycine vertegenwoordigt de atp-gekoppelde ademhaling. De resterende oligomycine-ongevoeligheidsgraad van zuurstofverbruik wordt protonlek genoemd, dat de stroom van protonen door de lipidebilayer of eiwitten in het binnenste mitochondriale membraan vertegenwoordigt, zoals de adenine nucleotide translocase9. De tweede injectie is de ontkoppelaar 2,4-dinitrophenol (DNP), een ionofoër die een massale binnenkomst van H+ in de mitochondriale matrix induceert, wat leidt tot depolarisatie van het mitochondriale membraan en verstoring van mitochondriale ATP-synthese. Cellen reageren op de dissipatie van de proton-bewegingskracht door het verhogen van de snelheid van elektronentransport en zuurstofverbruik tot maximale niveaus in een vergeefse poging om membraan potentieel (maximale ademhalingscapaciteit) te herstellen. Het verschil tussen de maximale ademhalingscapaciteit en de basale ademhaling is de reserveademhalingscapaciteit van de cel, die de hoeveelheid ademhaling vertegenwoordigt die niet door de cel wordt gebruikt om ATP in de basale toestand te genereren, maar mogelijk kan worden aangeworven als reactie op een toename van de ATP-vraag of onder stressomstandigheden8. De derde injectie is een combinatie van rotenone en antimycine A. Deze injectie stopt volledig de ETC en OCR daalt tot het laagste niveau, met de resterende zuurstofverbruik wordt niet-mitochondriaal (veroorzaakt door NADPH-oxidases, enz.).
Veranderingen in metabole trajecten kunnen op de een of andere manier de werking van NK-cellen voorspellen, omdat is gesuggereerd dat continue activering van NK-cellen met cytokines in vitro kan leiden tot NK-celuitputting door de studie van verschillende metabole trajecten10,11. De correlatie tussen NK cel metabolische status en functie is zeer belangrijk vanuit het oogpunt van kanker immunotherapie. Op dit gebied is de activering van NK-cellen met infusie van IL-15, alleen of in combinatie met monoklonale therapeutische antilichamen getest om het doden van tumorcellen12,13,14te verbeteren . De kennis van de metabolische status van de NK-cellen in reactie op deze behandelingsstrategieën zou een waardevolle voorspeller zijn van de NK-celactiveringsstatus en de moordfunctie.
De studie van metabole trajecten in andere myeloïde en lymfoïde cellen zoals monocyten, T- en B-cellen is beschreven15 en geoptimaliseerde methoden zijn gepubliceerd16. In dit protocol bieden we een methode die zowel een NK isolatieprotocol combineert dat hoge aantallen zuivere en levensvatbare NK-cellen oplevert als een geoptimaliseerd protocol om metabole activiteit te meten met behulp van een extracellulaire flux analyzer. Hier tonen we aan dat dit een geldige methode is voor de studie van metabole veranderingen in rust en IL-15 geactiveerde menselijke NK cellen. Voor de extracellulaire fluxtest zijn parameters zoals celnummer en medicijnconcentraties getest en geoptimaliseerd. Vergeleken met andere respirometrische methoden, de extracellulaire flux analyzer is volledig geautomatiseerd en in staat om te testen in real time, met zeer lage hoeveelheden cellen, tot 92 monsters tegelijk, en dus maakt hoge doorvoer screenings (met meerdere monsters en repliceert) in een relatief snelle manier17.
Deze methode kan worden gebruikt door onderzoekers die geïnteresseerd zijn in het beoordelen van NK celfunctie door het bestuderen van NK celmetabolisme. Het kan ook worden toegepast op cellen geactiveerd door andere cytokinen, antilichamen of oplosbare stimuli.

