물 비용의 증가와 물 사용 의 환경 문제로 인해 농장과 보육원의 재활용 물이 점점 더 인기를 끌고 있습니다. 식물 질환의 확산과 발생을 줄이기 위해 재배자를 위해 많은 관개 방법이 개발되었습니다. 물의 근원(관개 또는 강수량)에 관계없이 유출이 발생하고, 많은 채소와 보육 재배자가유출1을수집하고 재활용할 수 있는 연못이 있다. 이는 재활용물이 작물2,3,34를관개하는 데 사용될 때 병원균의 확산을 선호하는 병원균 축적 가능성을 위한저수지를만든다. 표적 미생물의 검출 및 진단을 위해 관개수를 테스트하는 전통적인 방법은 속도와 감도가 성공과 수익성 있는 작물생산(11,,12)에결정적인 시기에 구식 접근법이다. 표적 병원체(예를 들어, P. capsici용가지)에 취약한 식물 조직은 감염을 제거하고 검사하기 전에 장시간 관개 연못에 매달린 수정된 트랩에 부착된다. 식물 조직으로부터의 샘플은 반선택적 미디어(PARPH)에 도금되고 배양 성장을 위해 배양한 다음, 복합현미경(13)을이용하여 형태학적 식별이 수행된다. 선택적 매체를 이용하여 소량의 오염된 물을 소량도금하여14,,15를하위 배양하기 전에 다른 식물 병원균에 대한 다른 유사한 검출 방법이 있다. 이러한 방법은 유기체를 분리하기 위해 2 주에서 6 주, 여러 차례의 하위 배양이 필요하며, 각 종의 주요 형태학적 문자를 인식할 수 있도록 Phytophthora 진단에 대한 경험이 필요합니다. 이러한 전통적인 방법은 수원에 존재하는 다른 미생물에 의한 간섭과 같은 요인으로 인해 P. capsici에 의해 오염된 관개 수를 검출하는 데 잘 작동하지 않습니다. 파이티움 스프와 수인성 박테리아와 같은 일부 빠르게 성장하는 미생물은 접시에 과도하게 자랄 수 있어 P. capsici를 탐지할 수 없는16,,17.
이 연구의 목적은 관개 물에서 P. capsici 동물원포자를 검출하기 위해 현장 및 실험실 설정 모두에서 사용할 수있는 민감하고 구체적인 분자 방법을 개발하는 것이었습니다. 이 프로토콜은 P. capsici,18,19의1121베이스 쌍(bp) 단편을 기반으로 P. capsici를구체적으로 증폭시킬 수 있는 새로운 루프 매개 이스테어 증폭(LAMP) 프라이머 세트의 개발을 포함한다. 동외에서 이전에 개발된 LAMP 프라이머(2015)는 이 연구를 위해 개발된분석서(20)에비해 사용되었다.
