망막 색소 상피(RPE)는 눈의 후방 세그먼트를 일렬로 세워 망막 항상성을 유지하기 위해 중요한 기능을 발휘하는특수 세포의 단층이다. RPE는 광수용체와 초리오카필로혈관4,5사이의 영양분 및 대사 산물의 수송에 참여하여 시각 주기2라는공정에서 광자 포획 시각적 안료를 재생하여 광수용체를 지원합니다. RPE 기능의 이상은 노화와 관련된 황반 변성6,Leber의 선천성 수마우로시스7,8 및 최고의 비텔리폼 황반 이영양증9와같은 수많은 인간 망막 질환을 뒷받침합니다. 기증자 눈 조직은 종종 연구 목적으로만 얻기 어렵기 때문에 유전자 변형을 가진 동물 모델은 망막 질환의 발달을 연구하는 대체 방법을 제공 할 수있습니다(10,11). 또한, CRISPR cas9 기술의 출현 및 적용은 이제 이전 유전자 표적화기술의한계를 능가하는 간단한 1단계 공정에서 게놈 소개(knock-in) 또는 삭제(knock-out)를 허용한다. 새로운 마우스모델(13)의 가용성 붐은 비침습적으로 RPE 기능을 평가하기 위해 보다 효율적인 기록 프로토콜을 필요로 합니다.