Natural Killer (NK) cellen bemiddelen voornamelijk aangeboren anti-tumor en anti-virale immuunreacties en reageren op een verscheidenheid van cytokinen en andere stimuli ter bevordering van overleving, cellulaire proliferatie, productie van cytokinen zoals interferon gamma (IFNγ) en / of cytotoxiciteit programma's. NK celactivering door cytokine stimulatie vereist een aanzienlijke remodellering van metabole trajecten ter ondersteuning van hun bio-en bio-synthetische eisen. Er is een grote hoeveelheid bewijs dat suggereert dat verminderde NK celmetabolisme wordt geassocieerd met een aantal chronische ziekten, waaronder obesitas en kanker, die het klinische belang van de beschikbaarheid van een methode om NK celmetabolisme te bepalen benadrukt. In dit artikel beschrijven we een methode om glycolyse en mitochondriale ademhaling te meten in primaire menselijke Natural Killer (NK) cellen geïsoleerd van perifeer bloed, in rust of na IL15-geïnduceerde activering. Het beschreven protocol kan gemakkelijk worden uitgebreid tot primaire menselijke NK-cellen die worden geactiveerd door andere cytokines of oplosbare stimuli.

Deze methode zorgt voor de bepaling van natuurlijke killer cel metabolisme. Een belangrijke parameter in de activering door het meten van zuurstofverbruik en pH-veranderingen in het extracellulaire medium. Het voordeel van de extracellulaire flux analyzer is dat het volledig geautomatiseerd is en in staat is om tot 92 monsters in real time te testen met lage hoeveelheden cellen.Dus waardoor hoge doorvoerschermen. Een geldige methode om glycolyse en ademhaling in inktcellen te bepalen is erg belangrijk in de kliniek. En centrifugeren ze op 800 keer G gedurende vijf minuten. Om de natuurlijke killer cellen of engaves te isoleren, tel de PVM zeeën en resuspend ze in NK scheiding buffer op een keer 10 tot de achtste cellen per milliliter.Breng vervolgens 10 milliliter van de celsuspensie over in een nieuwe buis van 50 milliliter. Voeg 500 microliters van NK celisolatie antilichaam mix. En 10 microliters van anti-cd drie positieve isolatie antilichaam mix aan de PBMCs.En broed ze 10 minuten op kamertemperatuur. Vortex magnetische kralen en voeg een milliliter aan de PBMCs en antilichamen. Vervolgens de MIGS 10 minuten bij kamertemperatuur incubeermen.Voeg 35 milliliter NK isolatiebuffer toe en plaats de buis 15 minuten op de magneet. Verzamel de supernatant voorzichtig met een 50 milliliter plastic pipet zonder de zijkanten of de onderkant van de buis aan te raken. Tel de cellen en centrifugeren ze op 800 keer G gedurende vijf minuten.Om de NK-cellen te stimuleren met oplosbaar gangpad 15, worden 750.000 cellen in 100 microliter van IMDM met 10%HS in een put van een 96 putplaat opnieuw opgetrokken. Verdun het menselijk gangpad 15 tot 1 microgram per milliliter in de IMDM en 10%HS. En voeg 100 microliter van het verdunde menselijke gangpad 15 toe aan de cellen.Herstel een controlemonster met niet-geprisuleerde cellen en plaats beide monsters in de couveuse op 37 graden Celsius. Stimuleer de cellen gedurende 48 uur en voer vervolgens de extracellulaire fluxtest uit. Daag de dag voor het experiment de analyzer in en laat hem opwarmen tot 37 graden Celsius.Open het censuurcartridgepakket en scheid de cartridge van de hulpplaat. Voeg vervolgens 200 microliter van de kalibrantoplossing toe aan elke put van de nutsplaat. En zet de centrale cartridge terug om ervoor te zorgen dat de sensoren volledig onder water staan.Voor optimale resultaten incubeer de cartridge 's nachts bij 37 graden Celsius in een koolstofdioxide vrije couveuse. Die goed bevochtigd. Om een met lijm bekleed mes te bereiden, pipette 25 microliter van de celkleefoplossing op elke put van de