Les ostéocytes sont des cellules terminales différenciées des progéniteurs ostéoblastiques qui se sont intégrés dans leur matrice sécrétée1. Ce sont les cellules les plus abondantes et les plus anciennes parmi les populations de cellules osseuses. Ils résident dans des lacunes et ont une morphologie stellaire caractéristique avec des dendrites qui s’étendent à travers des canaux appelés canalicules formant un vaste réseau de communication et d’échange métabolique avec leur environnement et la surface osseuse2. Les ostéocytes chorégraphient à la fois les rôles des ostéoblastes et des ostéoclastes dans le remodelage osseux, ils sont les cellules mécanosensorielles primaires conférant une adaptation à la charge mécanique3, sont impliqués dans l’homéostasie phosphate4 et la minéralisation de la matrice osseuse5, et avec le système lacunocanaliculaire, ils agissent comme un organe endocrinien signalant des tissus distants6.
Les ostéocytes sont situés dans une matrice minéralisée, ce qui limite l’accessibilité et rend leur isolement difficile, entravant ainsi l’investigation in vitro. L’une des premières méthodes d’isolement a décrit des ostéocytes isolés du calvaire de poulet à l’aide d’un anticorps monoclonal spécifique aux ostéocytes (OB7.3)7 qui a été connu plus tard pour être la variante aviaire de PHEX (gène régulateur de PHosphate avec homologie aux endopeptidases sur le chromosome X)8. D’autres chercheurs ont utilisé la digestion séquentielle d’os longs de rat 9 ou de souris 10 pour obtenir des fractions riches en ostéocytes dans lesquelles la pureté des ostéocytes était d’environ70%9. Les limites de cette procédure comprennent la pureté sous-optimale des cultures contaminées par d’autres types de cellules que les ostéocytes et le fait que les ostéocytes pourraient être potentiellement envahis par d’autres cellules en culture puisque les ostéocytes ont perdu la capacité de se diviser. Ces défis limitent la facilité d’utilisation des cultures à long terme.
Pour surmonter ces limitations, différentes lignées cellulaires ostéopacytaires ont été développées. La lignée cellulaire MLO-Y411 et la lignée cellulaire MLO-A512 sont notamment les lignées cellulaires les plus étudiées qui sont utiles pour l’étude des ostéocytes à un stade précoce; cependant, ils sont moins utiles pour étudier la signalisation des ostéocytes matures car ils expriment de faibles niveaux de sclérostine et de FGF2313, qui sont tous deux des marqueurs ostéocytes matures. D’autres lignées cellulaires, y compris IDG-SW3 14 et Ocy45415, expriment des niveaux élevés de sclérostine et de FGF23 et sont utiles pour étudier le stade ostéocytaire tardif. Les lignées cellulaires s’avèrent être des outils de recherche utiles; Néanmoins, ils ne viennent pas sans limites car ils ne représentent pas pleinement la biologie de la cellule primaire. Différentes lignées cellulaires représentent différents stades de développement du spectre de maturité des ostéocytes, et les lignées cellulaires ne parviennent pas à représenter l’hétérogénéité des ostéocytes primaires16,17.
Pour obtenir des cultures pures d’ostéocytes primaires, les chercheurs ont tiré parti du modèle murin cre dans lequel le promoteur dmp1 de 8 kb est utilisé pour stimuler l’expression de la protéine fluorescente verte (GFP) dans les ostéocytes18,19. Des souris transgéniques doubles (pOB-Col 2.3- GFP-cyan et DMP1-GFP-topaz) de Paic et al.19 et des souris transgéniques dmp1-topaze de Nakashima et al.20 ont été utilisées pour obtenir des populations d’ostéocytes. Dans lequel ils ont utilisé la digestion séquentielle et le FACS des ostéocytes exprimant la GFP pour acquérir des cultures d’ostéocytes primaires19,20. La direction de cre chez la souris rapporteure dmp1 de 10 kb Ai9, qui active la protéine tdTomato, s’est avérée présente dans les ostéocytes, les ostéoblastes, les muscles et les cellules de la moelle osseuse. Le promoteur dmp1 de 8 kb avait le même schéma d’expression, mais seule une proportion d’ostéoblastes et de cellules de moelle osseuse exprimait la protéine, ce qui indique que le promoteur dmp1 de 8 kb est plus spécifique21. Malgré cela, les résultats obtenus à l’aide du promoteur dmp1 de 8 kb doivent être interprétés avec prudence, et les profils d’expression génique doivent être systématiquement réalisés à l’aide de marqueurs spécifiques aux ostéocytes par rapport aux ostéoblastes pour s’assurer que la population obtenue est d’une pureté suffisamment élevée.
