phosphoproteomic strategy for profiling osmotic stress signaling in arabidopsis

단백질 인산화는 삼투성 조건 하에서 효소 활성 및 유전자 발현의 조절에 매우 중요합니다. 질량 분광법 (MS)기반 인포 프로 테오믹스는 식물 신호 변환을 연구하는 방법을 변화시켰습니다. 그러나, 커버리지의 깊이를 달성하기 위해 많은 시작 재료와 장기간 MS 측정 시간의 요구 사항은 식물의 글로벌 인포 프로테오믹 변화에 대한 높은 처리량 연구에 대한 제한 요인이되고있다. 식물 인산염의 감도와 처리량을 개선하기 위해, 우리는 삼투압 스트레스에 대응하여 식물 인산화 의 신속하고 포괄적 인 분석을 위한 탠덤 질량 태그 (TMT) 라벨과 결합된 정지 및 이동 추출 (stage) 팁 기반 인포프로테오믹스 접근법을 개발했습니다. 단계 팁 기술의 단순성과 높은 처리량을 활용하여 전체 절차는 두 가지 팁을 사용하여 인포펩티드 농축, 분수 및 시료 세척 단계를 완료하는 데 약 1시간이 걸리며, 이는 접근 방식의 사용하기 쉽고 높은 효율을 시사합니다. 이 접근법은 심층적인 식물 인포프로테오믹스 분석(> 11,000인스포펩티드 식별)을 제공할 뿐만 아니라 인접한 분수 간의 우수한 분리 효율(&5% 중복)을 보여줍니다. 또한, 멀티플렉싱은 TMT 라벨링을 사용하여 야생형 및 snrk2 탈취 돌연변이 식물의 인포프로테오믹 변화를 정량화하고 있다. 이 접근법은 육상 식물의 초기 삼투압 신호에 대한 이해를 밝히는 삼투압 스트레스에 대한 응답으로 Raf와 같은 키나아제의 인광 이벤트를 성공적으로 밝히는 데 사용되었습니다.