plaat.En geïncubeerd bij kamertemperatuur gedurende 20 minuten. Verwijder vervolgens de oplossing en was de plaat twee keer met 200 microliter steriel water per put. Houd de plaat 15 minuten open in de celkweekkap om de putten te laten drogen.Centrifuge de eerder geïsoleerde NK cellen toe te voegen 200 keer G gedurende vijf minuten. Verwijder vervolgens supernatants en was de cellen in opgewarmde mitochondriale stresstest medium of glycolyse stress test medium. Pellet de cellen opnieuw en resuspend ze naar de gewenste celconcentratie in hetzelfde medium.Leg 180 microliter celsuspensie per put in de testplaat. Gebruik putten A een, A 12 H een en H 12 als controle putten voor achtergrondcorrectie. Het toevoegen van 180 microliters van het testmedium aan deze Putten Incubbate de plaat gedurende 30 minuten bij 37 graden Celsius in een koolstofdioxide vrije incubator.Centrifuge de plaat op 200 keer G gedurende vijf minuten. Observeer vervolgens de cellen onder de microscoop om te controleren of ze een manolaag vormen aan de onderkant van de put. Incubeer de cellen nog 25 minuten.Warm werkende oplossingen tot 37 graden Celsius. En hun pH aanpassen naar 7,4 indien nodig. Laad de eerder bereide verbindingen voor een mitochondriale stresstest of voor een glycolyse stresstest in de planken A, B en C van de gehydrateerde censuurcartridge Zet de geladen censuurcartridge terug in de couveuse tijdens het opzetten van het programma.Als u het extracellulaire fluxtestprotocol wilt instellen, opent u de software en gebruikt u groepsdefinities om de voorwaarden voor behandeling te definiëren. Gebruik het tabblad Plaatkaart om de groepen putten aan te geven die vergelijkbare voorwaarden hebben, geef ook de achtergrondcorrectie en lege putten aan. Stel vervolgens het programma in op het tabblad Protocollen.Start het programma en plaats de sensorcartridge en hulpplaat op de lade. Vervang na de kalibratiestap de rembrantplaat voor de testplaat door de bijgevoegde cellen. Zodra de run is voltooid, haalt u de gegevens op en analyseert u deze.Om de zuiverheid en levensvatbaarheid van de NK-cellen te beoordelen. Kleine Alec Watts van PVM zeeën en de geïsoleerde NK cel populatie werden gekleurd en geanalyseerd door stroom cytometrie. De zuiverheid van de NK cel populatie werd vastgesteld door dubbele verblijf tegen CD drie en CD 56 of NKP 46.Veel mitochondriaal zuurstofverbruik voor de menselijke NK cellen worden hier getoond. Zoals verwacht ik celnummers weergegeven hogere OCR waarden. Celnummers correleerden ook lineair met de hoeveelheid DNA of eiwit in de put.Enerzijds waren de hogere celaantallen niet optimaal. Omdat op aanvulling van DNP, de zuurstofconcentratie in de put was volledig uitgeput in elke cyclus. Die de nauwkeurige berekening van OCR verhinderde.In menselijke NK-cellen bleek 100 micromolar DNP de optimale dosis te zijn. Hier wordt een typisch mitochondriaal stresstestexperiment met 750.000 NK-cellen per put getoond. In deze test oligomycine leidt tot een dramatische daling van het zuurstofverbruik.En een toename van ECAR. Dat is een overstap naar glycolyse en een poging om cellulaire ATP-niveaus te behouden. Activering van de NK-cellen via gangpad 15 veroorzaakte een toename van zowel het mitochondriale zuurstofverbruik als de extracellulaire verzuring.Basale Maximal en ATP gekoppelde ademhaling verhoogd, maar niet proton lek of niet mitochondriale ademhaling. Bovendien daalde het OCR/ECA-tarief wat duidt op een verschuiving naar een glycolytisch metabolisme na IL 15 stimulatie. Bij het uitvoeren van dit protocol is het erg belangrijk om een zuivere en gezonde celpopulatie te verkrijgen om het optimale celaantal te bepalen en de concentratie van uncablers te titreren.