Les marqueurs ostéoclastes OSCAR et Dcstamp ont été trouvés dans des populations d’ostéocytes hématopoïétiques non appauvris vs appauvris, cette découverte a conduit les auteurs à conclure que les digestions obtenues par fractionnement de calvaire néonatal dmp1-topaze de 8 kb et le tri GFP sont contaminés par des cellules hématopoïétiques. La contamination par les cellules hématopoïétiques aurait pu être atténuée en resserrant la porte de tri de la GFP puisque les cellules hématopoïétiques GFP positives avaient une intensité de GFP raisonnablement inférieure à celle des cellules mésenchymateuses (ostéocytes) GFP-positives22.
Les méthodes d’étude des ostéocytes in vitro ont contribué à la richesse récente des informations sur la biologie des ostéocytes. Cependant, l’isolement des ostéocytes reste une procédure longue et laborieuse avec de faibles rendements cellulaires. La méthode décrite de digestion osseuse utilisant la collagénase et l’EDTA souvent jusqu’à la fraction 823, prend plusieurs heures au cours desquelles la viabilité des ostéocytes est taxée. Des chercheurs ont rapporté utiliser des fractions (2\u20125) pour le tri cellulaire 20, et ont montré que le profil d’expression des gènes associés aux ostéocytes par rapport à ceux des ostéoblastes confirme le succès de l’isolement des populations d’ostéocytes purs20. Dans cet article, nous décrivons le processus d’obtention des fractions (2\u20125), et nous comparons le rendement des ostéocytes de chaque fraction en commençant par la fraction 1 à 8 pour déterminer le retour des ostéocytes dans chaque fraction. Nous décrivons également la dissection du calvaria de souris dmp1-topaze nouveau-née et la digestion calvariaire à l’aide de collagénase et d’acide éthylènediaminetétraacétique (EDTA), ainsi que la préparation de cellules pour FACS.

L’ostéocytes, autrefois considéré comme un résident passif de l’os étant donné la fonction de détection de la charge mécanique en coulisses, est maintenant mis en lumière et a été montré pour avoir de multiples fonctions majeures comme la modification active de la matrice extracellulaire et la formation d’un organe endocrinien avec le système lacunocanaliculaire qui l’entoure envoyant des messages à des sites distants. Grâce aux méthodes qui ont permis de tester l’ostéocyte in vitro à partir d’ostéocytes primaires isolés en lignées cellulaires de type ostéocytes, les ostéocytes connaissent aujourd’hui un intérêt retentissant et une vague de connaissances sur la structure et la fonction. De nombreux aspects de la biologie des ostéocytes et de l’interaction avec d’autres composants moléculaires restent à découvrir. Dans ce protocole, nous décrivons en détail l’isolement efficace des ostéocytes primaires à partir du calvaire néonatal de souris dmp1-topaze, qui expriment la protéine fluorescente verte dans les ostéocytes, par fractionnement cellulaire et acquisition ultérieure de cultures d’ostéocytes primaires par FACS.

Le but de ce protocole est de démontrer le processus d’obtention de cultures d’ostéocytes primaires à partir de calvaire de souris néonatale dmp1-topaze par un processus de fractionnement utilisant la collagénase pour dégrader la matrice de collagène et l’EDTA pour la chélation du calcium, après quoi les cellules sont préparées pour FACS afin de séparer les ostéocytes des autres populations cellulaires.

Primary osteocyte isolation is a lengthy procedure that often leaves out in low cell areas. This protocol provides efficient methods for isolating high numbers of primary GFP-expressing osteocytes. This protocol reduces the handling time and allows a higher number of osteocytes to be obtained in a single procedure with minimal contamination from hematopoietic or mesenchymal cells.For the isolation of GFP-expressing osteocytes, place euthanized, postnatal day six to seven, transgenic mouse pups into a non-treated culture dish, and use scissors and tweezers to grasp the skin at the base of one's skull. Use the scissors to make an initial incision in the lifted skin, followed by cuts along both sides of the skull in front of the ears. Remove the skin to expose the calvaria and holding the head from the nasal bridge, cut the calvaria along the lamboid suture and along the edges of the parietal bones to the frontal part of the tissue.Separate the calvaria from the underlying brain tissue and place the bones concave side up in PBS. To obtain fraction one, pool up to five calvaria per 50-milliliter tube in five milliliters of freshly prepared two-milligram per milliliter collagenase, and incubate the tissues at 37 degrees Celsius and 300 revolutions per minute for 20 minutes. At the end of the incubation, discard the digest from fraction one and wash the calvaria with five milliliters of PBS.Replace the wash with five milliliters of five-millimolar EDTA supplemented with one milligram per milliliter of bovine serum albumin in PBS for 15 minutes at 37 degrees Celsius with shaking. At the end of the incubation, transfer the digest to a 50-milliliter conical tube and wash the calveria with five milliliters of PBS. Then transfer the wash solution to the digest.To obtain fraction two, place the digest for centrifugation and discard the supernatant. Re-suspend the pellet in eight milliliters of alpha minimum essential medium containing 10%fetal bovine serum, or FBS, and antibiotics, and seed the cells onto a 10-centimeter culture dish. Then place the fraction two cells into the cell culture incubator.After collecting and culturing fractions three and four as just demonstrated. Incubate the calveria in five milliliters of five-millimolar EDTA supplemented with one milligram per milliliter of bovine serum albumin in PBS for 15 minutes at 37 degrees Celsius with shaking. At the end of the incubation, collect and culture fraction five as demonstrated for fractions two, three, and four.Immediately after fractionation, collect the cells for sorting. Alternatively, collect the cells after 24 hours of culture by aspirating the medium and gently wash the cells two times with 10 milliliters of PBS per dish per wash. After the second wash, treat the cells with five milliliters of 0.5%tripsin EDTA in PBS per plate.After five minutes at 37 degrees Celsius, add five milliliters of complete alpha minimum essential medium to each plate and gently detach the cells with pipetting. To combine the cells in each fraction, filter the cell suspensions through a 40-micron cell strainer into a single 50-milliliter conical centrifuge tube. After washing the cells with 10 milliliters of PBS, add them to the tube.Filter the cells through a strainer and collect the cells by centrifugation. Once the cells are pelleted down, re-suspend the pellet in complete minimal essential medium to a one times 10 to the seventh cells per milliliter of medium concentration and filter the cells through a 35-micron nylon mesh capped tube. To test the efficiency of this method, fractions were separately sorted and cultured for 24 hours to determine the percentage and yield of osteocytes from each fraction.The density of the osteocytes in fractions two to five were higher than that observed for fraction one and decreased remarkably in fractions six, seven, and eight. Although the percentage of osteocytes obtained between all of the fractions is not statistically significant, the density of the osteocytes differs dramatically between fractions. Here the gating strategy for isolating GFP-positive osteocytes from their GFP-negative counterparts by fluorescence-activated cell sorting is shown.After sorting, osteocytes can be analyzed for the expression of osteocyte markers of interest. In this analysis, the osteocyte marker genes were expressed at higher levels in the isolated GFP-positive osteocytes compared to presort fraction two, which is known as a high osteoblast fraction. Here the morphology of a GFP-positive osteocyte retaining a stellate shape with dendrites extending from the cell body after 24 hours of culture on a plastic culture dish, post sort, can be observed.Pay attention when aspirating the digests as the calveria will be reduced to minute pellets. Remember to use a small tip aspirator to avoid cell loss. Osteocytes have been enigmatic because they are entombed in bone.This technique has paved the way to studying osteocyte molecular interactions and their control over osteoclasts and osteoblasts.
